The goal of this study is to see how an ingredient called kynurenic acid (which we named "FS2") affects scar formation in people with burn injuries that need skin graft surgery. A cream with FS2 will be used on both the area where the skin graft was placed and the area where the skin was taken (donor site). The cream will be applied after the skin has healed. This study will help us understand if FS2 is safe and effective for mitigating skin scar formation in burn patients.

Revealing the key antioxidant compounds and potential action mechanisms of Bauhinina championii honey based on non-targeted metabolomics, mineralogical analysis and physicochemical characterization

Bauhinia championii (BC) honey, highly regarded for its functional properties, is popular among consumers and holds significant economic potential, particularly in the food and health industries. However, limited knowledge of the bioactivities, especially its outstanding antioxidant activity, hampers the development of functional products. This study comprehensively compared BC honey's physicochemical properties, elemental composition, metabolic profile, and antioxidant properties with five other honeys. Results revealed that BC honey had a darker color, higher electrical conductivity (335.67 ± 6.81 μS cm^-1), and higher levels of minerals and flavonoids. It exhibited stronger radical scavenging activity, with DPPH (96.49 % ± 0.38) and ABTS (2.1593 mM ± 0.014) values. Multivariate analysis suggested that its superior antioxidant properties are likely due to high flavonoid content, particularly 7-O-methylchrysin. This study offers insights into BC honey's antioxidant characteristics and supports its use in functional products.

Early life imidacloprid and copper exposure affects the gut microbiome, metabolism, and learning ability of honey bees (Apis mellifera)

The pesticide imidacloprid and the heavy metal copper provide some degree of protection to plants, while at the same time causing varying degrees of damage to bees. However, few studies have investigated the negative effects of imidacloprid and copper exposure on newly emerged bees (young bees), especially when both are present in a mix. In this study, young bees were exposed to sterile sucrose solutions containing imidacloprid (10 μg/L, 100 μg/L), copper (10 mg/L, 50 mg/L), or a mix of both (10 μg/L + 10 mg/L) for 5 days to assess their gut system and behavior, with survival and dietary consumption recorded over 21 days. We found that imidacloprid and copper reduced honeybee survival, dietary intake, and learning ability, decreased gut microbiota diversity, and caused metabolic disruptions. Notably, the mix of imidacloprid and copper had a synergistic negative effect. Correlation analyses revealed that the honeybee gut microbiota influences bee immunity and behavior by regulating metabolic pathways related to ascorbate, tryptophan, and carbohydrates. Our results demonstrate that imidacloprid and copper, either alone or in a mix, alter young bee health through a complex mechanism of toxicity. These findings highlight imidacloprid and copper's negative effects on young honeybees, offering insights for future pesticide and heavy metal impact research.

Serum metabolomics after exercise in ambulatory individuals with cerebral palsy

AbstractAimTo evaluate whether serum metabolomics differ between ambulatory individuals with cerebral palsy (CP) compared with individuals with typical development and whether functional capacity is associated with metabolite abundance.MethodThirty‐eight adolescents and young adults were enrolled (CP: n = 19; typical development: n = 19). After functional capacity testing (10‐meter walk, sit‐to‐stand, and peak knee flexion/extension torques), blood was drawn. Targeted serum metabolomics on hydrophilic metabolites were performed by high‐performance liquid chromatography coupled with high‐resolution and tandem mass spectrometry. Metabolite dimensionality reduction, pathway analysis, fold change, and t‐tests evaluated changes in metabolite abundance. Associations were tested between functional measures and metabolite abundance.ResultsIndividuals with CP had a significant increase in the abundance of essential amino acids, catabolic products of protein metabolism, and tricarboxylic acid cycle substrates, such as valine, tryptophan, kynurenic acid, and pyruvate (p < 0.05). Importantly, the abundance of numerous metabolites was only highly associated with functional capacity in individuals with CP such that greater abundance was associated with greater capacity, but not in those with typical development.InterpretationOur findings show clear increases in serum metabolites in individuals with CP, which are associated with functional capacity for movement. The altered metabolite profile measured after exercise might reflect increased energy production needed for movement. Appropriate nutritional intake during exercise might be needed given increased energy requirements.

Kynexis Announces Positive Topline Results from Phase 1 Study of KYN-5356, a Potential Treatment for Cognitive Impairment Associated with Schizophrenia

KYN-5356, an innovative, first-in-class small molecule inhibitor of KAT-II, a key enzyme in the kynurenine pathway implicated in schizophrenia, was shown to be safe and well-tolerated in the phase 1 study in healthy volunteers Initial data show clear evidence of target engagement and suggestive evidence of cognitive improvement, adding to body of evidence that supports further study of KYN-5356 in patients with cognitive impairment associated with schizophrenia (CIAS), which is among the most debilitating symptoms of the disease Clinical trial in patients anticipated to begin in 2025 Dec. 17, 2024 -- Kynexis, a biotechnology company focused on precision therapeutics for brain diseases, today announced positive topline results from its first-in-human phase 1 clinical trial. The phase 1 study in healthy volunteers evaluated the safety, tolerability, and pharmacokinetics of single and multiple ascending oral doses of KYN-5356, a first-in-class small molecule inhibitor of KAT-II, being developed for the treatment of cognitive impairment associated with schizophrenia (CIAS). The study also evaluated central pharmacodynamic biomarkers and cognitive performance to determine target engagement in the multiple ascending part. Results show that KYN-5356 was safe and well-tolerated and exhibited excellent pharmacokinetic (PK) properties in both plasma and cerebrospinal fluid (CSF), suggesting appropriate penetration and exposure of KYN-5356 in the central nervous system (CNS). The study also tested exploratory pharmacodynamic (PD) endpoints, where KYN-5356 demonstrated statistically significant improvements in electroencephalography (EEG) activity relevant to cognitive pathways, as well as early, suggestive evidence of cognitive improvement. Findings further showed a dose-dependent reduction of kynurenic acid levels in the CSF, which have been shown to be elevated in people with schizophrenia and are believed to be a key driver of CIAS. “Cognitive impairment is a primary driver of disability for people with schizophrenia and affects most people with the disease. While still early, these data represent a milestone for Kynexis as we advance this potentially first-in-class treatment for cognitive impairment associated with schizophrenia, or CIAS,” said Kees Been, Chief Executive Officer of Kynexis. “As the first KAT-II inhibitor studied in humans, KYN-5356 has demonstrated an excellent safety profile and promising pharmacokinetic data, including evidence of significant brain penetration.
