Currently, there is concern about food products to which new psychoactive substances (NPS) have been added illegally. However, rapid, high-sensitivity, and high-throughput screening methods for such harmful substances have not been reported yet. In this study, a high-throughput non-targeted screening strategy was developed based on UHPLC-Q/Orbitrap. First, optimized pretreatment methods for typical high-risk matrices were established. Second, an accurate, efficient, and scalable high resolution mass spectrometry database containing 206 NPS and related compounds was constructed. The fragmentation pathways of three different types of fentanyl-related compounds were then systematically investigated to enhance compound identification. Additionally, a non-targeted screening strategy was developed using warning ions derived from 24 fentanyl-related compounds, streamlining the identification of NPS. This strategy was applied to 100 food samples with possible illegal additives and demonstrated its effectiveness. In summary, this study provides robust technical support for regulatory authorities in combating the illegal addition of food products.

Passive sampling of contaminants of emerging concern in a Caribbean urban estuary of Puerto Rico

Contaminants of emerging concern (CECs) have not been extensively studied in Caribbean estuaries, particularly in Puerto Rico. Polar organic chemical integrative samplers were deployed in surface waters at five sampling sites in the San Juan Bay Estuary to investigate the occurrence and distribution of 137 CECs, resulting in the detection of 64 chemicals (51 pharmaceuticals and personal care products "PPCPs", 12 hormones plus sucralose) at least in one sampling site, and with 35 chemicals obtaining a 100 % detection frequency. Wastewater intrusion was implicated in the presence of CECs based on the detection of sucralose and other wastewater tracers. Condado Lagoon was the most polluted site impacted by untreated wastewater, showing the highest levels of 30 of the 64 detected CECs, and the largest proportion of detected CECs, particularly PPCPs. Levels of estrone and 17β-estradiol represent a high potential risk for adverse effects on aquatic organisms in this Caribbean urban estuary.

Using umbilical cord tissue to identify prenatal ethanol exposure and co-exposure to other commonly misused substances

Abstract:

Objective:

Substance misuse during pregnancy can result in a variety of poor pregnancy outcomes. Objective data reporting the prevalence of neonates born with ethanol metabolites (evidence of prenatal ethanol exposure) in their fluids or tissues are limited.

Study design:

A secondary analysis of umbilical cord tissue specimens received for routine toxicological analysis was conducted. Prevalences of ethyl glucuronide (EtG), a long-term direct ethanol biomarker, were determined using a new laboratory tool, LDTD-MSMS. Additionally, other commonly misused substances were determined using routine procedures.

Results:

Of 12,995 specimens, 238 (1.8%) specimens contained EtG. Concentrations of EtG ranged from 5 ng/g to 6679 ng/g (median 47 ng/mg; IQR: 16 ng/g, 203 ng/g). Of those 238 EtG-positive specimens, nearly 58% (N = 138) contained additional substances or metabolites.

Conclusion:

Self-report of substance use during pregnancy is under-reported. We have demonstrated co-exposure of substances with ethanol is higher than previous reports.
