The Ad26.RSV.preF/RSV preF protein vaccine has previously demonstrated efficacyin protecting older adults against respiratory syncytial virus (RSV)-related lower respiratory tract disease in a phase 2b study. This study compared the immunogenicity of vaccine clinical trial material (CTM) representative of phase 2b clinical studies with CTM used in phase 3 clinical studies. A total of 248 adults aged 60-75 years, randomized in a 1:1 ratio, received one dose of either phase 3 CTM or phase 2b CTM. Solicited adverse events (AEs), unsolicited AEs, and serious AEs (SAEs) were assessed for 7-d, 28-d, and 6-month periods post-vaccination, respectively. RSV preF-ELISA antibody titers and RSV neutralizing titers were measured before and 14 d after vaccination. The phase 3 CTM-induced preF-ELISA response at Day 15, in terms of geometric mean titer, was shown to be non-inferior to that induced by phase 2b CTM. The RSV neutralizing antibody titers were also similar in the two groups at Day 15. The safety profile in terms of solicited AEs, unsolicited AEs, or SAEs was in general similar between the phase 3 CTM and phase 2b CTM groups, and solicited AEs were mostly mild to moderate in intensity. No related SAEs were reported, and no safety concerns were identified.

Quantitative Assessment of Bony Fish Tropomyosin Using a Monoclonal Antibody Competitive ELISA

Bony fish is one of the big-nine allergenic foods in the US. This study established a monoclonal antibody (mAb)-based indirect competitive enzyme-linked immunosorbent assay (ELISA) for the detection of bony fish tropomyosin (TM), a fish allergen. Immunoassay (Western blot and ELISA) was performed to characterize anti-TM mAb8F5 (target configuration, immunoaffinity, and species selectivity). Atlantic cod (Gadus morhua) tropomyosin (C_TM) and mAb8F5 were used for assay development. The immunosignal was captured by two light sources, i.e., colorimetry (cELISA) and luminescence (lELISA). During assay validation, cELISA had a wider working range (1-250 ng/μL) than lELISA (0.5-62.5 ng/μL). However, lELISA showed better sensitivity because of its lower half-maximal inhibitory concentration (IC₅₀, 8.8 ng/μL) than cELISA (37.1 ng/μL). The optimal cELISA was applied to measure C_TM-equivalents in 21 bony fish samples (0.03-4.2 mg/g) and laboratory-spiked samples (recovery rates of C_TM-spiked shrimp samples ranged from 104.7 to 120.8%). lELISA was used to quantify 5.25 μg C_TM residues on four food contact surfaces (3.4-6.9 μg from C_TM residues and 3.2-4.8 μg from Atlantic cod were detected). This immunoassay has the potential to surveil undeclared allergenic bony fish residues and validate the hygiene of food contact surfaces.

Effects of connective tissue massage and kinesiotaping in patients with fibromyalgia

BACKGROUNDFibromyalgia (FM) is a common soft tissue rheumatic disease. There is no established treatment plan for FM. Current treatments primarily focus on controlling symptoms and improving the quality of life. The aim of our study was to determine how connective tissue massage (CTM) and kinesiotaping (KT) affect pain and depression in patients with fibromyalgia, and to compare their effects on quality of life.METHODSThirty-four patients diagnosed with fibromyalgia were included. Participants divided into two groups. Group 1: CTM (n = 17, mean age = 35.47 ± 6.84 years), Group 2: KT (n = 17, mean age = 34.00 ± 5.46 years). This study was initiated by obtaining sociodemographic information. Pain (Visual Analogue Scale (VAS)), pain characteristics (McGill Pain Questionnaire), disease impact on patients (Fibromyalgia Impact Questionnaire (FIQ)), depression (Beck's Depression Inventory (BDI)), and quality of life (Short Form-36 (SF-36)) were evaluated. All participants received electrotherapy (TENS and infrared treatment) for 25 min, 5 days a week for 4 weeks. CTM was applied 3 days a week, KT was applied 2 days a week for 4 weeks.RESULTSA significant improvement with very large effect size was found in VAS, McGill, FIQ, BDI, and SF-36 variables in both groups (p < 0.05). The change after 4 weeks of treatment was similar between groups in terms of all parameters (p > 0.05).CONCLUSIONBoth CTM and KT improved pain, disease effects depression, and quality of life. These two interventions, which are not superior, can be added to the treatment program for patients with fibromyalgia.
