Preparation of slow-release fertilizer derived from rice husk silica, hydroxypropyl methylcellulose, polyvinyl alcohol and paper composite coated urea

There is a growing trend toward utilizing agricultural waste to create value-added products, addressing environmental concerns associated with their disposal. This study focuses on developing slow-release fertilizers (SRFs) using amorphous silica derived from rice husk, hydroxypropyl methylcellulose (HPMC), polyvinyl alcohol (PVA), waste paper, and urea. Experimental optimization was carried out using the response surface methodology central composite design (RSM-CCD). The optimal formulation included 8.63 g of silica, 1.04 g of HPMC, and 0.27 g of PVA. Two SRFs were prepared under these conditions: SRF1, consisting of silica, HPMC, and PVA, and SRF2, which additionally incorporated coated waste paper. Characterization techniques such as Fourier Transform Infrared (FTIR) spectroscopy, X-ray diffraction (XRD) Scanning Electron microscopy (SEM) and Brunauer-Emmett-Teller (BET) analysis were used to examine the materials. The rice-husk-derived silica exhibited a pore size of 2.140 nm and a BET surface area of 690 m²/g, providing an excellent surface for nutrient encapsulation. Although the addition of coated waste paper minimally influenced the slow-release behaviour of SRF2, however the other components effectively reduced nutrient leakage by trapping the nutrients. The swelling behaviour of the SRFs was analyzed in different media after 72 h, showing values of 2.66, 2.54 (g/g) in distilled water, 2.20, 2.58 (g/g) in pH 4, and 1.86, 3.09 (g/g) in pH 9 solutions. The swelling kinetics aligned with Scott's second-order kinetic model. Urea release tests in water revealed a release of 94 % and 97 % at 24 h for SRF1 and SRF2, respectively, compared to 98 % release of pure urea within 1 h. SRF2 demonstrated optimal nutrient release after 48 h. The release kinetics followed the first-order kinetic model for both SRF1 and SRF2, highlighting their potential as effective slow-release fertilizers.

Molecular characterization of SPATA6 and association of its SNPs with testicular size in sheep

The testis is an important organ for maintaining fertility in males, and testis size is positively correlated with ejaculate volume, sperm motility, thus fertility. Spermatogenesis-associated 6 (SPATA6) is an evolutionarily conserved testis-specific gene reported in many species. However, the effect of SPATA6 expression levels on testicular development and the effect of single nucleotide polymorphisms (SNPs) on testis and epididymis phenotype in sheep have not been studied. The purpose of the research was to investigate the expression profile of SPATA6 and its effect on testicular development and to confirm the effect of SNPs on the testis and epididymis phenotype. In this study, we detected a 1245bp coding sequence (CDS) of SPATA6 and encoded 414 amino acids. The expression levels of SPATA6 were significantly higher in the testis than in other tissues and gradually increased with testis development. Moreover, the expression level in the large testis was significantly higher than that in the small testis at six months. A total of 11 SNPs were detected in the coding region of SPATA6 by cDNA-pooling sequencing and improved multiplex ligation detection reaction (iMLDR) methods. Correlation analysis showed that SNP2 (c. 3631C > G) significantly affected left epididymis weight (LEW) and right epididymis weight (REW), and SNP10 (c. 937 A > G) significantly affected REW. And the combined genotype of SNP1 (c. 4245 G > A) and SNP2 significantly affected REW. The current study concluded that SPATA6 plays an important role in testicular development and the SNPs significantly associated with the epididymis phenotype can provide molecular markers for the early selection of high-fertility Hu sheep.

Identification of nonfunctional SPATA20 causing acephalic spermatozoa syndrome in humans

AbstractAcephalic spermatozoa syndrome (ASS) is a rare and severe type of teratozoospermia characterized by the predominance of headless spermatozoa in the ejaculate. However, knowledge about the causative genes associated with ASS in humans is limited. Loss‐of‐function of SPATA20 has been suggested to result in the separation of the sperm head and flagellum in mice, whereas there have been no cases reporting SPATA20 variants leading to human male infertility. In this study, a nonsense mutation in SPATA20 (c.619C > T, p.Arg207*) was first identified in an ASS patient. Moreover, this variant contributed to the degradation of SPATA20 and was associated with decreased expression of SPATA6, which plays a vital role in the assembly of the sperm head‐tail conjunction in humans. In addition, the infertility caused by loss‐of‐function mutation of SPATA20 might not be rescued by intracytoplasmic sperm injection (ICSI). Collectively, our findings suggested that SPATA20 might be required for sperm head‐tail conjunction formation in humans, the nonfunction of which may lead to male infertility related to ASS. The discovery of the loss‐of‐function mutation in SPATA20 enriches the gene variant spectrum of human ASS, further contributing to improved diagnosis, genetic counseling and prognosis for male infertility.
