Epigenetic profiling of ADHD symptoms trajectories: a prospective, methylome-wide study

Attention-deficit/hyperactivity disorder (ADHD) is a prevalent developmental disorder, associated with a range of long-term impairments. Variation in DNA methylation, an epigenetic mechanism, is implicated in both neurobiological functioning and psychiatric health. However, the potential role of DNA methylation in ADHD symptoms is currently unclear. In this study, we examined data from the Avon Longitudinal Study of Parents and Children (ALSPAC)-specifically the subsample forming the Accessible Resource for Integrated Epigenomics Studies (ARIES)-that includes (1) peripheral measures of DNA methylation (Illumina 450k) at birth (n=817, 49% male) and age 7 (n=892, 50% male) and (2) trajectories of ADHD symptoms (7-15 years). We first employed a genome-wide analysis to test whether DNA methylation at birth associates with later ADHD trajectories; and then followed up at age 7 to investigate the stability of associations across early childhood. We found that DNA methylation at birth differentiated ADHD trajectories across multiple genomic locations, including probes annotated to SKI (involved in neural tube development), ZNF544 (previously implicated in ADHD), ST3GAL3 (linked to intellectual disability) and PEX2 (related to perixosomal processes). None of these probes maintained an association with ADHD trajectories at age 7. Findings lend novel insights into the epigenetic landscape of ADHD symptoms, highlighting the potential importance of DNA methylation variation in genes related to neurodevelopmental and peroxisomal processes that play a key role in the maturation and stability of cortical circuits.

[Comparative proteomics of the serum in patients with nasopharyngeal carcinoma: a study with two-dimensional electrophoresis and MALDI-TOF-MS].

OBJECTIVETo screen the tumor specific antigens of nasopharyngeal carcinoma (NPC) in the serum of the patients.METHODSTwo-dimensional electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) was employed to screen the specific biomarkers of NPC in the sera from 42 healthy volunteers, 37 NPC patients with lymph node metastasis and 27 NPC patients without lymphatic metastasis.RESULTSAfter pretreatment including albumin and immunoglobulin (IgG) depletion and desalting, the sera were subjected to 2-DE and image analysis. The differentially expressed protein spots were identified by MALDI-TOF-MS. Sera pretreatment resulted in better repeatability and resolution on the reference gel because of albumin and IgG depletion. From optimized 2-DE gel images, 29 spots indicating differential protein expression were identified by MALDI-TOF-MS to represent 23 proteins. Transferrin, ZNF544 protein, transthyretin, FAD-synthetase and NM23-H1 proteins were down-regulated and 12-lipoxygenase, serum amyloid A1 protein precursor, cytochrome P450, sICAM-1, cathepsin G and lysine-specific histone demethylase 1 were upregulated in the two groups of NPC patients as compared with the healthy group. Increased expressions of 12-lipoxygenase, sICAM-1, cathepsin G and lysine-specific histone demethylase 1 were detected in NPC patients with lymph node metastasis as compared with the patients without lymph node metastasis, but heat shock protein 70 was expressed only in NPC patients with lymph node metastasis.CONCLUSIONThe 2-DE-based serum proteome analysis can be useful in detecting the protein expression alterations due to carcinogenesis and development of NPC, and the newly discovered biomarkers might help in the diagnosis of NPC.

Identification of a novel circRNA–miRNA–mRNA regulatory axis in hepatocellular carcinoma based on bioinformatics analysis

AbstractIn recent years, circular RNAs (circRNAs) have been found to play an essential regulatory role in hepatocellular carcinoma (HCC) through various mechanisms, particularly the endogenous competitive RNA (ceRNA) mechanism. Therefore, it is significant to explore the circRNAs in hepatoma. In this study, we constructed the ceRNA and survival network using Cytoscape. We also used R, Perl software, and multiple online databases and platforms, including Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG), to perform overall survival, immune cell infiltration, immune checkpoints, pathway activity, and anticancer drug sensitivity analysis of the genes. Finally, the receiver operator characteristic curve (ROC) analysis was performed to identify the diagnosis value of the genes. KEGG analysis revealed the T cell receptor signaling pathway as the main enrichment pathway. A total of 29 genes related to survival and prognosis were screened out. The findings suggest that ZNF544, WDR76, ACTG1, RASSF3, E2F3, ASRGL1, and POGK are associated with multilevel immune cell infiltration. Additionally, immune checkpoint analysis screened out the ACTG1, E2F3, RASSF3, and WDR76. It was also revealed that the WDR76, E2F3, ASRGL1, and POGK mainly activated the cell cycle and DNA damage response (DDR) pathway. The results suggest that the sensitivity toward trametinib, refametinib (RDEA119), and selumetinib correlates to the expression of WDR76. ROC analysis showed that the area under the curve (AUC) of all genes in the regulatory axis was greater than 0.7. The identified hsa_circ_0000417/hsa_circ_0002688/hsa_circ_0001387--hsa-miR-199a-5p--WDR76 regulatory axis may provide new insights into the progression, clinical diagnosis, and treatment of HCC.
