Study on the Effect of an Echinacea Formulation on the Clinical Manifestations and Evolution of Covid-19

The main objectives of ECCO-2 are: 1) Efficacy: to study whether EQUINACEA ARKOPHARMA, hard caplets containing cryogenized root of the plant Echinacea purpurea, show an improvement of the clinical manifestations and disease course in ambulatory patients with covid-19 with a respiratory presentation and not requiring hospitalization (i.e., mild covid-19). The drug being evaluated will be added as a supplement of the standard treatment, with its current recommended dose for treatment of the common cold. 2) Safety: to determine that the incidence of adverse events is not higher than that seen with the standard treatment applied in each case.

Open, Controlled Prospective, Explorative Clinical Trial to Assess Pharmacology and Effectiveness of Echinaforce Chewable Tablets "EC" to Prevent From Enveloped Virus Infections

It is a single-center, randomized, open-label, phase IV study for the explorative investigation of the pharmacological mode-of-action of Echinaforce® extract in the form of Echinaforce® Chewable tablets"- Further, it is aimed to evaluate the effectiveness of the study product for the prevention and treatment of respiratory tract infections in generally healthy adults following a real-life setting. The study covers 2x2 + 1 month of prevention in 120 randomized participants following a tight sampling set-up for sensitive detection of viral infections by RT-qPCR analysis of nasal swabs and seroconversion of SARS-CoV2 IgG/IgM in serum samples.

Developing a functional non-animal CNS stress model utilizing long-term potentiation with human iPSC-cortical neurons to evaluate therapeutics

Cortisol, the main stress hormone of the hypothalamic-pituitary-adrenal (HPA) axis, has been hypothesized to cause considerable detriment to cognitive function in both a time and concentration-dependent manner. However, there is a current lack of functional in vitro models available to evaluate the stress condition. Long-term potentiation (LTP) has served as a quantitative correlate for memory and learning through in vitro neuronal network responses to electrical stimuli, where a high-frequency stimulation (HFS) protocol on microelectrode arrays (MEAs) evaluates synaptic integrity related to higher-order cognition. As a novel alternative to animal studies, this study has employed a human iPSC-cortical neuron organ-on-a-chip (HoaC) system to establish a stress phenotype for synaptic dysfunction and evaluate the effects of therapeutic compounds to ameliorate stress-induced cognitive dysfunction. In our HoaC system, cortisol exposure was found to alter cortical neuron LTP, synaptic integrity, cellular morphology, and electrophysiology, confirming a cortisol-induced stress phenotype consistent with previous findings. Using this novel system, we investigated the ability of Echinacea purpurea and its active ingredient, dodeca-2E,4E,8Z,10Z-tetraenoic acid N-isobutyl amide (dodeca), to mitigate stress-induced functional decline. Following exposure to chronic stress (1 µM cortisol dosing for 7 days), both Echinacea purpurea and dodeca were found to significantly alleviate cortisol-induced cortical neuron stress in a time-dependent rescue of LTP. Together, these results characterize a novel, biologically-relevant model of neurological stress, and highlight its utility in identifying new therapeutic compounds capable of restoring stress-induced cortical neuron network deficits.

Role of Echinacea in the management and prevention of acute respiratory tract infections in children: A systematic review of the evidence

Abstract::

Acute cough and respiratory tract infections (RTIs), common in children, often lead to high antibiotic use. Echinacea, an herbal alternative, has been studied for its immune-modulating and antiviral effects to reduce the unnecessary use of antibiotics. This systematic review analyzed randomized controlled trials on Echinacea for pediatric acute cough and RTIs. PubMed, Cochrane Library, Semantic Scholar, and Google Scholar were searched on October 15, 2024. Five studies met all inclusion criteria and were retained for qualitative synthesis. The studies generally showed some concerns of bias in randomization and allocation concealment, as well as some issues about blinding and selective reporting. Grading of recommendations assessment, development, and evaluation assessment indicated high certainty of the reduction of symptom duration and antibiotic use, and moderate certainty for the prevention of infection. Evidence suggests that Echinacea purpurea, especially at higher doses (e.g., 2000 mg/day), may reduce RTI incidence, viral load, and symptom severity. Several trials have reported fewer antibiotic prescriptions and shorter fever duration, indicating public health benefits. Efficacy varied by formulation, extraction, and dosing, with tablet-based and hydroethanolic extracts showing more consistent results. Echinacea was well tolerated, only producing mild adverse effects of gastrointestinal discomfort and skin rashes. Some studies showed no significant difference from placebo, which underlines the need for standardized dosing and formulation. Overall, Echinacea may be a useful adjunct for pediatric RTI prevention and management, but further large, high-quality trials are needed to confirm optimal use, efficacy, and safety.

Critical analysis of Echinacea preparations marketed in Germany

Abstract:

Colds are the most common cause of incapacity to work in 2023 and therefore have a significant impact on the German economy. Echinacea preparations are considered a non-specific immune stimulant that is used as a phytotherapeutic agent for the treatment and prevention of colds. In the 2021 ZEIT ranking of the best-selling pharmacy-only medicines in Germany, four Echinacea preparations were among the top ten, and they experienced a new hype during the SARS-CoV-2 pandemic. In this study, 22 Echinacea monopreparations, which are approved as herbal medicinal products, were analyzed based on their package inserts. The preparations used in clinical studies were compared with the preparations available on the market with regard to the criteria of plant species used, plant part used, preparation (extract or pressed juice), drug-extract ratio, single dose administered, standardization of the preparations, prior performance of an ingredient analysis, galenics, dosage, duration of administration, and daily dose administered. The analysis revealed that the preparations available on the market are very heterogeneous. The composition can vary considerably in terms of ingredients. For customers, these products appear homogeneous in terms of their presentation on the market. The legal framework conditions promote this situation. The clinical studies are also unable to adequately reflect the market situation. New studies based on market reality, with an analysis of ingredients in the preparations used, a standardized study design, and the inclusion of hematological and biochemical parameters in the evaluation should improve this situation. Days of incapacity for work as an outcome parameter could be well suited.

New laboratory (in-vitro) study provides coronavirus insight: A.Vogel's Echinaforce demonstrates anti-viral activity against SARS CoV-2, the virus that causes Covid-19.

MONTREAL, Jan. 26, 2021 /CNW/ - An article titled "In vitro virucidal activity of Echinaforce®, an Echinacea purpurea preparation, against coronaviruses, including common cold coronavirus 229E and SARS-CoV-2" was recently published in the renowned Virology Journal. The study was carried out by researchers of the Spiez laboratory in Switzerland. According to the laboratory (in-vitro) study, A.Vogel's Echinaforce was shown to be effective against coronaviruses in cell experiments, including the SARS-CoV-2 which is commonly known as COVID-19.
