Pressurized pyrolysis of mattress residue: An alternative to landfill management

Mattresses are a difficult waste to manage in landfills due to their large volume and low density. Pyrolysis treatment could reduce its volume while producing fuel or products valuable for the chemical industry. Pressurized pyrolysis at 400, 450, and 500 °C is carried out in a lab-scale autoclave at initial pressures 4.2, 8.4, and 16.8 bar. Product gas yield increases slightly along with elevated pressure as well as temperature. However, beyond 8.4 bar the initial pressure makes no discernible differences. CO and CO₂ are the major gas species followed by CH₄. CO contributes the most to the product gas energy content followed by C₃ species, C₂H₆, and H₂. Calculated energy content (heating value) is between 2 and 15 MJ·Nm^-3. In terms of product gas energy content, low pressure pyrolysis is favorable over high pressure pyrolysis. According to integration areas of chromatographic measurements the liquid phase contains up to 25 % of N-compounds, with benzonitrile being the most abundant, followed by toluene, o-xylene, and ethylbenzene. The solid char maintains constant properties across operating conditions, with carbon and energy contents of approximately 75 wt% and 30 MJ·kg^-1, respectively.

Thiamine pretreatment improves endotoxemia-related liver injury and cholestatic complications by regulating galactose metabolism and inhibiting macrophage activation

BACKGROUNDAs a major metabolic site, the liver is an important target organ of endotoxemia. High serum lipopolysaccharide (LPS) levels can cause hepatocyte necrosis and produce cholestasis, which results in severe liver injury. Contrastingly, thiamine (THA) has shown anti-inflammatory effects against severe infections and may be indicated for systemic endotoxemia treatment. Therefore, the present study was conducted to investigate the effective treatment of endotoxemia-induced liver injury with THA and the possible molecular mechanisms.METHODOLOGY/PRINCIPAL FINDINGSin vivo, We established two models of endotoxemia-induced liver injury at the in vivo level using LPS and bile duct ligation (BDL) + LPS, administering prophylactic THA intraperitoneally to mice. In vitro, the effects of THA on RAW264.7 and THP-1 administration of LPS-induced inflammatory macrophage activation were observed. Metabolomic analysis screening and subsequent validation experiments were also performed. THA has different degrees of preventive therapeutic effects on different causes of endotoxemia-induced liver injury, as evidenced by a decreased alanine aminotransferase (ALT) and decreased inflammatory factors. This study aimed to clarify the specific mechanism. We subsequently found that THA reduced the inflammatory macrophages produced by RAW264.7 and THP-1 in response to LPS. Additionally, THA reduced galactose liver accumulation and improved glucose metabolism. Moreover, Galectin-3 (Gal-3), as a point of interaction between macrophage activation and galactose metabolism mechanisms, was observed to inhibit Gal-3 expression by THA at both in vivo and in vitro levels.CONCLUSIONSThis study revealed that THA may be a viable prophylactic treatment option for the prevention of liver injury occurring in endotoxemia, which is associated with its effects on the modulation of Gal-3 to improve the inflammatory response and the inhibition of galactose metabolism. Additional evidence is provided for its clinical application.

Combination of planar chromatography with matrix-assisted laser desorption/ionization mass spectrometry for the analysis of biologically active compounds

MALDI mass spectra of 33 pharmacol. substances recorded from standard metal targets using four different matrixes (HABA, DHB, IAA and AT) have been obtained for the first time.They revealed only peaks for ions [M + H]⁺, [M + Na]⁺, and [M + K]⁺ with different intensities.It has been shown that MALDI spectra of the compounds can be obtained as a result of the desorption/ionization of analytes from spots on thin-layer chromatogram (TLC) (silica stationary phase) in the presence of the same tested matrixes.It has been found that graphite can be efficiently used as a matrix in combination of TLC with MALDI mass spectrometry, as it eliminates the danger of the broadening of spots on TLC plates in the treatment with matrix solution and reduces the time of sample preparation
