Introduction

Nucleolar protein that acts as a regulator of RNA polymerase I by connecting RNA polymerase I with enzymes responsible for ribosomal processing and modification (PubMed:12777385, PubMed:26399832). Development of a nanobody-based immunoassay for the analysis of the disinfectant triclosan in water

Triclosan (TCS), a broad-spectrum antimicrobial agent in pharmaceuticals and personal care products, necessitates environmental monitoring due to its antimicrobial properties and widespread in the environment. In this study, two anti-TCS single-domain antibodies (i.e., nanobodies, Nbs), T1 and T2, were isolated from a phage-displayed Nb library derived from a camel immunized with a mixture of TCS immunogens. The T1-based enzyme-linked immunosorbent assay (ELISA) exhibited a better sensitivity to TCS than the T2-base ELISA. Motivation at enhancing specificity, sensitivity, and stability of Nb-based immunoassays promoted exploring use of a bivalent strategy to enhance performance. The bivalent Nb T1-T1 was tandemized via a linker-(GGGGS)3- between. The thermal stability of the bivalent Nb was improved in comparison with that of a monovalent Nb. The sensitivity of T1-T1-based ELISA, with an IC₅₀ value of 4.3 ng mL^-1 of TCS, was improved approximately 2-3 fold in comparison to those of T1-or T2-based ELISAs (8.5 and 14.6 ng mL^-1, respectively). The average recovery of TCS from water samples measured with T1-T1-based ELISA was in the range of 99-125 %, which correlated well with that measured by a high-performance liquid chromatography (HPLC) method (R² > 0.99). TCS in river water samples was detected by the resultant ELISA and an HPLC method, showing a satisfactory correlation.

Protein-inorganic hybrid flowers with a two-stage accelerated strategy for stimulated activation of CRISPR/Cas12a enhance polynucleotide kinase biosensing

Polynucleotide kinases (PNK) play a crucial role in DNA damage repair and are closely associated with specific diseases, making them promising targets for therapeutic intervention. In this study, we propose a two-stage accelerated strategy that utilizes protein-inorganic hybrid flowers (PHFs) to enhance the performance of the terminal deoxynucleotidyl transferase (TdT)-combined CRISPR/Cas12a system (TCS) for efficient detection of PNK activity. In TCS, the participation of PHFs confines the substrate probes (SPs) to a limited space, thereby significantly enhancing the local concentration of phosphorylated 3' termini of SPs and effectively promoting the enzymatic reaction kinetics as the first step in the accelerated strategy. Upon encountering the target PNK, the phosphorylated 3' termini were promptly recognized and dephosphorylated to 3'-OH termini. Subsequently, TdT catalyzed the assembly of deoxyadenosine triphosphates (dATPs) without a template, rapidly activating the CRISPR/Cas12a system by forming multiple polyadenine (poly-A) chains. PHF-fixed poly-A chains then substantially boosted the localized concentration of CRISPR/Cas12a systems and vastly enhanced their efficacy in cleaving reporter nucleic acids. Our findings indicated that the spatial confinement effect facilitated by PHFs promoted frequent molecular collisions and accelerated multiple enzymatic reactions. The developed sensing strategy allows for the detection of PNK activity within a linear range of 0.001-1 U/mL, with a detection limit of 1.82 × 10^-4 U/mL. Additionally, this strategy has been successfully applied to detect PNK activity in cell extracts and to screen for PNK inhibitors. Owing to these advantages, PNK can be rapidly and accurately detected with a high sensitivity, specificity, and biostability.

Analysis of the two-component system gene family and the positive role of CsRR5 in cold stress response in tea plants

The two-component system (TCS), a ubiquitous signaling network consisting of histidine kinases (HKs), phosphotransfers (HPs), and response regulator proteins (RRs), participates in various functions, including responses to abiotic stresses. However, a comprehensive identification of TCS genes in tea plants is still lacking. Here, we identified 60 CsTCS members in tea plants, including 23 HKs, 10 HPs, and 27 RRs. The analysis of promoter cis-regulatory elements indicated that CsTCS genes are involved in phytohormone signaling, stress responses, and growth and development. The expression of CsETR1, CsETR2b, CsERS1b, and CsEIN4b from the HK subfamily was down-regulated by ethylene, whereas CsHK2b and CsHK3a were down-regulated by cytokinin. Conversely, CsHK4a and CsHK4b were up-regulated by cytokinin. CsTCS genes were widely expressed in various tissues, with the majority associated with multiple stresses. Furthermore, we demonstrated that suppressing the type-A response regulator CsRR5 in tea plants reduced cold tolerance and the expression of CBF-COR pathway genes, indicating that CsRR5 positively regulates the cold stress response through the CBF pathway. Therefore, our study establishes a connection between the two-component system and its downstream regulation in tea plant cold stress response.
