Introduction

Transcriptional modulator activated by BMP (bone morphogenetic proteins) type 1 receptor kinase. SMAD1 is a receptor-regulated SMAD (R-SMAD). SMAD1/OAZ1/PSMB4 complex mediates the degradation of the CREBBP/EP300 repressor SNIP1. May act synergistically with SMAD4 and YY1 in bone morphogenetic protein (BMP)-mediated cardiac-specific gene expression.

Bone-brain communication mediates the amelioration of Polgonatum cyrtonema Hua polysaccharide on fatigue in chronic sleep-deprived mice

This study aimed to investigate the anti-fatigue efficacy and underlying mechanisms of Polygonatum cyrtonema Hua polysaccharide (PCP) in chronic sleep-deprived mice. Following three weeks of oral administration, PCP demonstrated significant efficacy in alleviating fatigue symptoms. This was evidenced by the prolonged swimming and rotarod time in the high-dose group of PCP, which increased by 73 % and 64 %, respectively. Additionally, serum activities of CAT, GSH-Px, and SOD enzymes rose by 53.56 %, 37.69 % and 53.67 %, respectively, while MDA, lactic acid and BUN levels decreased by 22.90 %, 17.48 % and 24.61 %. The crosstalk between bone and brain is crucial for maintaining energy homeostasis. Molecular docking studies indicated a spontaneous and strong mutual binding between PCP and the bone-promoting target protein BMPR1A. Furthermore, it was observed that PCP enhanced osteogenic differentiation via the BMP-2/Smad1 pathway, leading to an upregulation of osteocalcin expression, which in turn regulated neurotransmitter balance and improved central arousal capacity. Moreover, PCP treatment stimulated neurogenesis by activating the CREB/BDNF/Akt signaling cascade, exhibiting neurotrophic effects. Additionally, PCP increased AMPK phosphorylation and destabilized TXNIP, facilitating astrocyte glucose uptake, glycolysis, and lactate conversion to support neuronal activity. These findings suggested that PCP could effectively respond to energy demands through bone-brain crosstalk, ultimately exerting anti-fatigue properties.

Genome‐Wide Association Study of Temporomandibular Disorder‐Related Pain in Finnish Populations

ABSTRACTBackgroundTemporomandibular disorders (TMD) are multifactorial musculoskeletal pain and dysfunctions in temporomandibular joints (TMJs) and masticatory muscles. Genetic factors play a role in TMD‐related pain, but only a few genome‐wide association studies (GWAS) have been conducted.ObjectiveThe aim of this GWAS was to explore genetic factors associated with painful TMD in Finnish populations.MethodsData from two epidemiological surveys, the Northern Finland Birth Cohort 1966 (NFBC1966) and the Health 2000 Survey in Finland, including altogether 468 cases and 6833 controls, were used. Case definition was based on pain on palpation of masticatory muscles and/or TMJs. GWASs of the whole data and stratified by sex were conducted from both cohorts using additive models, followed by meta‐analysis of the two cohorts. Replications of the previously reported TMD risk loci (rs73460075, DMD; rs4794106, SGCA; rs73271865, SP4; rs60249166, RXP2; rs1531554, BAHCCI; rs5862730, OTUD4/SMAD1; rs10092633, SFRP1; rs34612513, SOX14/CLDN18; rs878962, TSPAN9) were also investigated.ResultsFour genome‐wide significant loci were found in sex‐stratified analysis of NFBC1966, including associations at three loci in males (rs1023114, PRIM2, p = 5 × 10−9; rs4244867, ALG10, p = 3 × 10−8; rs79841648, ADCYAP1, p = 4 × 10−9) and one locus in females (rs148476652, DNER, p = 4 × 10−9). However, the results could not be replicated in the Health 2000 Survey or in the meta‐analysis of these two cohorts. The previous TMD GWAS associations did not replicate in our data either.ConclusionSeveral TMD pain‐associated variants were found in sex‐stratified analysis of NFBC1966, suggesting the role of neuroendocrine stress responses and central nervous system. These findings need to be confirmed in future studies.

Thyroid transcriptomic profiling reveals the differential regulation of lncRNA and mRNA related to prolificacy in Small Tail Han sheep with FecB ⁺⁺ genotype

The thyroid gland is an important endocrine gland in animals, which mainly secretes thyroid hormones and acts on various organs of the body. Long-chain non-coding RNA (lncRNA) plays an important role in animal reproduction. However, there is still a lack of understanding of their expression patterns and potential roles in the thyroid of Small Tail Han (STH) sheep. In this study, RNA-seq was used to examine the transcriptome expression patterns of lncRNAs and mRNAs in the follicular phase (ww_FT) and luteal phase (ww_LT) in FecB⁺⁺ genotype STH Sheep. A total of 17,217 lncRNAs and 39,112 mRNAs were identified including 96 differentially expressed lncRNAs (DELs) and 1054 differentially expressed mRNAs (DEGs). Functional analysis of genes with significant differences in expression level showed that these genes could be enriched in Ras signalling pathway, hedgehog (HH) signalling pathway, ATP-binding cassette (ABC) transporters and other signalling pathways related to animal reproduction. In addition, through correlation analysis for lncRNA-mRNA co-expression and network construction, we found that LNC_009115 and LNC_005796 trans target NIK-related kinase (NRK) and poly(A)-specific ribonuclease (PARN). LNC_007189 and LNC_002045 trans target progesterone-induced blocking factor 1 (PIBF1), LNC_009013 trans targets small mothers against decapentaplegic (SMAD1) are related to animal reproduction. These genes add new resources for elucidating the regulatory mechanisms of reproduction in sheep with different reproductive cycles of the FecB⁺⁺ genotype STH sheep.
