Introduction

Plays a role in the inhibition of RLR-mediated type I interferon signaling pathway by targeting RIGI for autophagic degradation. Interacts specifically with ISG15-associated RIGI to promote interaction between RIGI and the autophagic cargo receptor p62/SQSTM1 to mediate RIGI degradation via selective autophagy (PubMed:29288164). Also plays a role in the inhibition of NF-kappa-B signaling pathway and inflammatory response by promoting the degradation of p65/RELA.

Myeloid-lineage-specific membrane protein LRRC25 suppresses immunity in solid tumor and is a potential cancer immunotherapy checkpoint target

Leucine-rich repeat containing 25 (LRRC25), a type I membrane protein, is specifically expressed in myeloid cells including neutrophils and macrophages. The anti-inflammatory role of LRRC25 was suggested in a few pathogenic models. However, its role in cancer immunity has not been interrogated. Here, we demonstrate that LRRC25 is robustly expressed in tumor-associated macrophages (TAMs). Lrrc25 deficiency in the tumor microenvironment (TME) suppresses growth of multiple murine tumor models by reprogramming TAMs toward an anti-tumor phenotype and thereby enhancing infiltration and activation of CD8⁺ T cells. The Nox2-ROS-Nlrp3-Il1β pathway is elevated in Lrrc25-deficient TAMs. Furthermore, a human myeloid cell line or mice with loss of Lrrc25 appear normal, indicating that LRRC25 is a safe immune target. Our results suggest that as an unappreciated immune checkpoint for tumor immunotherapy, the myeloid-specific membrane protein LRRC25 orchestrates the activity of TAMs via the canonical Nlrp3-IL1β inflammatory pathway and influences CD8⁺ T cell chemotaxis to the TME.

LRRC25 Is a Potential Biomarker for Predicting Immunotherapy Response in Patients with Gastric Cancer

BACKGROUNDLeucine-rich repeat containing 25 (LRRC25) is distinguishingly expressed in different tumor types, but the relationship with immune cell infiltration in gastric cancer stills unclear. We analyzed LRRC25 expression using pan-cancer data from The Cancer Genome Atlas and gene data from Gene Expression Omnibus. The clinical significance was further evaluated using gastric cancer tissues derived from clinical trials (no. NCT04208347).METHODThrough bioinformatics analysis of TCGA database and the UCSC Xena database, we found the correlation between LRRC15, gastric cancer, and immune cell infiltration. Further, multiplex immunohistochemistry/immunofluorescence (mIHC/IF), tissue microarray, and image acquisition and quantitative analysis confirmed our theory.RESULTSIt was discovered that LRRC25 was highly expressed in gastric cancer. Further analysis revealed that the expression of LRRC25 associated with gene sets implicated in immunity, including those in innate immunity, adaptive immunity, and chemokine signaling pathways. The result of (mIHC/IF) suggests a negative relevance between LRRC25 and response of anti-PD-1 treatment and reveals a trend of consistent change on LRRC25 + cells and CD16 expression. We also discovered that LRRC25 expression significantly associated with immune cell infiltration level. In particular, there is a relationship between LRRC25 and the phenotype and function of NK cells.CONCLUSIONHigh LRRC25 expression contributes to immunosuppressive microenvironment by influencing chemokine axis in gastric cancer. LRRC25 may serve as a clinically useful biomarker for predicting neoadjuvant immunotherapeutic response in patients with gastric cancer. LRRC25 can affect the phenotype and function of NK cells.

Green synthesis of magnetic aromatic polyamide composites via mechanochemical ball milling with a deep eutectic system for the magnetic solid-phase extraction of antibiotics from water

Magnetic aromatic polyamide (MAPA) composites were synthesized using a green mechanochemical ball milling process and used for magnetic solid-phase extraction (MSPE) and detection of trace level fluoroquinolone antibiotics (FQs) from water samples. The formation of MAPAs was facilitated by deep eutectic systems (DESys) created from magnetic beads and organic monomers during ball milling. A comprehensive study was conducted to investigate the kinetics, thermodynamics, and mechanisms of MAPA adsorption of ciprofloxacin hydrochloride (CIP) and ofloxacin (OFL) from water. The maximum adsorption capacities for CIP and OFL were found to be 151.5 mg/g and 164.1 mg/g, respectively. Additionally, the recyclability and selective adsorption capabilities of MAPAs were evaluated. MAPAs were subsequently used for MSPE of CIP and OFL in water samples. Both UV-Vis spectrometry and HPLC methods can be used for analysis, the MSPE-HPLC method showed superior linearity for CIP (0.06-1500 μg/L) and OFL (1-1200 μg/L), with R² values of 0.9995 and 0.9999, and limit of detection (LOD) as low as 0.006 μg/L and 0.02 μg/L, respectively. Both analytical methods are highly effective and practical for use with real water samples. MAPA used for MSPE offer a promising green approach for detecting antibiotic contamination in aquatic ecosystems.
