A New Experimental Method for Evaluating Drugs in the Nasal Cavity

A method was described that permitted evaluation of nasal decongestant activity in anesthetized rats and guinea pigs on spontaneous nasal resistance, histamine-induced congestion, and exptl. allergic rhinitis.Drugs to be tested were administered into the nasal cavities, the more closely simulating their clin. application.The decongestant activity of known standards could be detected at clin. effective concentrations, and the method could differentiate decongestant drugs with different durations of action.
Pharmacologic action and tissue distribution of epinephrine bitartrate in the rabbit eye

Topical administration of 0.5% 14C-labeled epinephrine bitartrate (I.bitartrate) [51-42-3] in 0.5% hydroxypropylmethylcellulose or saline as drops or in a freon propelled aerosol to unanesthetized rabbits decreased intraocular pressure.The most label was observed in eyes treated with I in hydroxypropylmethylcellulose and least in those treated by aerosol.The greatest localization of I occurred in the nictitating membrane which decreased with time.This may act as a reservoir and account for the redistribution of I causing a second peak in most component parts of the eye.Radioactivity was detected in the blood and also in the untreated contralateral eye throughout the experimentOnly 13% of the calculated dose of I was delivered to the eye by the aerosol adaptor, suggesting that less drug may be needed than is used.Aerosolized drugs may prove to be an effective dosage form for delivery of ophthalmic drugs.
