Randomized Controlled Trial Comparing Micronized Progesterone (Amelgen ®) 400 mg BID Versus 400 mg TID for Luteal Support in Artificial Vitrified/Warmed Single Blastocyst Transfer Cycles With Low Progesterone on Day of Embryo Transfer

RSAD2: A pathogenic interferon-stimulated gene at the maternal-fetal interface of patients with systemic lupus erythematosus

Pregnancy disorders in patients with autoimmune diseases or viral infections are often associated with an excessive response of type I interferons. We identify radical S-adenosyl methionine domain containing 2 (RSAD2) as a pathogenic interferon-stimulated gene (ISG) associated with pregnancy complications in systemic lupus erythematosus (SLE). The increased expression of RSAD2 mainly occurs in macrophages and structural cell populations at the maternal-fetal interface of pregnant patients with SLE. The elevation of RSAD2 leads to the accumulation of diacylglycerol lipids in the placenta, impairing the necessary vascular development for the fetus. Depletion of Rsad2 in pregnant mice models exposed to type I interferon inducers significantly reduces lipid accumulation, vascular injury, and embryo development disorders. An RSAD2 inhibitor, L-chicoric acid (LCA), alleviates lipid accumulation and vascular damage, improving pregnancy outcomes in SLE-induced and spontaneous mouse models. This study proposes the potential of targeting RSAD2 to improve pregnancy outcomes in individuals with heightened type I interferon response.

Aneuploidy-driven gene expression profiling in human blastocysts: insights from RNA-Seq analysis

PURPOSEPreimplantation aneuploidy in humans is one of the primary causes of implantation failure and embryo miscarriage. This study was conducted to gain insight into gene expression changes that may result from aneuploidy in blastocysts through RNA-Seq analysis.METHODSThe surplus embryos of preimplantation genetic testing for aneuploidy (PGT-A) candidate couples with normal karyotype and maternal age < 38 were collected following identical ovarian stimulation protocol. The embryos were selected based on trophectoderm biopsy and array comparative genomic hybridization in three groups: normal group, small chromosomes aneuploidy group (SCA), including single aneuploidy for small chromosomes 16, 20, 21, 22, and other chromosomes aneuploidy group (OCA), including single aneuploidy for other chromosomes.RESULTSPrincipal component analysis revealed overall differentiation of transcriptome of the groups, confirming embryo classification. The Gene Ontology indicated that transcription, ubiquitination, autophagy, and DNA repair pathways were upregulated in aneuploid embryos. The overexpression of five genes, UBE2E2 and VPS4A, BUB1B, CDCA8, and COX14 was confirmed by quantitative real-time PCR. Additionally, overexpression was observed in translation and protein synthesis pathways in aneuploid embryos. Mitochondrial pathway upregulation was notable in both SCA and OCA groups, while the apoptosis pathway was overexpressed only in the OCA group. Only cellular lipid synthesis pathway differed between SCA and OCA, the two aneuploid groups.CONCLUSIONSThis study highlights the impact of aneuploidy on the gene expression in blastocysts independent of aneuploidy type and paves the way for understanding the molecular mechanisms underlying the generation of aneuploidy.

Microplastics aggravate the adverse effects of methylmercury than inorganic mercury on zebrafish (Danio rerio)

The potential health risks of microplastics (MPs) and their combined exposure with heavy metals such as mercury (Hg) in aquatic environment are increasingly concerned recently. In this work, zebrafish embryos were exposed to different levels of polystyrene microplastics (PS-MPs, ∼0.1 μm) coupled with Hg(II) or/and MeHg at 20 μg/L, to investigate the tissue biodistribution and accumulation of PS-MPs and Hg species, and their interaction, as well as embryo toxicity, oxidative stress and metabolic profiles. With zebrafish embryo development, PS-MPs were ingested and then primarily translocated to yolk sac, liver, and intestinal tissues, further acted as a significant vector for improving the bioaccumulation of MeHg vs. Hg(II). Whatever single or combined exposure of PS-MPs and Hg species, embryo disorders, such as delayed hatching, developmental abnormalities, and motor behavioral, and increased oxidative stress indications were obviously found. Herein, PS-MPs + MeHg aggravated oxidative stress compared with MeHg alone, which might been relevant to the high accumulation of Hg level in zebrafish larvae induced by PS-MPs. Non-targeted metabolomics results proved PS-MPs involvement disturbed lipid metabolism, amino acid metabolism, and energy metabolism compared with alone Hg(II) or MeHg exposure, of which excessive energy metabolism by activating the glycolysis process was found in PS-MPs + MeHg treatment. This work reveals the enhancement efficacy of PS-MPs on MeHg induced toxicity and adverse stress, further proving the differentiated effect of elemental chemical forms with microplastics. In the future, elemental species must be considered for the combined toxicity evaluation and ecological risk assessments of microplastics and heavy metals.
