Tauroursodeoxycholic acid inhibits endothelial-mesenchymal transition in high glucose-treated human umbilical vein endothelial cells

The high glucose-induced endothelial-mesenchymal transition (EndMT) may be the initial and underlying mechanism of diabetic vascular complications. Although tauroursodeoxycholic acid (TUDCA) plays various protective roles in diabetes and its complications, it's unclear whether TUDCA inhibits the high glucose-induced EndMT. In this study, human umbilical vein endothelial cells (HUVECs) were treated with high glucose and intervened with TUDCA. The mRNA expression of fibroblast as well as endothelial markers, fibroblast specific protein 1 (FSP1), collagen I, CD31 and calcium adhesion protein 5, was detected. The protein content of FSP1 and CD31 was ascertained, with FSP1 distribution illustrated. The scratch assay was performed to evaluate the migratory ability of HUVECs. The protein content of TGF-β1 and Smad3, the distribution of Smad3 and the binding of Smad3 to the gene promoter of FSP1, were measured. The results firstly showed that TUDCA reversed the expression of EndMT-related genes in high glucose-treated HUVECs. Furthermore, TUDCA reduced FSP1 content with elevation in CD31, inhibited FSP1 distribution and attenuated morphological changes of high glucose-treated HUVECs. Meanwhile, TUDCA inhibited the high glucose-enhanced migratory ability of HUVECs. Mechanically, TUDCA prevented the binding of Smad3 to the gene promoter of FSP1 in high glucose-treated HUVECs, although it had little effect on the content of TGF-β1 and Smad3. In conclusion, TUDCA inhibited the high glucose-induced EndMT via preventing Smad3 from binding to the gene promoter of fibroblast markers, such as FSP1, in HUVECs.

Comprehensive Characterization of a Subfamily of Ca²⁺-Binding Proteins in Mouse and Human Retinal Neurons at Single-Cell Resolution

Ca2+-binding proteins (CaBPs; CaBP1–5) are a subfamily of neuronal Ca2+sensors with high homology to calmodulin. Notably, CaBP4, which is exclusively expressed in rod and cone photoreceptors, is crucial for maintaining normal retinal functions. However, the functional roles of CaBP1, CaBP2, and CaBP5 in the retina remain elusive, primarily due to limited understanding of their expression patterns within inner retinal neurons. In this study, we conducted a comprehensive transcript analysis using single-cell RNA sequencing datasets to investigate the gene expression profiles of CaBPs in mouse and human retinal neurons. Our findings revealed notable similarities in the overall expression patterns of CaBPs across both species. Specifically, nearly all amacrine cell, ganglion cell, and horizontal cell types exclusively expressed CaBP1. In contrast, the majority of bipolar cell types, including rod bipolar (RB) cells, expressed distinct combinations of CaBP1, CaBP2, and CaBP5, rather than a single CaBP as previously hypothesized. Remarkably, mouse rods and human cones exclusively expressed CaBP4, whereas mouse cones and human rods coexpressed both CaBP4 and CaBP5. Our single-cell reverse transcription polymerase chain reaction analysis confirmed the coexpression CaBP1 and CaBP5 in individual RBs from mice of either sex. Additionally, all three splice variants of CaBP1, primarily L-CaBP1, were detected in mouse RBs. Taken together, our study offers a comprehensive overview of the distribution of CaBPs in mouse and human retinal neurons, providing valuable insights into their roles in visual functions.

Characterization of Retinal Development in 13-Lined Ground Squirrels

PurposeThe cone-dominant, 13-lined ground squirrel (13-LGS) retina mimics the human central retina, but a thorough examination of retinal development in this species has not been reported. Here, the embryonic and postnatal development of the 13-LGS retina was studied to further characterize 13-LGS as a practical alternative animal model for investigating cone-based vision in health and disease.MethodsThe spatiotemporal expression of key progenitor and cell type markers was examined in retinas from defined embryonic and postnatal stages using immunohistochemistry. Postnatal gene expression changes were validated by quantitative PCR.ResultsThe 13-LGS neuroblastic layer expressed key progenitor markers (Sox2, Vsx2, Pax6, and Lhx2) at E18. Sequential cell fate determination evidenced by the first appearance of cell-type-specific marker labeling was at embryonic stage 18 (E18) with ganglion cells (Brn-3A, HuC/D) and microglia (Iba1); at E22.5 with photoreceptor progenitors (Otx2, recoverin) followed shortly by horizontal and amacrine cells (Lhx1, Oc1) at E24 to E25.5; and at postnatal stage 15 (P15) with bipolar cells (Vsx1, CaBP5) and Müller glia cells (GS, Rlbp1). Photoreceptor maturation indicated by opsin-positive outer segments and peanut agglutinin (PNA) labeling of cone sheaths was completed at the time of eye opening (P21-P24).ConclusionsThe timeline and order of retinal cell development in the 13-LGS generally matches that recorded from other mammalian models but with a stark variation in the proportion of various cell types due to cone-dense photoreceptors.Translational RelevanceThis thorough examination of an emerging translationally relevant cone-dominant specie provides a baseline for future disease modeling and stem cell approach studies of human vision.
