Baicalein attenuates ovalbumin-induced allergic rhinitis through the activation of nuclear receptor subfamily 4 group a member 1

Baicalein, one of the major active flavonoids found in Scutellaria baicalensis, has been revealed to exhibit potent anti-inflammatory properties in allergic airway inflammation. This study aimed to explore the role of baicalein and its relevant mechanism in the treatment of allergic rhinitis (AR). The bioinformatics tools were used to predict the targets of baicalein and AR-related genes. AR mice were induced by ovalbumin (OVA) and treated with lentivirus-encapsulated knockdown of nuclear receptor subfamily 4 group A member 1 (NR4A1) or protein arginine N-methyltransferase 1 (PRMT1) plasmids and baicalein. IL-4/IL-13-induced human nasal mucosal epithelial cells (HNEpC) were transfected with knockdown of NR4A1 or PRMT1 plasmids and baicalein treatment. Baicalein alleviated AR-like symptoms and reduced the levels of immunoglobulin E, histamine, and LTC4 in serum and IL-4, IL-25, and IL-33 concentrations in nasal lavage fluids of mice induced with OVA by increasing NR4A1 expression. NR4A1 blocked the NFκB/p65 pathway by mediating transcriptional repression of PRMT1. Knockdown of PRMT1 overturned the effects of NR4A1 knockdown on IL-4/IL-13-induced HNEpC and OVA-induced mice. Collectively, these findings provide evidence that baicalein activation of NR4A1 mediates transcriptional repression of PRMT1 and relieves AR in mice by blocking the NFκB/p65 pathway.

An ultrasensitive electrochemical sensor for the simultaneous detection of baicalin and baicalein in pharmaceuticals and serum samples

Developing an accurate and ultrasensitive method for the simultaneous detection of baicalin and baicalein, two key flavonoid from Scutellaria baicalensis, is critical, particularly in the context of traditional Chinese medicine. This study focuses on an innovative electrochemical approach, utilizing a biochar (BC)/ZIF-67/MnCo₂O₄ composite, which was fabricated via a combination of ultrasonic, calcination, and hydrothermal treatments. The resulting BC/ZIF-67/MnCo₂O₄ electrode exhibits exceptional sensitivity, with linear response ranges of 0.01 μM-400 μM. The limits of detection (LOD) for baicalein and baicalin were as low as 0.056 nM and 0.49 nM, respectively. The practical evaluations in serum and Scutellaria baicalensis demonstrate high recovery rates of 93.65 %-95.25 % for baicalein and 98.23 %-101.52 % for baicalin, with relative standard deviations below 3.99 % and 2.92 %, respectively. This study not only presents a promising technique for quality control in traditional Chinese medicine but also expands the potential applications of BC/ZIF-67/MnCo₂O₄ composites in electrochemical sensing, paving the way for applications in food safety monitoring and herbal medicine standardization.

Facile synthesis of nitrogen self-doped carbon dots from rapeseed meal for highly sensitive fluorescence detection of baicalein

The rapeseed meal, a type of residual by-product of rapeseed oil production was used as the precursor to prepare nitrogen self-doping carbon dots RM-CDs through an easy hydrothermal process. Thanks to the introduction of nitrogen element and oxygen-containing functional groups, RM-CDs had a fluorescence quantum yield of 18.6 %. The RM-CDs demonstrated highly stable to the ionic strength, pH, and ultraviolet radiation. However, their bright blue fluorescence was significantly weakened by baicalein owing to the inner-filter effect and static quenching effect. As a result, RM-CDs could be used as the fluorescent sensing materials for baicalein detection with a wide range of linear response (0.5-170 μM) as well as low limit of detection (0.046 μM). Moreover, the as-developed sensing platform was also applicable to detect baicalein in actual samples. The developed RM-CDs was sustainably prepared from the biomass by-product of rapeseed meal, this approach expanded the application scopes of CDs.
