The emergence of a huge number of combinatorial libraries has created an immediate need for the development of automated and universal techniques for high throughput (HT) anal. and purificationThe ability to efficiently analyze and purify the libraries is essential to reduce the costs of drug development.Our approach of using online coupling of HPLC with solid phase extraction (SPE) under high pressure can be used to fully automate the semi-prep purification process.The technique can remove DMSO from samples on line before injection onto the HPLC column.In the post-column stage, purified enriched organic compounds can be easily accessed buffer-free in any organic solvent in micro-titer plate format for easy integration into the HT screening process.The talk will also describe, a parallel HPLC approach where it is possible to sep. simultaneously eight different samples using a single pump, multi-channel detector and expert software system resp.Samples are separated by an array of HPLC columns, providing one column for each sample.A sophisticated flow control ensures equal chromatog. conditions in each channel.The results combined with our existing SPE coupling method of purification for throughput increase will also be discussed.
