Mass spectrometric detection of keratins in tear fluid

PURPOSEKeratin contamination is a common problem in mass spectrometry proteomic analyses, particularly in bottom-up mass spectrometry. The purpose of this study was to determine the protein contaminants introduced during the proteomic analysis of tear fluid.METHODSHuman tear fluid samples were collected using Schirmer strips. Proteomic analyses were performed using liquid chromatography-tandem mass spectrometry (LC-MS/MS) on blank Schirmer strips and tear fluid samples, with empty vials serving as controls for assessing environmental contaminant proteins.RESULTSWe detected 26 contaminant proteins (18 keratins and 8 non-keratins). 98.2% of the total protein contamination can be attributed to the 9 keratins, including KRT10 (23.6%), KRT1 (23.5%), KRT2 (15.7%), KRT14 (7.6%), KRT16 (7.0%), KRT5 (6.1%), KRT9 (5.9%), KRT6B (4.6%), and KRT6A (4.3%). A comparison to the proteomic profile of blank Schirmer strips and controls (empty vials) found a strong correlation (R² = 0.9753), indicating that these proteins were not from the blank Schirmer strips but are environmental contaminants. On the other hand, several keratins including KRT19, KRT13, KRT4, KRT7, KRT15, KRT8 and KRT18 were present in tear fluid, but either not detected or were negligible in blank strips. Another set of keratins, including KRT5, KRT6A, KRT14, KRT16, and KRT17, were identified as components of tear fluid as well as environmental contaminants.CONCLUSIONSThis study revealed nine major contaminant keratins in the mass spectrometry analysis. Several other keratins were identified as constituents of tear fluid. Background subtraction is necessary for the accurate analysis of tear fluid using mass spectrometry.

Microscopic and Molecular Analysis of the Urethral Plate: A Step Towards Improved Hypospadias Surgery Outcomes

OBJECTIVETo investigate the microscopic and molecular structure of the urethral plate in hypospadias patients compared to normal urethral tissue, focusing on epithelial characteristics, cytokeratin expression, vascular patterns, and extracellular matrix components, including elastin fibers.METHODSWe analyzed 8 full-thickness mid urethral plate samples from pediatric hypospadias patients using immunohistochemistry to evaluate epithelial structure, cytokeratin expression (PanCK, CK7, CK10, CK13, CK14), vascular patterns (vWF, CD31), and extracellular matrix components (elastin). Control tissues included fetal urethras and penile urethras from gender confirmation surgery.RESULTSThe hypospadias urethral plate exhibited a multi-layered epithelium, contrasting with the single-layered normal urethral epithelium. Cytokeratin expression differed significantly: CK7 was predominantly negative in hypospadias samples but strongly positive in controls; CK10 showed variable expression; CK13 and CK14 were consistently positive in hypospadias samples, with CK14 exhibiting strong expression in the basal layer. Vascular analysis revealed fewer subepithelial capillaries and smaller vascular spaces in hypospadias tissue. Elastin expression was positive in hypospadias samples, while fetal urethra showed lower expression and adult urethra displayed distinct elastin fiber bundles. Minimal smooth muscle tissue was observed in hypospadias and fetal samples compared to adult controls.CONCLUSIONThe urethral plate in hypospadias patients demonstrates significant structural and molecular differences from normal urethral tissue, suggesting a developmental delay or arrest. These differences may contribute to surgical challenges and post-operative complications. Understanding these unique tissue characteristics could inform the development of novel surgical techniques or tissue engineering approaches to improve hypospadias repair outcomes.

Transcriptome analysis identifies potential molecular mechanisms for growth of fast muscle in Chinese perch juvenile

Chinese perch (Siniperca chuatsi) is an important commercial fish species in China. Understanding the molecular mechanisms of growth and development of skeletal muscle is helpful for selection breeding and improving the growth rate of Chinese perch. We analyzed histological and transcriptomic differences in fast muscle of Chinese perch between 30 days post hatching (dph) and 60 dph using histological sections and high-throughput RNA-Seq. The results showed that the diameter of muscle fibers in 30 dph Chinese perch was mainly distributed in range of 30 - 40 μm, and that of 60 dph was primarily in the range of 40 - 50 μm. 34 differentially expressed genes (DEGs) were identified in the fast muscle of Chinese perch between 30 and 60 dph, of which 9 were up-regulated and 25 were down-regulated (60 dph vs 30 dph). The DEGs, including MYH4, ENO3, Bag3, krt13 and krt18, are associated with muscle cell differentiation and fusion in Chinese perch. The analysis of the protein-protein interaction network of DEGs revealed that FOS, junb and EGR1 may involve in the development of fast muscle. KEGG enrichment results showed that the up-regulated genes in the 60 dph were associated with several pathways related to metabolism and protein synthesis, such as glycosphingolipid biosynthesis and aminoacyl-tRNA biosynthesis. The results suggest that the development of fast muscle in Chinese perch from 30 to 60 dph is accompanied by an increase in muscle fiber diameter and changes in gene expression related to muscle cell differentiation and protein synthesis.
