The method for determination of methanesulfonic acid tropisetron by HPLC was established.The compound was separated by column C18 with methanol-water-acetonitrile-triethylamine (565: 40: 35: 0.3, V/V/V/V) as mobile phase A and methanol-water-acetronitrile-triethylamine (800: 100: 100: 0.3, V/V/V/V) as mobile phase B for gradient elute, and the detection wavelength was 284nm.The detection limit of methanesulfonic acid tropistron was 0.24ng, and the principal component can separated from the other relevant compoundsThe method is simple, accurate with good specificity, and can be applied to determining the relevant compounds of methanesulfonic acid tropistron.

Influence of oxycodone hydrochloride injection and fentanyl on Stretch reaction in appendectomy under epidural block

The objective was to observe the clin. effect of oxycodone hydrochloride injection and fentanyl on Stretch reaction in appendectomy under epidural block and the security.The method includes selecting 60 patients who will receive appendectomy, randomly dividing them into observation group and control group with 30 patients into each group, performing appendectomy after epidural block effect is definite, i.v. injection of oxycodone hydrochloride 0.1 mg/kg+Tropisetron Mesylate 6 mg and then pump injection of oxycodone 2μg/(kg·min) until the abdomen was closed, i.v. injection of fentanyl 1μg/kg+Tropisetron Mesylate 6 mg and then pump injection oxycodone 2μg/(kg·min) until the abdomen was closed, monitoring SBP, DBP, HR, ECG and SpO₂ change before the appendectomy, at skin incision, incision of abdominal cavity, Stretch appendix and the end of appendectomy, and assessment of the internal organs stretch reaction state.The results show that oxycodone hydrochloride injection has good therapeutic effect, little untoward effect and big assurance coefficient for preventing Stretch reaction in appendectomy under epidural block.

HPLC determination of content and dissolution of tropisetron mesylate tablets

A method for content determination and dissolution assay of tropisetron mesylate in tablets was established by HPLC on Zorbax SB - C₁₈ column (4.6 mm x 150 mm, 5 μm) with 0.02 mol L^-1 potassium dihydrogen phosphate solution (adjusted to pH = 5.5 with potassium hydroxide) - methanol - acetonitrile (35:40:25) as mobile phase and at a flow rate of 0.8 mL min^-1 and UV detection at 285 nm.There was a good linear between 0.1 - 1.0 μg of tropisetron mesylate and 2 μg internal standard (r = 0.999 9, n = 6).The recovery of the method was 100.1%, the intra-day RSD and inter-day RSD was 0.52% and 0.69%, resp.The method was specific, accurate and convenient, and could be used for quality control of the tropisetron mesylate tablets.
