Plasma levels of erythromycin from 3 treatments were compared in an open, single dose, 3-way cross-over study in 12 volunteers.The treatments were enteric-coated pellets of erythromycin base (500 mg) sprinkled on yogurt or jam, or erythromycin ethyl succinate suspension (1 g).Plasma samples were collected immediately prior to dosing and after 1-9 h.Samples following erythromycin ester dosage were assayed for total erythromycin and erythromycin ester, the concentration of erythromycin base then being calculated by difference.The erythromycin base formulation displayed very similar plasma concentration vs. time curves when administered on either yogurt or jam.However, total erythromycin levels from the ethyl succinate formulation were significantly lower than those produced by the base formulation and less than half of the erythromycin was present as the active base.There were no adverse experiences reported on any of the treatments.
