Potential Molecular Markers for Cholecystic Inflammation-Induced Carcinogenesis Based on RNA-Seq Gene Screening

Background:Uncontrolled inflammation plays an important role in the initiation and progression of tumors. The repeated circulation and continuous stimulation of gallbladder epithelium caused by gallstones is an important risk factor for gallbladder cancer.Methods:To study pathogenesis, samples were collected for chronic cholecystitis caused by gallstones and early and advanced gallbladder cancer with gallstones and subjected to RNA-seq analysis. Gene Ontology and Kyoto Gene and Genome Encyclopedia analyses were used to elucidate the protein–protein interaction network and identify differentially expressed genes.Results:Nine potential molecular markers, VTN, CHAD, AKR1C4, ABCC2, AOX1, ADH1A, ADH1C, PLA2G2A, and CYP4F3, with elevated expression gradients in cholecystitis and early and advanced gallbladder cancer, were identified. Using qPCR and immunohistochemistry on clinical tissues, we confirmed three factors, VTN, CYP4F3, and AOX1, to be worthy of further research. To demonstrate that these three genes are potential molecular markers for gallbladder cancer, their cellular biological functions were confirmed in gallbladder cancer cell lines through siRNA transfection.Conclusion:The potential molecular markers CYP4F3, VTN, and AOX1 for cholecystitis and different stages of gallbladder cancer were identified. Further studies on differentially expressed genes vital in gallbladder cancer progression can help provide potential targets for the early diagnosis and treatment of gallbladder cancer.

Integrated metabolome and transcriptome analysis reveals key genes and pathways associated with egg yolk percentage in chicken

Yolk percentage is a critical index in the egg product industry, reflecting both nutritional value and economic benefits. To elucidate the underlying mechanisms that contribute to variations in egg yolk percentage, we performed integrated transcriptome and metabolome analyses on the liver, ovary, and magnum tissues of Rhode Island Red chickens with high and low yolk percentages. A total of 322 differentially expressed genes (DEGs) and 128 significantly differential metabolites (SDMs) (VIP>1, P < 0.05) were identified in the liver, whereas 419 DEGs and 215 SDMs were detected in the ovary, and 238 DEGs along with 47 SDMs were found in the magnum. In the liver, genes such as HMGCR, DHCR7, MSMO1, and CYP7A1 were linked to cholesterol metabolism, essential for steroid hormone synthesis and yolk formation, while ACACB, ACSL1, ACSL4, LPL, and SGPP2 were involved in fatty acid biosynthesis, a key process for supplying energy and structural components of the yolk. In the ovary, COL6A6, COMP, CHAD, ITGA7, THBS2, and TNC contributed to extracellular matrix-receptor interactions, which are fundamental for follicle development and oocyte maturation. In the magnum, UGT1A1, MAOB, and ALDH3B2 participated in drug metabolism-cytochrome P450 and amino acid metabolism, ensuring a proper environment for egg white formation and potentially influencing nutrient allocation to the yolk. Metabolic pathway enrichment revealed that steroid hormone biosynthesis, glycerophospholipid metabolism, and betaine metabolism were predominant in the liver; pyruvate, taurine, and hypotaurine metabolism in the ovary; and phenylalanine metabolism in the magnum. Moreover, integrated analysis highlighted key metabolites and genes potentially regulating yolk deposition, including 7,8-dihydroneopterin and Pg 38:4 in the liver (related to immune modulation and lipid metabolism, respectively), thalsimine in the ovary, as well as DL-glutamine in the magnum, all of which may be crucial for maintaining metabolic homeostasis and supporting egg formation. Collectively, these findings deepen our understanding of how distinct molecular and metabolic pathways in the liver, ovary, and magnum orchestrate yolk proportion and deposition. Such insights may advance future strategies to improve egg quality and productivity in poultry breeding programs.

Transcriptomic insight into the underlying mechanism of induced molting on reproductive remodeling, performance and egg quality in laying hen

This study aimed to clarify the reproductive remodeling mechanism in enhancing production performance and egg quality during the fasting-induced molting process of laying hens. A total of two-hundred and forty 380-days-old Jingfen No. 6 laying hens, with an average laying rate of 78% were divided into four replicates, with 60 hens in each replicate to receive a four-stage molt induction experiment. The stages encompassed the pre-molt stage (T1), the molt stage (T2), the recovery stage (T3), and the second peak-laying stage (T4). The egg-laying rate and egg quality were recorded during all stages, and sample collection (serum, magnum of oviduct, ovary) was conducted at the end of each stage. The length and index of oviduct, the number of hierarchical follicles, and serum reproductive hormone levels were further measured, followed by the transcriptomic sequencing process on the magnum of the oviduct and ovarian tissues at each stage. Results showed that the fasting treatment induced atrophy of the oviducts, the disappearance of large yellow follicles in the ovaries, and the decrease in serum reproductive hormone levels compared to the pre-molt stage. Transcriptomic analysis revealed that differentially expressed genes were notably enriched in cytokine-cytokine receptor interactions, cell adhesion molecules, and the arachidonic acid metabolism signaling pathway during the remodeling phases of oviduct and ovary tissues. Key candidate genes such as BMPR1B, NEGR1, VTN, and CHAD emerged as pivotal in influencing reproductive function remodeling in molt-treated chickens. Additionally, genes associated with steroid biosynthesis showed significant up-regulation in the ovaries of molted hens, correlating positively with egg-laying rates. Furthermore, genes related to collagen and laminin displayed significant positive associations with Albumen height and Haugh unit values. The results indicate that fasting interventions might modulate the remodeling of reproductive functions in laying hens by altering cytokine-cytokine receptor interactions, cell adhesion molecules, and arachidonic acid metabolic pathways. Enhanced ovarian steroid biosynthesis and up-regulation of gene expression, including oviductal collagens post-molting, are crucial for enhancing laying rates and egg quality. These findings could offer novel thinking for refining molting protocols.
