Introduction

Involved in transcription regulation by sequestering in the cytoplasm nuclear receptor coactivators such as NCOA1, NCOA2 and NCOA3 (PubMed:17476305). Involved in regulation of caspase-independent apoptosis by sequestering the proapoptotic factor AIFM1 in mitochondria (PubMed:22371500). Pro-apoptotic stimuli can induce its proteasomal degradation allowing the translocation of AIFM1 to the nucleus to induce apoptosis (PubMed:22371500). Involved in the negative control of vitamin D receptor signaling pathway (PubMed:24671081). Involved in actin stress fibers formation through its interaction with ARHGDIA and the regulation of the Rho signaling pathway (PubMed:17996375, PubMed:25961457). May thereby play a role in cell adhesion and migration, regulating for instance podocytes migration during development of the kidney (PubMed:25961457). Through the Rho signaling pathway may also regulate cell proliferation (By similarity).

Hydroa vacciniforme lymphoproliferative disorder, a clinicopathologic and genetic analysis

Hydroa vacciniforme lymphoproliferative disorder (HV-LPD) is a rare Epstein-Barr virus (EBV)-associated disease and the systemic form shows a propensity to progress and some cases may eventually develop a disease simulating NK/T-cell lymphoma or even aggressive NK-cell leukemia. We report 12 patients with systemic HV-LPD of median age 15.8 years with dermatosis involving the face, trunk, extremities and serous membranes. Ten of 12 patients (83%) had systemic symptoms including 9 with prominent facial edema and 2 with bone marrow involvement. All cases were positive for EBER and CD3 and lacked CD20. Additional positive markers included CD8 in 11 of 12 (91.6%), CD56 in 1 of 12 (8.3%), granzyme B in 6 of 7 (85.7%), TIA1 in 2 of 2 (100%) and CD30 in 1 of 3 (33.3%). Two cases studied demonstrated monoclonal TCR-γ and TCR-β rearrangements. Five cases were sequenced and showed recurrent mutations in ALMS1, ALPK2, BCORL1, KANK2, KMT2D, NCOR1, PTPRD, RHOA, TNIK, TP53 and ZFHX3, and single cases showed a variety of mutations including BCOR, CREBBP, DDX3X, DMXL2, IRF4, IRF8, KDM6A, MGA, NCOR2, PLCG1, PRDM1, RNF213, SMARCA4, STAT5B and TET2 mutation. Most patients were treated with immunomodulating therapy, two received methotrexate, three received multiagent chemotherapy and one underwent hematopoietic stem cell transplant. Follow-up was available in 10 patients of whom 6 died of disease and one was alive without disease. Our results showed that patients with persistent/progressive systemic HV-LPD have a poor prognosis and do not respond well to chemotherapy. The mutation spectrum bore some resemblance to extranodal NK/T lymphomas but also had notable differences.

Identification of biomarkers associated with pathological tumor staging and their utility in the diagnosis and prognosis of prostate cancer

AbstractObjectivePathological tumor (pT) staging plays a crucial role in prostate cancer (PCa) diagnosis. This study aimed to identify pT stage-associated biomarkers and explored their utility in PCa prognosis.MethodsGSE69223 was used to identify potential targets differentially expressed between level 2 of pT staging (pT2) and level 3 of pT staging (pT3). Quantitative reverse transcriptase–polymerase chain reaction and immunohistochemistry were performed on tissues from patients with PCa to screen the pT stage-associated targets and to explore the prognostic value of these targets in PCa.ResultsCENPI and SLC38A11 were most significantly upregulated, whereas ANO6 and KANK2 were mostly decreased in pT3 tumors compared with pT2 staging. ANO6 levels were negatively associated with preoperative prostate-specific antigen (PSA) levels, lymph node staging (N staging), Gleason score, and overall survival (OS); CENPI was positively associated with preoperative PSA levels, N staging, and OS, but was not associated with the Gleason score; SLC38A11 and KANK2 were not associated with OS. ANO6 and KANK2 were correlated with neutrophil markers, whereas CENPI was correlated with macrophage M2 types.ConclusionWe identified 4 reliable PCa biomarkers associated with pT staging that would be valuable for diagnosing and determining PCa prognosis.

Pan-Cancer Analysis of KANK2: Clinical and Molecular Insights into Tumor Progression and Therapeutic Implications

BACKGROUNDKANK2, a gene crucial for cell migration and movement, is implicated in neoplastic and non-neoplastic diseases. This study aimed to analyze KANK2's expression and its diagnostic and prognostic significance across 33 cancers using multiple online databases.METHODSThis study aimed to comprehensively analyze KANK2 in 33 cancers using The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx) database. Multiple web platforms and software were used for data analysis, including R, Cytoscape, HPA, TISIDB, UALCAN, GEO, cBioPortal, STRING, GSCALite, and CancerSEA. WB and qPCR experiments were used to verify the results.RESULTSKANK2 is widely expressed in various tissues and has significant diagnostic value in multiple cancers, with AUC values exceeding 0.75 in 13 cancer types. Survival analysis indicated that KANK2 expression is significantly associated with overall survival (OS) and disease-specific survival (DSS) in several cancers. KANK2 expression varied significantly across different molecular and immune subtypes and was associated with specific genetic mutations and DNA methylation patterns. Functional state analysis highlighted correlations with processes such as EMT, angiogenesis, and apoptosis. GSEA identified pathways related to proliferation, migration, and extracellular matrix remodeling. The key interacting proteins were identified by PPI network analysis, and the sensitive drug molecules were found by GSCA database. The results were also confirmed by two GEO datasets and WB and qPCR results.CONCLUSIONKANK2 serves as a valuable biomarker for diagnosis and prognosis in various cancers, and its expression is intricately linked to multiple molecular and cellular processes, offering potential therapeutic targets for future research.
