Hypoxia-inducible factor-1α involves in regulating anti-lipopolysaccharide factors expression via NF-κB under hypoxia stress in Chinese mitten crab (Eriocheir sinensis)

Oxygen is essential for the survival of organisms. Hypoxia profoundly affects the immune response in aquatic crustaceans, nevertheless, the precise mechanisms of immunological regulation under hypoxic conditions remain unclear. Hypoxia-inducible factor 1-alpha (HIF-1α), a key regulator of oxygen homeostasis, also plays a vital role in the immunological responses of mammals. Nonetheless, it remains uncertain whether HIF-1α regulates the immune response of crustaceans under hypoxia stress. This study investigated the expression patterns of EsHIF-1α and anti-lipopolysaccharide factors (ALFs) in response to Aeromonas hydrophila stimulation under hypoxia stress in Eriocheir sinensis. The mRNA expression levels of EsHIF-1α in haemocytes were significantly increased after hypoxia treatment, while were markedly reduced following A. hydrophila stimulation under hypoxic condition. Similarly, the EsALFs mRNA expression levels were also significantly decreased post A. hydrophila injection under hypoxic condition. Subsequently, the effect of EsHIF-1α on EsALFs mRNA expression was detected. The mRNA transcripts of EsALFs significantly diminished in HIF-1α inhibitor (KC7F2) injected crabs, however, a significant increase was observed in HIF-1α activator (IOX4) injected crabs. Furthermore, the mRNA expression and phosphorylation levels of NF-κB exhibited a similar trend following the inhibition or activation of EsHIF-1α, indicating that EsHIF-1α has a positive effect on the expression and activity of NF-κB. In addition, the bacterial clearance of haemolymph in the HIF-1α activated group was significantly higher, whereas in the HIF-1α inhibited group it was significantly lower, compared to the control group. Our findings collectively suggested that EsHIF-1α regulated ALFs expression through NF-κB activation in E. sinensis in response to A. hydrophila stimulation under hypoxic conditions. This research improves the understanding of the immunological regulation mechanisms in crustaceans under hypoxia stress.

Litopeidin28-51, a novel antimicrobial peptide from Litopenaeus vannamei, combats white spot syndrome virus infection through direct virus lysis and immunomodulatory effects

White spot syndrome virus (WSSV) poses a critical threat to crustacean aquaculture, particularly shrimp, causing widespread pandemics. In crustaceans, hemocytes function as a key component of the innate immune system and play a pivotal role in both cellular and humoral immune responses by producing various immune factors, such as antimicrobial peptides (AMPs), to defend against pathogenic microorganisms. In this study, an uncharacterized functional gene named Litopeidin was identified in Pacific white shrimp (Litopenaeus vannamei). It exhibited heightened expression in hemocytes and demonstrated a significant response to WSSV infection. Further, a truncated peptide, Litopeidin_28-51, derived from this gene, was characterized and identified as a novel AMP with robust antibacterial and antifungal properties, especially against common aquatic pathogens, including Vibrio spp. Moreover, Litopeidin_28-51 significantly suppressed the expression of viral genes (IE1 and VP28, WSSV replication-related genes) and the VP28 protein, as well as reduced viral copy numbers in hematopoietic tissue (Hpt) cells following WSSV infection. Mechanistic studies revealed that Litopeidin_28-51 exhibited a direct virucidal effect on WSSV and significantly upregulated immune-related gene expression (including Relish, ALF, Crustin, and LYZ1) in Hpt cells. Notably, in Cherax quadricarinatus and L. vannamei, either co- or pre-treatment with Litopeidin_28-51 markedly reduced animal mortality and viral replication in tissues. Collectively, the findings suggest that Litopeidin_28-51, a newly identified AMP with potent antibacterial activity, effectively inhibits WSSV replication by disrupting the viral envelope and regulating the cellular antiviral responses, making it a promising candidate for developing anti-infective agents or immunostimulants in aquaculture.

Impact of amyloid-like ovalbumin fibril consumption on health-related markers: An in vitro approach

Induction of amyloid-like morphology in food proteins offers high potential to induce new techno-functional properties in food products (e.g. use as emulsifier, thickener or gelling agent in e.g. bakery and confectionery products). However, the health impact of amyloid-like fibril (ALF) consumption remains widely understudied and merits additional research. The aim of this study was to (partially) elucidate the general health impact of food-borne ALF consumption, using egg white ovalbumin as a case study. Based on in vitro cell culture models it was demonstrated that ovalbumin ALFs (i) do not induce direct cytotoxic effects on intestinal (Caco-2, IPEC-J2) and neuronal (SH-SY5Y) cell lines, but (ii) are able to induce a Toll-like-receptor-mediated innate immune response, similar to endogenous amyloids, in activated THP-1 cells. Furthermore, the consecutive in vitro digestion and absorption (enterocyte and M-cell) experiments demonstrated that ovalbumin ALFs (i) do not completely lose their ALF morphology upon in vitro gastrointestinal digestion, and that (ii) the ALF core sequences, located at the center of the ALF structure, are transported across Caco-2 based cell models, suggesting aggregate transport. In vivo, intestinal translocation of ingested ALFs would imply potential cross-seeding of endogenous, disease-related precursor proteins. The ability of ovalbumin ALFs to induce aggregation of a disease-related precursor protein, αSyn, was evaluated in a precursor overexpressing cell model. Here, it was illustrated that only homologous (αSyn) - but not heterologous (ovalbumin) - seeding resulted in intracellular aggregation bodies of (phosphorylated) αSyn. The lack of cross-seeding supports the assumption that ovalbumin ALF consumption is not a risk factor for the development of α-synucleinopathies like Parkinson's disease.
