In dogs anesthetized with pentobarbital, l-nordefrin was 100-200 times more active than its d isomer in raising blood pressure.Adrenaline was 2-2.5 times more active than l-nordefrin.In approx. equipressure doses l-nordefrin produced a somewhat greater increase in heart rate than adrenaline but was considerably less active in stimulating the retractor penis.In equipressure doses d-nordefrin produced greater contractions of the retractor penis than its l isomer.On the nonpregnant rat uterus in vitro adrenaline was 15 times more inhibitory than l-nordefrin, and the latter approx. 5000 times more active than its d isomer.The intravenous L.D.₅₀ ± S.E. of l-nordefrin in white mice was 12.6 ± 2.7 mg./kg.; it was 10 times more toxic than the d isomer, and approx. 1/5 as toxic as adrenaline. l-Nordefrin prolonged significantly the duration of local anesthesia.In a solution containing ravocaine-procaine, 1:10,000 racemic nordefrin produced a slightly longer duration than did 1:20,000 l-nordefrin.
