Introduction

Broad specificity cytosolic 5'-nucleotidase that catalyzes the dephosphorylation of 6-hydroxypurine nucleoside 5'-monophosphates (PubMed:10092873, PubMed:12907246, PubMed:1659319, PubMed:9371705). In addition, possesses a phosphotransferase activity by which it can transfer a phosphate from a donor nucleoside monophosphate to an acceptor nucleoside, preferably inosine, deoxyinosine and guanosine (PubMed:1659319, PubMed:9371705). Has the highest activities for IMP and GMP followed by dIMP, dGMP and XMP (PubMed:10092873, PubMed:12907246, PubMed:1659319, PubMed:9371705). Could also catalyze the transfer of phosphates from pyrimidine monophosphates but with lower efficiency (PubMed:1659319, PubMed:9371705). Through these activities regulates the purine nucleoside/nucleotide pools within the cell (PubMed:10092873, PubMed:12907246, PubMed:1659319, PubMed:9371705).

Risk-adapted treatment protocols conferred remarkable improvement in the survival rates of pediatric acute lymphoblastic leukemia/lymphoma (ALL/LBL). Nevertheless, clinical management is still challenging in certain molecular subgroups and in the presence of alterations associated with an increased rate of relapse. In this study, disease-relevant genomic and transcriptomic profiles were established in a prospective, multicenter, real-world cohort involving 192 children diagnosed with ALL/LBL. Gene fusions were detected in 34.9% of B-ALL and 46.4% of T-ALL patients, with novel chimeric genes involving JAK2, KMT2A, PAX5, RUNX1, and NOTCH1, and with KMT2A-rearranged patients displaying the worst 3-year event-free survival (P = .019). Nonsynonymous mutations were uncovered in 74.9% of the analyzed patients, and pairwise scrutiny of genetic lesions revealed recurrent clonal selection mechanisms commonly converging on the same pathway (eg, Ras, JAK/STAT, and Notch) in individual patients. Investigation of matched diagnostic and relapse samples unraveled complex subclonal variegation, and mutations affecting the NT5C2, TP53, CDKN2A, and PIK3R1 genes, emerging at the time of relapse. TP53 and CREBBP mutations, even as subclonal aberrations, were associated with shorter 3-year event-free survival among all patients with B-ALL (TP53 mutant vs wild-type: P = .008, CREBBP mutant vs wild-type: P = .010), and notably, B-ALL patients showing no measurable residual disease on day 33 could be further stratified based on TP53 mutational status (P < .001). Our in-depth molecular characterization performed across all risk groups identified novel opportunities for molecularly targeted therapy in 55.9% of high-risk and 31.6% of standard/intermediate-risk patients.

Specific glycomacropeptide detection via polyacrylamide gel electrophoresis with dual imaging and signal-fusion deep learning

Herein, we report a sodium dodecyl-sulfate polyacrylamide gel electrophoresis (SDS-PAGE) method featuring dual imaging and signal-fusion deep learning for specific identification and analysis of glycomacropeptide (GMP) in milk sample. Conventional SDS-PAGE methods lack specificity because of the signle staining of protein bands, and the overlap between GMP and β-lactoglobulin (βLg). Our dual imaging method generated a pair of complementary detection signals by recruiting intrinsic fluorescence imaging (IFI) and silver staining. Comparing the IFI image with the staining image highlighted the presence of GMP and differentiated it from βLg. Additionally, we trained a signal-fusion deep learning model to improve the quantitative performance of our method. The model fused the features extracted from the paired detection signals (IFI and staining) and accurately classified them into different mixing ratios (proportion of GMP-containing whey in the sample), indicating the potential for quantitative analysis on the mixing ratios of GMP added into whey sample. The developed method has the merits of specificity, sensitivity and simplilcity, and has potential to analysis of protein/peptides with unique IFI properties in food safety, basic research and biopharming etc.

Combined deletion of cytosolic 5′-nucleotidases IA and II lowers glycemia by improving skeletal muscle insulin action and lowering hepatic glucose production

Obesity and type 2 diabetes (T2D)-linked hyperglycemia, along with their associated complications, have reached pandemic proportions, constituting a major public health issue. Genetic deletion or pharmacological inhibition of purine nucleotide-metabolizing enzymes has emerged as a potential strategy for treating diseases. We previously showed that cytosolic 5'-nucleotidase II (NT5C2)-deficient mice were protected against high-fat diet (HFD)-induced insulin resistance. This study investigated the effects of dual deletion of cytosolic 5'-nucleotidases IA (NT5C1A) and II (NT5C2) in mice. We found that NT5C1A/NT5C2 double-knockout (NT5C-dKO) mice exhibited mild hypoglycemia, associated with enhanced skeletal muscle insulin action and reduced hepatic glucose production. This phenotype was accompanied by liver and skeletal muscle proteomic alterations notably related to amino acid metabolism, besides the potential involvement of adenosine monophosphate (AMP)-activated protein kinase (AMPK). Our findings support the development of novel anti-diabetic treatments using small-molecule cytosolic 5'-nucleotidase inhibitors.
