A Novel Effect of Id2 in Microglia TNFα Regulation

Microglia are the most important immune cells in the central nervous system (CNS), which can defend against external pathogens and stimuli. Dysregulation of microglia releases excessive proinflammatory cytokines and leads to neuroinflammation, which is fundamental to the pathophysiology of multiple neurological diseases. However, the molecular mechanisms underlying the regulation of proinflammatory cytokines in microglia are still not well-understood. Here, we identified that inhibitor of DNA binding protein 2 (Id2) was a negative regulator of tumor necrosis factor-α (TNFα) in cultured microglia. Knockdown of Id2 significantly increased the expression of TNFα in microglia, while overexpression of Id2 inhibited TNFα expression. Furthermore, by interacting with the p65 subunit of nuclear factor kappa-B (NF-κB), Id2 suppressed the transcription activation of NF-κB and inhibited TNFα expression. Interestingly, in lipopolysaccharides (LPS)-treated microglia, Id2 increased and underwent a cytoplasmic relocation. Immunoprecipitation and immunostaining results showed that by binding to the LIM domain of Id2, a scaffold protein PDZ and LIM 5 (PDLIM5) involved in the Id2 cytoplasmic relocation, which inactivated Id2 and resulted in higher TNFα expression in LPS-treated microglia. Collectively, our data delineate a novel effect of Id2 on TNFα regulation in microglia, which may shed a light on the proinflammatory cytokines regulating in microglia associated neuroimmune disorders.

Enhancing porcine oocyte quality and embryo development through natural antioxidants

During fetal development, primordial oocytes maintain their developmental potential through a ROS-minimizing metabolic mechanism. Maturation increases ROS levels, causing stress and damage, which are countered by in vivo antioxidants. In vitro maturation (IVM) worsens this due to fewer antioxidant presence and medium factors. To address this, we evaluated the effects of incorporating various natural antioxidants in the porcine oocyte IVM media. Our findings revealed that 10 μM Dendrobine (DEN), 1 μM Polydatin (PD), 20 μM Limonin (LIM), and 25 μM Nobiletin (NOB) significantly improved the first polar body extrusion rates (p < 0.05), reduced ROS, and increased GSH levels. Individual addition of 100 μg/mL Lycium barbarum polysaccharides (LBP), 0.1 μM Kaempferol (KAE), 250 μM Salidroside (SAL), 10 μM Curcumin (CUR), DEN, PD, LIM, and NOB to the porcine IVM system showed that KAE, LIM, NOB, and LBP treatments yielded the most favorable results. At the gene level, LIM, LBP, and NOB were found to upregulate the expression levels of GPX1, SIRT1, and TFAM, while downregulating Caspase3 and increasing the BCL2/BAX ratio. The inclusion of LIM, NOB, and LBP, either alone or in combination, into the IVM media effectively alleviated oxidative stress in porcine oocytes, decreased cell apoptosis, preserved mitochondrial membrane potential, and enhanced the blastocyst rate. These results offer valuable insights for optimizing the porcine oocyte IVM culture system.

HAGLR, A Long Non-coding RNA of Potential Tumor Suppressive Function in Clear Cell Renal Cell Carcinoma: Diagnostic and Prognostic Implications

Research works suggested the role of long non-coding RNAs (lncRNAs) in pathogenesis of clear cell renal cell carcinoma (ccRCC). lncRNA HAGLR is studied in several malignancies, but not in ccRCC. From The Cancer Genome Atlas Kidney Renal Clear Cell Carcinoma (TCGA-KIRC) dataset, we analyzed molecular alterations of HAGLR and constructed a competitive endogenous RNA (ceRNA) network with related miRNAs and mRNAs. Gene Ontology analysis was done to identify important pathways enriched with HAGLR recovered mRNAs. Clinical importance of HAGLR and related mRNAs was assessed and, the impact of selected mRNA-encoding genes on tumor immune infiltration was studied using TIMER. HAGLR expression was reduced in ccRCC than in normal kidneys, and correlated significantly with gene promoter methylation. Low HAGLR level in tumors showed diagnostic potency, and was associated with clinicopathological parameters (stage/grade/metastasis) and poor patient survival. The HAGLR-associated ceRNA network constituted 13 miRNAs and 23 mRNAs differentially expressed in the TCGA-KIRC dataset. From HAGLR recovered mRNA-encoding genes, we developed a 5-gene (PAQR5, ARHGAP24, HABP4, PDLIM5, and RPS6KA2) prognostic signature in the training dataset and validated it in testing as well as entire datasets. The expression level of signature genes showed negative correlation with tumor infiltration of immune cells having adverse impact on ccRCC prognosis and also with tumor derived chemokines facilitating the infiltration. In conclusion, HAGLR seemed to play a tumor suppressive role in ccRCC. HAGLR and associated gene signature may have implementation in improving existing prognostic measure and developing effective immunotherapeutic strategies for ccRCC.
