Chromosome 22q Allelic Losses at Microsatellite Loci in Human Astrocytic Tumors

Common regions of deletion(s) on chromosome 22q and the correlations between loss of heterozygosity and patient survival were analysed in 18 deoxyribonucleic acid samples from astrocytic tumors (3 astrocytomas, 5 anaplastic astrocytomas, and 11 glioblastomas) and matched normal brain tissues. The polymerase chain reaction products using five microsatellite markers were electrophoresed on polyacrylamide gels and the ethidium bromide stained bands were photographed. Loss of heterozygosity was observed in 14 (74%) of 19 samples, with similar incidences in astrocytomas, anaplastic astrocytomas, and glioblastomas (67%, 80%, and 82%, respectively). The locus D22S300 (q12.1-q13.1) was most frequently involved, with loss of heterozygosity in eight (80%) of 10 informative glioblastomas at this locus. Increased loss of heterozygosity during tumor progression or recurrence was seen in two patients at the D22S300 (q12.1-q13.1) and TOP1P2 (q11.2-q13.1) loci. No correlation between loss of heterozygosity on chromosome 22 and the postoperative survival was found. These findings suggest that loss of heterozygosity on chromosome 22q probably occurs quite frequently in astrocytic tumors. The chromosome segment 22q12.1-q13.1, around the D22S300 locus, may be the common region of deletion in glioblastomas.

We have initiated a genetic analysis of the physiologically important enzyme type I DNA topoisomerase in mouse. The exon-intron structures of the 5' part and the 3' part of the active gene, Top-1, were determined and shown to be quite similar to those of the previously determined human gene TOP1. The active mouse gene was mapped to the distal Chromosome (Chr) 2. In addition, the mouse genome contains one truncated processed topoisomerase-I-related pseudogene (retroposon), Top-1ps, on Chr 16. The Top-1ps locus, together with the immunoglobulin-lambda-light-chain locus, defines an additional conserved linkage group common to murine Chr 16 and human Chr 22, the site of the human pseudogene TOP1P2. The mapping data suggest that the pseudogene was established before mammalian radiation. Structural features, shared by the mouse and the human pseudogene, support this possibility.

The cDNA clone T1B was used to detect a 3-allele TaqI restriction fragment length polymorphism (RFLP) in the human topoisomerase I pseudogene locus on chromosome 22q11.2-13.1.Band sizes observed were 8.4 kb (A1), 7.5 Kb (A2), and 5.2 Kb (A3) whose resp. frequencies were 0.34, 0.59, and 0.07 in 70 unrelated caucasians.Constant bands were observed at 6.5, 4.9, 3.9, 3.3, 2.8, 2.1, 1.6, 1.4, and at 1.0 kb.The 2.1-kb constant band mapped to chromosome 1q, whereas all other constant bands mapped to chromosome 20 which contains the active gene for topoisomerase 1.Co-dominant segregation of the RFLP was demonstrated in a 3-generation family.
