Benzodiazepines (BZDs) have been reported to cause negative impacts on motor as well as cognitive functions, which in turn could result in lethal incidents including falls especially in the elderly. This notwithstanding, few trials have evaluated a feasibility and benefits of discontinuing BZD-derivative hypnotics in a systematic manner in this frail population. In this 8-week open-label study, we examined changes in motor and cognitive functions following the discontinuation of BZD hypnotics in older persons.
OBJECTIVES & HYPOTHESES
Primary Objective The primary objective is to examine the feasibility of discontinuing BZD-derivative hypnotics in older people.
Secondary Objectives
One of the secondary objectives is to examine the magnitude of discontinuing BZD-derivative hypnotics in the stability of body.
Another secondary objective is to examine the magnitude of discontinuing BZD-derivative hypnotics in cognitive function.
Hypotheses
1. More than 80% of the participants will complete and tolerate all the study procedures.
2a. Participants will show an improvement in the stability of body. 2b. Participants will show an improvement in the cognitive function globally as well as specifically in attention.

Determination of Sedative and Anesthetic Drug Residues in Aquatic Food Products Using Solid Phase Extraction (SPE) and Liquid Chromatography–Tandem Mass Spectrometry (LC–MS/MS)

The residues of sedative and anesthetic drugs in aquatic products are an important factor affecting food safety.In order to screen the residues of sedative and anesthesia drugs in aquatic products, a rapid and sensitive method was established using solid phase extraction (SPE) and high-performance liquid chromatog.-tandem mass spectrometry (HPLC-MS/MS).The samples were extracted by 80% aqueous acetonitrile by an alumina column and separated by a C18 column.The mass spectra were determined in pos. ion mode.The standard curves were matrix matched and quantified by the external standard method.The results showed that the linear ranges of the drugs in aquatic products were from 0.5 to 100 μg kg-1.The correlation coefficient (R2) was greater than 0.990 and the detection limits were between 0.001 and 0.322 μg kg-1.The average recovery rate and relative standard deviation were between 78.64% and 112.15% and from 0.41%-8.26%.Overall, the developed method showed promising results for the simultaneous qual. and quant. determination of the drug residues in aquatic products.

Eu-MOF-based highly sensitive and selective luminescence probe for trace, in situ and visual detection of flunitrazepam

Flunitrazepam, as an emerging new psychoactive substance classified as a third-generation drug that is more harmful and camouflaged, is gradually proliferating globally. Maliciously used as a criminal tool in homicide and rape cases, it has already caused serious harm to public safety and social stability. Owing to its special molecular structure, low concentration level and rapid metabolic process in the human body, accurate detection of flunitrazepam remains a major challenge, especially for real sample and on-site detection. In this paper, a lanthanide MOF (Eu-MOF) based on bi-ligand was constructed as a luminescence probe and used for the first time to detect trace amounts of flunitrazepam. The 'antenna effect' promotes strong luminescence of Eu-MOF, while the lower LUMO orbital energy level of flunitrazepam allows it to accept electrons from the electron donor leading to quenching of Eu-MOF luminescence. The probe has a high sensitivity and can detect flunitrazepam in the range of 0-800 μM with a detection limit as low as 73 nM. Moreover, flunitrazepam was detected in urine from real samples as well as in a variety of beverages to further validate its accuracy and practicality. The reported Eu-MOF represents one of the pioneering luminescence probes for the detection of flunitrazepam, which offers great promise for the on-site or on-line analysis of flunitrazepam.

Surface-grafted MoS₂ for ketoconazole sensing in biological and aqua samples

Fungal infections necessitate crucial antifungal agents, yet despite their effectiveness, these drugs can induce severe reactions, interact with various medications, and have detrimental environmental effects.Consequently, there is a pressing need for accurate, sensitive, and reliable detection methods to mitigate and ideally prevent these potential neg. impacts.Due to that, the present work is devoted to a biocompatible β-cyclodextrin (β-CD) functionalized molybdenum disulfide (MoS₂) nanocomposite using a liquid exfoliated hydrothermal method. β-CD/MoS₂ was assessed for its electrochem. detection capabilities of ketoconazole (KTZ) by modifying the glassy carbon electrode (GCE).The synthesized β-CD/MoS₂ undergoes thorough characterization using various anal. and spectroscopic techniques, such as X-ray crystallog. (XRD), XPS, Field emission SEM (FE-SEM), and Transmission electron microscopy (TEM) anal.Furthermore, the β-CD/MoS₂ sensor is tested under diverse conditions, including scan rate and pH variations, successfully detecting KTZ at concentrations as low as 1.0 nM.It demonstrates an impressive sensitivity of 1.3567μA μM-1 cm-2 and a limit of quantification (LOQ) matching at 0.4 nM, surpassing other electrodes in sensing performance.The proposed sensor was successfully applied to determine KTZ in pharmaceutical formulations and biol. samples with good recoveries.In addition, the sensor displayed the high selective detection of KTZ in the existence of other co-interferences, good repeatability, reproducibility, and high storage stability.As a result, this method proves to be practical and devoid of any matrix effect on the KTZ sensor.
