High-effective, convenient and environmental-friendly MOFs-chitosan-glyoxal composite film for ceftizoxime adsorption: Behavior and mechanisms

In this work, a novel PCN-222-chitosan-glyoxal (PCN@CSG²) composite film was constructed by loading PCN-222 in chitosan substrate, and used for ceftizoxime adsorption. The results demonstrated that the PCN@CSG not only had excellent adsorption properties for ceftizoxime, but also maintained excellent structural properties in harsh environments (strong acids and alkalis), making it have good recycling performance. Specifically, the adsorption kinetics and isotherms investigation demonstrated that the adsorption process followed the pseudo-second-order kinetic model and Freundlich isotherm model respectively, indicating that it was a multilayer process mainly controlled by chemisorption. The PCN@CSG possesses excellent absorptive capacity of 561.7 mg·g^-1 and reaches equilibrium rapidly within 60 min, which is attributed to the structural advantages of PCN-222 and chitosan. The amino, carboxyl and hydroxyl functional groups of PCN-222 provide numbers of active sites and cationic chitin greatly promoted the electrostatic adsorption with negative ceftizoxime. In addition, the PCN@CSG has the advantages of renewable and environment-friendly for the biodegradation of chitosan. After five consecutive adsorption-desorption cycles, the removal rate was still higher than 90 %, confirming the excellent reusability of PCN@CSG. This work provided a great prospect for the design and application of shaped-MOFs composite materials for the removal of cephalosporins in environmental water.

Staphylococcus aureus, a facultative anaerobic gram-positive bacterial pathogen, has posed major threat to public health worldwide. Upon S. aureus infection, the host immune system is activated for clearance. However, intracellular S. aureus, which remains viable for an extended time, has evolved the ability to escape from immune response and extracellular antibiotics. One of possible strategies is the formation of persisters. Persistence is one of the major causes of S. aureus relapse infection but the underlying mechanisms remain obscure. Here, we identified two co-transcribed genes SA1833-SA1832 that are involved in persister formation in S. aureus. Dysfunction of SA1833 and/or SA1832 significantly reduces persister formation in the presence of ceftizoxime. Additionally, we found that the expression of SA1833 and SA1832 under the induction of oxidative stress and SOS response is strictly regulated by the LexA-RecA pathway. Interestingly, SA1833-SA1832 contributes to persister formation in an lrgA-dependent manner. Moreover, the mouse RAW264.7 macrophage infection model indicated that disrupting SA1833-SA1832 inhibits S. aureus from infecting macrophages and impairs its ability to survive in the intracellular environment.

Impact of ciprofloxacin with autophagy on renal tubular injury

Backgrounds:Renal tubular injury caused by oxidative stress and inflammation results in acute kidney injury. Recent research reported that antibiotics may protect renal tubules from progressive deterioration, but the underlying mechanism remains unclear. Therefore, we investigated the efficacy and mechanism of action of antibiotics against renal tubular injury.Methods:We screened ciprofloxacin, ceftizoxime, minocycline, and netilmicin and selected ciprofloxacin to examine further because of its low toxicity towards renal tubular cells. We evaluated the effect of ciprofloxacin on cell survival by analyzing apoptosis and autophagy.Results:Terminal deoxynucleotidyl transferase-mediated d-UTP nick end labeling (TUNEL) assay results showed that the ciprofloxacin group had less apoptotic cells than the control group. The ratio of cleaved caspase 3 to caspase 3, the final effector in the apoptosis process, was decreased, but the ratio of B-cell lymphoma 2 (Bcl-2)-associated X protein (Bax) to Bcl-2 located upstream of caspase 3 was not decreased in the ciprofloxacin group. Therefore, apoptosis inhibition does not occur via Bax/Bcl-2. Conversely, the levels of phosphorylated Bcl-2, and Beclin-1, an autophagy marker, were increased, and that of caspase-3 was decreased in the ciprofloxacin group.Conclusion:This indicates that ciprofloxacin enhances autophagy, increasing the amount of free Beclin-1 via phosphorylated Bcl-2, and inhibits caspase activity. Therefore, ciprofloxacin might protect against renal tubular injury through the activation of autophagy in the setting of acute kidney injury.
