Carbon Quantum Dot Nanoparticles Enhance the Efficacy of Spodoptera littoralis Nucleopolyhedrovirus Suspoemulsion

ABSTRACTThis study evaluates the efficacy of Spodoptera littoralis nucleopolyhedrovirus (SpliNPV) and laboratory‐synthesized carbon quantum dot nanoparticles (CQDNPs) against the second instar Spodoptera littoralis larvae under laboratory and greenhouse conditions. Individually, both SpliNPV and CQDNPs exhibited substantial lethality (91.6% and 83.3% at 1 × 108 OBs/ml and 700 mg/ml, respectively) (p < 0.05). The LC50 values were 1.88 × 105 OB/ml and 434.2 mg/mL, and the LT50 values were 8.9 and 9.8 days, respectively. Four LC‐based combined treatments demonstrated significant additive effects, with the SpliNPV (LC50) + CQDNPs (LC25) combination achieving the optimum effect with a mortality rate of 86.3% and an LT50 value of 6.6 days, leading to its selection for the suspoemulsion nanoparticle (SENP) formulation. The SENP formulation displayed superior performance, achieving the highest mortality rates and fastest killing times across all environments: 89.0% in laboratory conditions, 83.3% on eggplant plants, and 76.6% on pepper plants. In contrast, the suspoemulsion (SE) and unformulated (UF) formulations showed lower efficacy, emphasizing the importance of formulation in enhancing the biological activity of SpliNPV. The LT50 values further supported these findings, with the SENP formulation demonstrating the shortest LT50 values, indicating faster lethality. A significant decrease in CHS‐B, IIM2, PER3, REPAT14, and CDA1 expression was observed, particularly in the combined CQDNPs + SpliNPV treatment, while API expression increased significantly. These findings highlight the potential of nanoparticle‐enhanced formulations like SENP, and integrating CQDNPs with SpliNPV can significantly enhance pest control efficacy.

Evidence for the Involvement of Gene Regulation of Inflammatory Molecules in the Accumulation of Intracellular Cholesterol: The Mechanism of Foam Cell Formation in Atherosclerosis

Background:The relationship between the cellular pro-inflammatory response and intracellular lipid accumulation in atherosclerosis is not sufficiently studied. Transcriptomic analysis is one way to establish such a relationship. Previously, we identified 10 potential key genes (IL-15, CXCL8, PERK, IL-7, IL-7R, DUSP1, TIGIT, F2RL1, TSPYL2, and ANXA1) involved in cholesterol accumulation in macrophages. It should be noted that all these genes do not directly participate in cholesterol metabolism, but encode molecules related to inflammation.Methods:In this study, we conducted a knock-down of the 10 identified key genes using siRNA to determine their possible role in cholesterol accumulation in macrophages. To assess cholesterol accumulation, human monocyte-derived macrophages (MDM) were incubated with atherogenic LDL from patients with atherosclerosis. Cholesterol content was assessed by the enzymatic method. Differentially expressed genes were identified with DESeq2 analysis. Master genes were determined by the functional analysis.Results:We found that only 5 out of 10 genes (IL-15, PERK, IL-7, IL-7R, ANXA1) can affect intracellular lipid accumulation. Knock-down of the IL-15, PERK, and ANXA1 genes prevented lipid accumulation, while knock-down of the IL-7 and IL-7R genes led to increased intracellular lipid accumulation during incubation of MDM with atherogenic LDL. Seventeen overexpressed genes and 189 underexpressed genes were obtained in the DGE analysis, which allowed us to discover 20 upregulated and 86 downregulated metabolic pathways, a number of which are associated with chronic inflammation and insulin signaling. We also elucidated 13 master regulators of cholesterol accumulation that are immune response-associated genes.Conclusion:Thus, it was discovered that 5 inflammation-related master regulators may be involved in lipid accumulation in macrophages. Therefore, the pro-inflammatory response of macrophages may trigger foam cell formation rather than the other way around, where intracellular lipid accumulation causes an inflammatory response, as previously assumed.

A comprehensive immunoinformatic analysis of chitin deacetylase’s and MP88 for designing multi-epitope vaccines against Cryptococcus neoformans

Cryptococcus neoformans causes life-threatening pneumonia and meningitis and is regarded as one of the leading killers of immunocompromised individuals. There is currently no vaccine against this pathogen. Recently, WHO placed it at the top among the critical priority groups in the fungal priority pathogens to accelerate the development of effective treatments. Numerous studies suggested the potential of subunit vaccines to overcome the challenges associated with live and inactivated whole-cell vaccines. Therefore, this study exploited integrated reverse vaccinology and immunoinformatic approach to construct and characterize multi-epitope vaccines targeting chitin deacetylases (Cda1, Cda2, Cda3) and MP88 of C. neoformans. 4 CTL, 8 HTL and 6 B cell epitopes were fused with different adjuvants and appropriate linkers to design two multi-epitope vaccines (VC1 and VC2). Both chimeric constructs were predicted to be highly antigenic, non-allergenic, non-toxic, soluble and had satisfactory physicochemical properties. Molecular docking and binding free energy calculation revealed strong binding interactions between vaccine constructs and human TLRs (TLR-2 and TLR-4). Classical MD Simulation and Normal mode analysis verified the stability of the vaccine-TLR complex in the biological environment. Codon adaptation, cloning and in silico expression suggested the efficient expression of recombinant vaccine proteins in E. coli. Both candidates also generated robust immune profiles comprising innate, adaptive and humoral immune responses. Taken together, experimental validations of our findings through extensive in vitro and in vivo testing might provide an effective vaccine for prophylactic control of C. neoformans.Communicated by Ramaswamy H.
