The method has been developed for the simultaneous determination of five essences in milk powder by high-performance liquid chromatog. with the diode array detector (UPLC-DAD).Firstly, the sample was extracted by ultrasound, and then purified by HLB solid phase extraction column.Agilent SB-Aq column (C18, 150mmx2.1mm, particle size 1.8μm), the mobile phase:methanol + 0.01 mM sodium dihydrogenphosphate (PH=2.9 adjusted by phosphoric acid), the flow rate:1.0mL♂min∼(-1) and the detection wavelength:278 nm were used.The linear range of five essences is from 0.2μg♂mL∼(-1)to 10μg♂mL∼(-1).The detection limit was 2.5mg♂kg∼(-1).The quant. limit was 7.5mg♂kg∼(-1).The average recovery was 83.1%∼ 88.1% and the precision was 2.8%∼7.3%.The method is easy to operate with high recovery rate and can be used to detect five essences simultaneously in milk powder.
