Scrutinizing the human TEX genes in the context of human male infertility

AbstractBackgroundInfertility affects around 15% of all couples worldwide and is increasingly linked to variants in genes specifically expressed in the testis. Well‐established causes of male infertility include pathogenic variants in the genes TEX11, TEX14, and TEX15, while few studies have recently reported variants in TEX13B, TEX13C, FAM9A (TEX39A), and FAM9B (TEX39B).ObjectivesWe aimed at screening for novel potential candidate genes among the human TEX (“testis expressed”) genes as well as verifying previously described disease associations in this set of genes.Materials and methodsTo this end, we screened the exome sequencing data of 1305 men, including 1056 crypto‐ and azoospermic individuals, and determined cell‐specific expression by analyzing testis‐specific single‐cell RNA sequencing data for genes with identified variants. To investigate the overarching role in male fertility, we generated testis‐specific knockdown (KD) models of all 10 orthologous TEX genes in Drosophila melanogaster.ResultsWe detected rare potential disease‐causing variants in TEX10, TEX13A, TEX13B, TEX13C, TEX13D, ZFAND3 (TEX27), TEX33, FAM9A (TEX39A), and FAM9B (TEX39B), in 28 infertile men, of which 15 men carried variants in TEX10, TEX27, and TEX33. The KD of TEX2, TEX9, TEX10, TEX13, ZFAND3 (TEX27), TEX28, TEX30, NFX1 (TEX42), TEX261, and UTP4 (TEX292) in Drosophila resulted in normal fertility.DiscussionBased on our findings, the autosomal dominant predicted genes TEX10 and ZFAND3 (TEX27) and the autosomal recessive predicted gene TEX33, which all three are conceivably required for germ cell maturation, were identified as novel potential candidate genes for human non‐obstructive azoospermia. We additionally identified hemizygous loss‐of‐function (LoF) variants in TEX13B, TEX13C, and FAM9A (TEX39A) as unlikely monogenic culprits of male infertility as LoF variants were also found in control men.ConclusionOur findings concerning the X‐linked genes TEX13B, TEX13C, and FAM9A (TEX39A) contradict previous reports and will decrease false‐positive reports in genetic diagnostics of azoospermic men.

AbstractAs the world population continues to increase to unsustainable levels, the importance of birth control and the development of new contraceptives are emerging. To date, male contraceptive options have been lagging behind those available to women, and those few options available are not satisfactory to everyone. To solve this problem, we have been searching for new candidate target proteins for non-hormonal contraceptives. Testis-specific proteins are appealing targets for male contraceptives because they are more likely to be involved in male reproduction and their targeting by small molecules is predicted to have no on-target harmful effects on other organs. Using in silico analysis, we identified Erich2, Glt6d1, Prss58, Slfnl1, Sppl2c, Stpg3, Tex33, and Tex36 as testis-abundant genes in both mouse and human. The genes, 4930402F06Rik and 4930568D16Rik, are testis-abundant paralogs of Glt6d1 that we also discovered in mice but not in human, and were also included in our studies to eliminate the potential compensation. We generated knockout (KO) mouse lines of all listed genes using the CRISPR/Cas9 system. Analysis of all of the individual KO mouse lines as well as Glt6d1/4930402F06Rik/4930568D16Rik TKO mouse lines revealed that they are male fertile with no observable defects in reproductive organs, suggesting that these 10 genes are not required for male fertility nor play redundant roles in the case of the 3 Glt6D1 paralogs. Further studies are needed to uncover protein function(s), but in vivo functional screening using the CRISPR/Cas9 system is a fast and accurate way to find genes essential for male fertility, which may apply to studies of genes expressed elsewhere. In this study, although we could not find any potential protein targets for non-hormonal male contraceptives, our findings help to streamline efforts to find and focus on only the essential genes.

[Preparation and application of rabbit polyclonal antibody against mouse Tex33].

Objective To prepare rabbit-antimouse testis expressed 33 (Tex33) polyclonal antibody and detect the expression of Tex33 during mouse spermatogenesis. Methods Tex33 open reading frame was amplified by PCR from mouse testis cDNA. The PCR product was finally sub-cloned into pET-30a vector. The prokaryotic expression plasmid pET-30a-Tex33 was constructed and then transformed into E. coli BL21. Tex33 prokaryotic protein was induced by IPTG and then purified by Ni-NTA affinity chromatography. Tex33 polyclonal antibody was obtained from adult male New Zealand white rabbits immunized with purified Tex33 protein. ELISA, Western blotting and immunofluorescence assays were used to determine the potency and specificity. Results pET-30a-Tex33 recombinant vector was successfully constructed. After induced by IPTG, the recombinant Tex33 protein could be expressed effectively. The titer of polyclonal antibody was 1:1 000 000. Western blotting showed that the antibody could recognize Tex33 protein in mouse testis. Immunofluorescence assay revealed that Tex33 gene was expressed in spermatids and sperms of adult mouse testis. And Tex33 was located on the acrosome and flagellum of spermatozoa. Conclusion We have prepared rabbit-antimouse Tex33 polyclonal antibody with high specificity and found that Tex33 gene was expressed in mouse testis.
