1) A preliminary dose-duration-effect study was conducted to investigate the dose-duration-effect relationship of the reference formulation of diflucortolone valerate cream (strength: 0.1% (30 g); licensed by LEO Pharma A/S; trade name: Nerisona®) in Chinese health study participants. This study was used to determine the dose-duration (ED50) and the proportion of participants expected to meet the "detector" criteria in the subsequent pivotal in vivo bioequivalence study. 2) Based on the results of the preliminary dose-duration-effect study, appropriate study conditions were designed. A pivotal in vivo bioequivalence study was conducted using WSEFKL Cream (strength: 10 g: 10 mg) provided by Tianjin Pharmaceutical Peace (Tianjin) Co., Ltd. as the test formulation and diflucortolone valerate cream (strength: 0.1% (30 g); trade name: Nerisona®) licensed by LEO Pharma A/S as the reference formulation. Investigating the Telomerase Downregulatory Potential of Steroidal Drugs Following Pharmacophore‐Based Rational; Computational and In Vitro Assessments

ABSTRACT:

Inhibiting telomerase could lead to telomere shortening, chromosomal instability, and eventually cell death in cancer cells. The use of steroids in cancer treatment is significant; besides, drug repurposing can be a quicker and less expensive method of drug discovery than de novo drug development. Accordingly, diverse 280 steroidal drugs—following a pharmacophore‐based rationale—were docked against telomerase receptor and compared to the co‐crystallized inhibitor (BIBR1532) as a reference standard. Additionally, a molecular dynamics simulation for 200 ns was performed to confirm the docking results. Moreover, the six steroidal drugs (Betamethasone disproportionate, Beclomethasone disproportionate, Clobetasone butyrate, Desonide, diflucortolone valerate, and Hydrocortisone butyrate) were selected for further in vitro investigation. The antitumor activities of the six steroidal drugs against H1299, HuH7, HCT116, A549, MDA‐MB‐231, MCF7, PC3, MG63, and A375 cancer cell lines, besides OEC and HSF normal cell lines, were evaluated and compared to doxorubicin (Dox) as a reference positive standard. Especially, Clobetasone butyrate represented the better cytotoxicity against seven cancer cell lines (H1299, HuH7, HCT116, MDA‐MB‐231, PC3, MG63, and A375) with IC 50 values of 12.56, 13.95, 12.00, 17.51, 11.69, 20.64, and 20.21 µg/mL, respectively. The outstanding antitumor six steroidal drugs were further investigated for their telomerase downregulatory potentials. All analogs recorded outstanding downregulatory results against telomerase, especially Clobetasone butyrate, Desonide, Diflucortolone valerate, and Hydrocortisone butyrate, where the protein expression for telomerase was downregulated up to 0.66‐, 0.76‐, 0.56‐, and 0.73‐fold change for clobetasone butyrate, desonide, diflucortolone valerate, and hydrocortisone butyrate, respectively, compared to the control.

Chromatographic retention assisted in-silico prediction of anticancer potential of glucocorticoids on cancer cell lines

Glucocorticoids (GCs) are hallmarks of anti-inflammatory activity. They are used as adjuvant therapy in oncology medications to alleviate some of the associated side effects. Although recent research has indicated that GCs have favorable anticancer potential, some scientific evidence suggests a pro-proliferation impact of GCs on cancer cells. This may create a scientific confusion on the utility of GCs and the choice of which GC enhances the anticancer potential and mitigates the negative effect. Accurate in-silico prediction and correct ranking of biological activity may be limited by the impact of the physicochemical interaction between small molecules and biological membranes. Chromatographic retention is inherently dependent on the physicochemical properties of the test molecule. It can scale the relative hydrophobicity and tentatively indicate the membrane permeability. In this study, the relationship between the in-silico binding affinity, the chromatographic retention and the in-vitro anticancer activity was investigated. Fifteen GCs were chromatographically separated on an Inertsil® C18 (4.6 * 250 mm; 5 μm) HPLC column. The binding affinity of the test GCs was determined on three receptors involved in cancer cell proliferation (topoisomerase II (TOPII), glucocorticoid receptor (GR) and ATP-binding cassette (ABCG2)). The antiproliferative potential of the test steroids was determined as per their IC₅₀. The correlation between chromatographic retention and the binding affinity to the observed IC₅₀ was investigated by multiple linear regression (MLR). Results revealed that some GCs exhibited a remarkably favorable inhibitory potential against cancer cell lines over normal cell lines. Our data indicated a significant correlation between the retention times of different GCs and the determined binding affinity, especially to the GR (r² = 0.677; p = 0.011) and the estrone sulphate (ESS) binding site of the ABCG2 (r² = 0.643; p = 0.018). Concurrently, the retention times were well correlated to the observed IC₅₀ on the colorectal cancer cell lines (r² = 0.580; p = 0.038) and the hypopharyngeal cell lines (r² = 0.638; p = 0.019). Significant MLR models (n = 4) correlating the retention times of the tested steroids and the binding affinity to the observed IC₅₀ were created. The MLR model correlating the retention times and the ABCG2_ESS binding affinity to the IC₅₀ on lung cancer was the most significant (p = 0.004). The accuracy of the model was 107.12 ± 29.18 % indicating good IC₅₀ prediction abilities. In conclusion, chromatographic retention can be employed as a low-cost and simple auxiliary tool for improving the in-silico prediction of the in-vitro activities of small molecules.

Abstract:

Frequent outbreaks of infectious diseases in aquaculture have led to significant economic losses. The leopard coral grouper (Plectropomus leopardus) often suffers from vibriosis. Improving host immunity presents a superior strategy for disease control, with minimal side effects compared to the use of antibiotics, highlighting the necessity of exploring the mechanisms underlying the fish's response to pathogen infections. Here, we conducted a comparative metabolomic analysis on the livers of the P. leopardus infected with Vibrio harveyi. A total of 1124 differential metabolites (DMs) were identified, with 190, 218, 359, and 353 DMs being identified at 6, 12, 24, and 48 h post‐infection (hpi), respectively. Then, based on the time series analysis, we found that the lipid metabolism pathways were modulated in response to the Vibrio infection, with an increase in the quantity of eicosanoids and gycerophospholipids (GPLs), as well as a decrease in the quantity of bile acids (BAs), vitamin D, and sex hormones. Furthermore, 13 enriched pathways involving 31 DMs were identified through KEGG (Kyoto Encyclopedia of Genes and Genomes) enrichment analyses. We identified histamine, 15(S)‐HpETE, and anandamide in the transient receptor potential (TRP) channels pathway, as well as (7S,8S)‐DiHODE, 5S,8R‐DiHODE, and 13(S)‐HpODE in the linoleic acid (LA) metabolism pathway. The DM levels increased, which may be attributed to inflammation. The DMs in the thyroid hormone synthesis pathway were identified, and the contents of nicotinamide adenine dinucleotide phosphate (NADPH) and glutathione (GSH) decreased, which may be crucial in antioxidants. Our findings highlighted the dynamic adjustments in lipid metabolism and the response to inflammation and oxidative stress during the infection of V. harveyi in P. leopardus. This study not only deepens our understanding of the metabolic underpinnings of fish immune responses but also lays the groundwork for research into functional metabolomics and mechanisms of disease resistance.

In Second Big Dermatology Deal this Week, Almirall Buys 5 Products from Allergan

Almirall, headquartered in Barcelona, Spain, announced it is buying five dermatology products from Allergan. Those include Aczone (dapsone), Tazorac (tazarotene), Azelex (azelaic acid), Cordran Tape (fludroxycortide) and Seysara (sarecycline). Seysara is a new product, a first-in-class tetracycline-derived antibiotic with anti-inflammatory properties to treat moderate to severe acne vulgaris in patients nine years of age and older. The U.S. Food and Drug Administration (FDA) is slotted to approve the drug in the fourth quarter of this year. The drug, if approved, is projected to have $200 million annual sales at its peak. “This is a transformational deal for Almirall,” said Peter Guenter, Almirall’s chief executive officer, in a statement. “It will reinforce and consolidate our position in the world’s largest dermatology market and is a well-balanced portfolio of mature and growth brands with a major launch opportunity of an innovative New Chemical Entity (NCE). It is perfectly complementary to our existing platform and will be immediately accretive to our earnings. It offers us medium to long term top and bottom line growth opportunities. Moreover, it will allow for an expanded platform to launch KX2-391, which has the potential to become a new standard of care in actinic keratosis.” In the first half of this year, the portfolio had net sales of $70 million. The acquisition was for $550 million upfront cash and up to $100 million based on performance. The company stated, “The acquired portfolio entails an excellent fit with Almirall U.S.’s selling capabilities and its current team, which has deep knowledge of the U.S. oral acne market and the acquired products. The four products already marketed generated total sales in U.S. of $70 million in the first six months of 2018. In addition, Seysara (sarecycline) offers strong medium to long term growth potential.” Almirall also indicates that the acquisition will make dermatology the company’s primary growth driver, increasing its net sales to nearly 45 percent of the group total from 34 percent. This deal marks the second big prescription dermatology deal this week. Earlier, Dutch company LEO Pharma announced it was acquiring Bayer ’s global prescription dermatology portfolio, including Skinoren for acne, Travogen and Travocort for fungal skin infections, Finacea for rosacea, and a number of topical steroids including Advantan, Nerisona and Desonate. It didn’t include Bayer’s over-the-counter products, including Canesten, an antifungal. PharmaPhorum notes that, “Bayer had attempted to sell its prescription dermatology portfolio in 2016, but struggled to find a buyer willing to meet the $1 billion asking price. No financial details of the new arrangement have been disclosed, but it is likely to be worth considerably less than two years ago, as since then some products have lost patent protection.” As part of the deal, LEO Pharma picks up the global product rights, except for Afghanistan and Pakistan, and will take over the sales and marketing teams in 14 countries, in addition to Bayer’s factory in Segrate, Italy.
