The Phase 3 SUCCESSOR-2 trial (NCT05552976) demonstrated that adding mezigdomide (a next-generation CELMoD/molecular glue degrader) to the established carfilzomib + dexamethasone (Kd) backbone produced a statistically significant and clinically meaningful improvement in progression-free survival (PFS) in patients with relapsed/refractory multiple myeloma (RRMM). The trial met its primary endpoint, with MeziKd showing superior PFS and higher response rates versus Kd alone. The overall evaluation was Positive, representing a landmark Phase 3 validation of the CELMoD drug class in combination with a proteasome inhibitor.
