Introduction

Component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle (PubMed:18485873). The APC/C complex acts by mediating ubiquitination and subsequent degradation of target proteins: it mainly mediates the formation of 'Lys-11'-linked polyubiquitin chains and, to a lower extent, the formation of 'Lys-48'- and 'Lys-63'-linked polyubiquitin chains (PubMed:18485873). The APC/C complex catalyzes assembly of branched 'Lys-11'-/'Lys-48'-linked branched ubiquitin chains on target proteins (PubMed:29033132). APC7 is not required for the assembly of the APC/C complex, but has an enzyme-substrate adapter activity mediating the processive ubiquitination of specific substrates (PubMed:34942119). Involved in brain development through the specific ubiquitination and clearance of MKI67 from constitutive heterochromatin after neuronal progenitors exit mitosis (PubMed:34942119).

Utility of a Large Series of B‐Cell Precursor Acute Lymphoblastic Leukemia Cell Lines as a Model System

ABSTRACTBackgroundIn B‐cell precursor acute lymphoblastic leukemia (BCP‐ALL), chromosomal translocations are strongly associated with prognoses. RNA sequencing (RNA‐seq) is a powerful technology that reveals a close correlation between types of translocation and patterns of gene expression in clinical samples of BCP‐ALL. Cancer cell lines are powerful research tools, and thus, we built a larger series of BCP‐ALL cell lines and performed RNA‐seq analysis to confirm their utility as a model system.MethodsWe performed RNA‐seq in a total of 94 BCP‐ALL cell lines, including 80 cell lines with 8 representative types of translocations.ResultsIn the UMAP visualization, a close association was confirmed between the types of fusion genes and patterns of gene expression. In the cluster analysis of the gene expression profile, each type of fusion gene showed a clear association with the expression profile in the top 51 variable genes. Of clinical importance, the majority of the top variable genes in the BCP‐ALL cell lines also showed a significant association with the types of fusion genes in the clinical samples. When an association of 125 cell cycle‐related genes with the percentage of S and G2/M phases in 67 cell lines was evaluated, a significant positive correlation with cell cycle progression was confirmed in 10 cell cycle‐related genes (HDAC2, CDC23, YWHAG, MAD2L1, CCNH, ANAPC7, CDC6, ANAPC5, ORC3, andRBX1). Moreover, significant upregulation and downregulation of 40 and 10 genes, respectively, were observed in the cell lines established at relapse compared with those established at diagnosis. Four (SP6, CCNE1, HIST1H2BH, and DECR2) and two (EVI2B and SYN1) of these genes were also significantly higher and lower, respectively, in the clinical samples at relapse than in those at diagnosis.ConclusionLarge series of BCP‐ALL cell lines is a powerful research tool for studying the mechanisms of leukemogenesis and the disease progression of BCP‐ALL.

The Multifunctional Anaphase Promoting Complex 7 (APC7) Gene Is Associated With Increased Plant Growth and Improved Resistance to DNA and RNA Viruses

ABSTRACTThe anaphase promoting complex 7 (AtAPC7) is an APC/C subunit expressed in different organs of Arabidopsis thaliana and conserved among eukaryotes. A variant of the complete APC7 protein, containing its C‐terminal region (named APC‐CT), shows a high homology with a tobacco viral replication inhibitor (IVR‐like) protein that reduces plant susceptibility to RNA viruses. Here, the role of the AtAPC7 gene was investigated by characterizing Arabidopsis plants overexpressing the full‐length AtAPC7 (APC7OE) and the C‐terminal portion (APC7‐CTOE), by phenotypical, physiological and molecular approaches. APC7OE plants showed improved growth of vegetative organs, earlier flowering and increased photosynthetic efficiency, CO2 assimilation and productivity, compared with Col‐0 control plants. Conversely, APC7‐CTOE plants showed reduced susceptibility to both RNA and DNA viruses, along with an improvement in plant growth, although not surpassing APC7OE plants. Altogether, the data provide evidence for the role of the AtAPC7 in regulating cell division, expansion and differentiation, accompanied by an increase in photosynthetic capacity, resulting in enhanced plant biomass and seed yield. AtAPC7‐CT might reduce growth‐defence trade‐offs, enabling plants to simultaneously defend themselves while promoting better growth. Our findings highlight the multifunctional role of AtAPC7, unveiling the potential of its orthologous genes as valuable biotechnological tools in important crops.

AP‐1/C‐FOS and AP‐1/FRA2 differentially regulate early and late adipogenic differentiation of mesenchymal stem cells

AbstractObesity is defined as an abnormal accumulation of adipose tissue in the body and is a major global health problem due to increased morbidity and mortality. Adipose tissue is made up of adipocytes, which are fat‐storing cells, and the differentiation of these fat cells is known as adipogenesis. Several transcription factors (TFs) such as CEBPβ, CEBPα, PPARγ, GATA, and KLF have been reported to play a key role in adipogenesis. In this study, we report one more TF AP‐1, which is found to be involved in adipogenesis. Human mesenchymal stem cells were differentiated into adipocytes, and the expression pattern of different subunits of AP‐1 was examined during adipogenesis. It was observed that C‐FOS was predominantly expressed at an early stage (Day 2), whereas FRA2 expression peaked at later stages (Days 6 and 8) of adipogenesis. Chromatin immunoprecipitation‐sequencing analysis revealed that C‐FOS binds mainly to the promoters of WNT1, miR‐30a, and ANAPC7 and regulates their expression during mitotic clonal expansion. In contrast, FRA2 binds to the promoters of CIDEA, NOTCH1, ARAF, and MYLK, regulating their expression and lipid metabolism. Data obtained clearly indicate that the differential expression of C‐FOS and FRA2 is crucial for different stages of adipogenesis. This also raises the possibility of considering AP‐1 as a therapeutic target for treating obesity and related disorders.
