READS (restriction endonucleolytic anal. of differentially expressed sequences) gel display process involves selective PCR amplification of the 3'-end restriction fragments of the cDNAs based on the N1N2 nucleotides 5'-to the poly-(A) tail of the message.As a result, this approach produces well-defined products from each polyadenylated transcript and, in general, produces only one major product per poly-(A) site on the transcript.The approach has a wide dynamic range and predicts changes in the relative levels of different RNA species.READS can be carried out even with limited amounts of total RNA and hence can be applied to small tissue biopsies.READS is a very reproducible, consistent approach to study differential gene expression and is very easy to carry out.A detailed protocol of READS is provided.
