Development of novel electrospun nanofibers loaded with Levosulpiride and Cannabidiol encapsulated in cyclodextrin-phospholipid inclusion complex for ameliorated treatment of anxiety and depression

Anxiety and depression are major global health challenges in the modern world, with current therapies often hindered by poor bioavailability and limited therapeutic efficacy. Advancing drug delivery systems to overcome these limitations is critical for achieving improved outcomes. This study presents a novel formulation of Levosulpiride and Cannabidiol (Levo-CBD) loaded in a Cyclodextrin/Phospholipid supramolecular inclusion complex (Levo-CBD-C-P-IC), embedded into electrospun nanofibers for targeted brain delivery. The Levo-CBD-C-P-IC was successfully prepared, exhibiting 222.8 nm of mean particle size along with 0.197 and -17.58 mV of polydispersity index and zeta potential, respectively. Morphological analysis confirmed the successful incorporation of the complex into nanofibers, and the average size was found in the range of 177 nm to 350 nm, as observed under a scanning electron microscope. Gamma scintigraphic studies demonstrated enhanced drug delivery to the brain with the nanofiber formulation compared to conventional approaches. Behavioural studies revealed significant improvements in therapeutic outcomes across multiple parameters, indicating the superior efficacy of the developed formulation. Histopathological evaluation showed reduced tissue damage in treated animals, with inflammatory markers approaching normal levels, reflecting effective management of neuroinflammation. The biochemical profile of the treated group further corroborated the formulation's improved therapeutic potential. In conclusion, the Levo-CBD-C-P-IC-loaded nanofiber system offers a promising approach for the targeted management of depression and anxiety by addressing limitations in drug bioavailability and delivery.

A framework to assess pharmaceutical accumulation in crops: from wastewater irrigation to consumption

The reuse of treated wastewater for irrigation can inadvertently introduce a suite of emerging contaminants such as pharmaceuticals into agri-ecosystems. However, current monitoring efforts to characterise exposure usually focus on a limited range of analytes. A modelling framework was developed that employs a sequence of pre-developed models to predict accumulative potential in a model crop, Zea mays (corn), using chemical structure and excretion rate as the only model inputs. Z. mays was selected as the model crop as it is a major food source, stands as one of the highest cultivated crops globally, and is characterised as having a medium uptake potential. The framework was used to predict uptake in Z. mays in three regions characteristic of high wastewater reuse (Australia, the US and the Middle East). Despite regional and plant specific differences, 72.7 % of the calculated concentrations were within a factor of ten of those reported in the literature. Topiramate, furosemide, and gemfibrozil were observed to accumulate to the greatest extent in Z. mays, predicted concentrations ranged between 50.27 and 418.01 ng/g (dw) for the top 10. Acids predominantly accumulated in leaves and fruit whereas a higher proportion of bases were predicted to accumulate in the roots. To the best of our knowledge 56.7 % of the 30 highest-ranked pharmaceuticals have not been previously documented in existing literature or monitoring campaigns. This presented framework demonstrates a method to assess risk posed by pharmaceutical compounds with limited experimental data.

Crystallization Resolution of a Racemic Compound (R,S)-Sulpiride Promoted by Chiral Ionic Liquids: Insight into the Effect on Selective Nucleation

This study systematically investigates the influence of chiral ionic liquids (CILs) on the enantiomeric resolution efficiency of chiral drugs via crystallizationFour CILs-1-pentyl-3-methylimidazolium-R-mandelate ([Pmim][R-MA]), 1-pentyl-3-methylimidazolium-R-pyroglutamate ([Pmim][R-PgA]), 1-pentyl-3-methylimidazolium-S-mandelate ([Pmim][S-MA]), and 1-pentyl-3-methylimidazolium-S-pyroglutamate ([Pmim][S-PgA])-previously synthesized and characterized in prior research were employed in a methanol-based system to evaluate their effects on the nucleation kinetics and crystallization resolution of the model drug RS-sulpiride (RS-Sul).Nucleation probability anal. revealed that R-enantiomeric CILs (R-CILs) preferentially facilitated the nucleation of S-sulpiride (S-Sul), exhibiting intermol. interactions with S-Sul that are significantly stronger than those with R-sulpiride (R-Sul).Furthermore, a CIL-based cosolvent mixture (CCM) with a mass fraction (ω) of 0.7 (CCM-0.7) was identified as a highly effective nucleation inhibitor, selectively suppressing the crystallization of enantiomers with identical chiral configurations while promoting the nucleation of their opposite enantiomers.In a methanol-based system containing 7 wt % R-CILs, variations in solvent composition and seeding concentrations significantly influenced the crystallization resolution efficiency of RS-Sul.The chiral recognition mechanism between CILs and Sul enantiomers was revealed by an independent gradient model based on Hirshfeld partition (IGMH) anal. as well as binding energy (BE) calculations, which further demonstrated that [Pmim][R-PgA] exhibited superior chiral recognition compared to [Pmim][R-MA], elucidating the mol.-level mechanism of CIL-mediated stereoselective nucleation.Addnl., cyclic stability assessments revealed that after six reuse cycles, the CILs retained a 93.8% recovery efficiency and structural integrity, highlighting their potential for practical applications in chiral resolution

Akums Launches Rabeprazole + Levosulpiride SR Capsules

Akums Drugs & Pharmaceuticals Ltd. announced that it has launched Rabeprazole + Levosulpiride SR Capsules. Approved by the Drug Controller General of India (DCGI), the company claims this new formulation aims to enhance relief for patients suffering from gastrointestinal tract (GIT) disorders. GERD, or gastro-oesophageal reflux disease, is a debilitating condition characterised by the reflux of stomach contents into the oesophagus, leading to troublesome symptoms and potential complications. With GERD often dubbed as chronic acid reflux, its prolonged presence not only impacts the quality of life but also poses risks of serious health complications, including stomach and esophageal cancer. “Rabeprazole sodium, a potent antisecretory compound, selectively inhibits gastric acid secretion by targeting the H+ and K+ ATPase at the surface of gastric parietal cells. It has demonstrated efficacy in treating gastric and duodenal ulcers, as well as GERD. Complementing rabeprazole, Levosulpiride, the levo-enantiomer of sulpiride, exhibits significant central antidopaminergic activity and antiemetic effects,” the company said in a statement. Its peripheral anti-dopaminergic action modulates the motor activity of the upper digestive tract, making it valuable in gastro-enterology, it claimed. “We are excited to introduce a solution for patients suffering from GERD and related gastrointestinal disorders. GERD diagnosis and treatment rely on symptom management and acid suppression therapies. Our Rabeprazole + Levosulpiride SR Capsules aim to offer an effective remedy, ease symptoms, enhance gastrointestinal function, and boost patients’ overall well-being. Levosulpiride offers rapid relief and better healing outcomes with fewer side effects, elevating the overall treatment experience,” Arushi Jain, Director, Akums Drugs & Pharmaceuticals Ltd. said in a statement. Each hard gelatin capsule contains Rabeprazole Sodium IP 40 mg (as enteric-coated form) and Levosulpiride 75 mg (as uncoated sustained-release form). The recommended dosage is one capsule once daily or as directed by a physician.
