European photopatch test baseline series: A 3‐year experience

BackgroundIn 2012, a consensus was reached regarding a baseline photopatch test series on the basis of the results of a European multicentre study.ObjectivesTo describe experience with the European photopatch test series.MethodsA retrospective analysis of 116 patients tested with the European photopatch test series between 2014 and 2016 was performed.ResultsFifty‐five positive photopatch test reactions in 25 subjects were recorded, most commonly caused by the topical non‐steroidal anti‐inflammatory drugs ketoprofen, dexketoprofen, and etofenomate. Organic ultraviolet (UV) absorbers constituted the second main category of agents eliciting positive photopatch test reactions. Among UV absorbers, benozophenone‐3 and octocrylene were the most frequent photoallergens. UV absorbers that have been introduced more recently rarely elicited positive photopatch test reactions. Positive patch test reactions were less commonly observed than positive photopatch test reactions, namely, 21 reactions in 14 patients.ConclusionsWe present the largest clinical experience with the European photopatch test baseline series hitherto reported. The results are similar to those underlying the above consensus process, reaffirming the usefulness of this series.

Aim: Histamine plays an important role in the pathogenesis of allergic diseases. Genetic variations in histamine receptors (HRH) may influence the expression of allergic diseases. This study analyzes the association between HRH variants and NSAID hypersensitivity reactions. Patients & methods: The authors analyzed copy number variations (CNVs) and common functional SNPs in genes HRH1, HRH2 and HRH4 in 442 unrelated patients with hypersensitivity to NSAIDs and in 414 healthy unrelated controls. Results: The authors identified, both in patients and control subjects, individuals carrying CNVs in HRH genes. The most common genotype corresponded to two copies of each gene, but carriers of one or three copies of HRH1 (5% of individuals), HRH2 (1.1%) and HRH4 genes (0.9%) were also identified. Conclusion: For the first time, we describe CNVs in human HRH genes. Neither common functional SNPs in HRH genes nor CNVs influenced the risk of developing hypersensitivity to NSAIDs. Solvent Versatility of Immobilized Amylose and Cellulose-Based Chiral Stationary Phases in Enantioselective LC Separation and Monitoring of Bio-Catalyzed Resolutions of Acidic Drugs in Non-Standard Organic Solvents

The solvent versatility of Chiralpak IA (amylose tris(3,5-dimethylphenylcarbamate)), Chiralpak IB (cellulose tris(3,5-dimethylphenylcarbamate)) and Chiralpak IC (cellulose tris(3,5-dichlorophenylcarbamate)) immobilized onto silica gel, are investigated for the enantioselective separation of a set of acidic drugs in liquid chromatog.Non-standard LC organic solvents like dichloromethane, Et acetate, THF, methyl-tert-Bu ether were used in mobile phase compositions and/or diluent agent for the analyte on all new columns.Furthermore, the enantioselective separations of the reported compounds were compared on both immobilized columns (Chiralpak IA and IB) and their conventionally coated versions (Chiralpak AD-H and Chiralcel OD-H, resp.), using a mixture of n-hexane/2-PrOH/TFA (80:20:0.1 volume/volume/v).The versatility of the immobilized Chiralpak IB in monitoring reactions performed in non-standard solvent was studied on a representative example consisting of the lipase-catalyzed enantioselective esterification of flurbiprofen with n-butanol in methyl-tert-Bu ether as organic solvent.
