Introduction

Lysosomal thiol reductase that can reduce protein disulfide bonds. May facilitate the complete unfolding of proteins destined for lysosomal degradation. Plays an important role in antigen processing. Facilitates the generation of MHC class II-restricted epitodes from disulfide bond-containing antigen by the endocytic reduction of disulfide bonds (By similarity). Facilitates also MHC class I-restricted recognition of exogenous antigens containing disulfide bonds by CD8+ T-cells or crosspresentation (By similarity).

Integrative gut microbiota, metabolomics and proteomics studies unraveled the mechanism of Shaoteng decoction in treating Sjogren's syndrome

BACKGROUNDSjögren's syndrome (SS) is a complicated autoimmune disorder, encompassing multifaceted pathogenesis of inflammatory response, immune dysregulation and metabolic abnormalities. Shaoteng Decoction (STD) is a type of traditional Chinese medicine preparation that has been shown to effectively improve inflammatory damage and immune dysfunction in patients with SS. Nevertheless, the exact mechanism has not been unspecified.PURPOSEThis work aims to determine the mechanism of STD treatment on SS, identifying potential therapeutic targets and their relationships.METHODSNon-obese diabetic mice served as a disease model. This study analyzes potential signaling pathways of STD treatment for SS through network pharmacology, and assesses the role of STD in reducing inflammatory damage using pathological staining, ELISA, and immunohistochemistry. Additionally, the study apply gut microbiota, metabolomics, and proteomics analyses to identify the key microbiota, metabolites and proteins, aiming to find potential action targets of STD. We use Western blotting and immunohistochemistry to verify the authenticity of the relevant targets and study the interactions among gut microbiota, metabolites, and proteins.RESULTSProteobacteria is the important intestinal bacteria, Bile Acid Biosynthesis is the main metabolic pathway, IfI30, Ndufv3, and Ndufs6 are the crucial differential expressed proteins. Moreover, there is a strong correlation among the three. STD treats SS by reducing the abundance of Proteobacteria, increasing Bile Acid Biosynthesis, decreasing IfI30 expression, and increasing the expression of Ndufv3 and Ndufs6.CONCLUSIONSTD inhibits inflammatory responses, improves immune dysregulation and energy metabolism abnormalities, and prevents the progression of SS through regulating the gut microbiota, enhancing Bile Acid Biosynthesis, and modulating proteins expression levels.

Effects of dietary oregano essential oil on digestive tissue structure and function, antioxidant and immune responses and gut microbiota of red swamp crayfish (Procambarus clarkii)

Modifying the diet is an effective strategy to enhance the production and health of farmed red swamp crayfish (Procambarus clarkii). In this study the effects of adding dietary oregano essential oil (OEO) at 0, 100, 300, and 500 mg/kg concentrations were investigated in red swamp crayfish after a 3-week feeding trial. Histology showed no negative impacts on the structural integrity of the hepatopancreas and intestine. Total protease activity in the hepatopancreas, hemolymph alkaline and acid phosphatase activity, hemolymph superoxide dismutase activity and hepatopancreas glutathione levels were higher in the 500 mg/kg OEO group than in the 0 mg/kg group. No significant differences were observed in hepatopancreas phenoloxidase activity or hemolymph malondialdehyde content between treatment groups. Supplementing the diet with OEO upregulated the expression of antioxidant and immune-related genes in the hepatopancreas including metallothionein, gamma interferon-inducible lysosomal thiol reductase, and catalase in the 500 mg/kg OEO group relative to the 0 mg/kg control. Ferritin gene expression was unaltered. The intestinal microbiota of crayfish fed OEO showed an increased relative abundance of Proteobacteria and Actinobacteriota, and a lower abundance of Firmicutes. This study demonstrates the potential of OEO as a novel feed additive for enhancing antioxidant capacity and the immune response in red swamp crayfish. This research addresses the growing need for sustainable aquaculture practices and highlights the application of plant-derived bioactivities in improving crustacean health.

Whole exome sequencing for identifying rare genetic variants related to idiopathic granulomatous mastitis

AbstractBackgroundsTo reveal rare genetic factors that cause susceptibility to idiopathic granulomatous mastitis (IGM).MethodsWhole exome sequencing (WES) was performed in 30 patients with histopathologically diagnosed idiopathic granulomatous mastitis. WES analysis mainly focused on 317 genes linked to autoimmunity, autoinflammation, and immune dysregulation.ResultsA total of 141 variants were detected in 100 genes. The 40% (12/30) of patients had pathogenic or likely pathogenic variants. The pathogenic/likely pathogenic variants were 10.6% of all variants, and the rest of the variants (89.4%) were classified as VUS. Most of the variants were heterozygous (97.2%) only 4 variants (2.8%) were homozygous. Pathogenic/likely pathogenic variants were detected in FCGR1A, MPO, F5, IL36RN, CLUH, C9, NAXD, PROC, THRB, IFI30, COQ2, RNASEH2B, and SLC29A3 genes. The highest number of variants were detected in UNC13D, VPS13B, EPHB4, NLRP12, TCIRG1, TOM1, IRF9, and PIK3CG.ConclusionUp to date, our study is the first whole exome sequencing study of IGM patients which aims to find out the rare variants related to etiopathogenesis of the disease. We identified 141 single nucleotide variants of 100 genes, and most of these variants were found in innate immunity-related genes. The current study provides clues for identifying the etiologic factors and designing further functional studies in this rare disease with unknown etiopathogenesis. Key Points•Autoimmunity/autoinflammation-related genetic factors are blamed for etiopathogenesis of idiopathic granulomatous mastitis (IGM).•Mutation in genes related to innate immunity, especially in macrophage functions and phagocytosis, may lead to IGM susceptibility.•Potential candidate genes for genetic susceptibility to IGM may shed light for new treatment options.
