Introduction

Does not have alcohol dehydrogenase activity. Binds NADP and acts through a one-electron transfer process. Orthoquinones, such as 1,2-naphthoquinone or 9,10-phenanthrenequinone, are the best substrates (in vitro). May act in the detoxification of xenobiotics. Interacts with (AU)-rich elements (ARE) in the 3'-UTR of target mRNA species. Enhances the stability of mRNA coding for BCL2. NADPH binding interferes with mRNA binding.

Identification of key chromium resistance genes in Cellulomonas using transcriptomics

Cellulomonas fimi Clb-11 can reduce high toxic Cr(VI) to less toxic Cr(III), and transcriptomics was used to reveal the key Cr(VI) uptake and reduction genes of C. fimi Clb-11 in this study. The results showed that under 0.5 mM Cr(VI) stress, 654 genes were upregulated. Among the upregulated genes, phosphate transport protein encoding genes phoU and TC.PIT, and molybdate transport protein encoding genes modA, modB, and modC were involved in the cell uptake of Cr(VI). Cytochrome c subunits encoding genes qcrA and qcrC were involved in the intracellular reduction of Cr(VI), and cytochrome c oxidase subunits encoding genes coxB and coxC were involved in extracellular electron secretion. Additionally, several unreported genes were found to be upregulated in C. fimi Clb-11 under Cr(VI) stress. The upregulated manganese transport protein encoding gene mntH may also assist Cr(VI) uptake in C. fimi Clb-11. Proton pump subunit encoding genes atpA, atpB, atpE, atpF, and atpH, as well as sodium-hydrogen antiporter subunit encoding genes mnhA and mnhC were upregulated, it may be involved in the extracellular proton secretion to reduce Cr(VI). Iron complex transport system substrate-binding protein encoding gene ABC.FEV.S, vacuolar iron transporter encoding gene VIT, FMN reductase gene encoding gene ssuE, and quinone oxidoreductase encoding genes qor and qorB were upregulated to reduce Cr(VI) in the intracellular. Our study showed theoretical importance by revealing the Cr(VI) reduction mechanism using chrome-resistant microorganisms.

An Expanded Substrate Spectrum of Sulfide:Quinone Oxidoreductase Found in Pollutant Degrading Bacteria

AbstractSulfide:Quinone Oxidoreductase (Sqr) Catalyzes The Initial Procedure On Sulfide Transformation, Alongside Sulfide (H2S, S2−) Oxidization Coupled With Coenzyme Q (CoQ) Reducing And Reactive Sulfur Species (RSS) Production. Here, We Assessed The Reactivity Of Propanethiol (PT) As An Alternative Substrate For Sqr To Maintain Intracellular Homeostasis In Strain S‐1 Capable Of Degrading Emerging Sulfur‐Containing Pollutants. We Deleted A Gene Encoding Sqr, And Serial Transcriptional Difference Induced By RSS Dynamics Was Therefore Revealed. Next, The Reaction Properties Of Two Sqr Homologs From Strains JMP134 And S‐1 Were Comparatively Characterized, Respectively. As A Result, An Additional Role Of Sqr In Yielding RSS From PT Was Found In Reaction Mixture Prepared By Cell‐Free Extracts Or Purified Enzymes. Interestingly, The Transformation Velocity Of PT By Sqr Was Slower Than That Of Sulfides. From This Scenario, It Was A Rate‐Determining Step That PT As A Nucleophilic Compound Can Be Added Into Sqr Cysteine To Form Disulfide Bond And Likely Serve Nonoptimal Sulfur Recipient. In Addition, The Role Of Persulfidation Driven By RSS In Combating Oxidative And Sulfur Stresses Required To Be Further Clarified. Nevertheless, This Promiscuity Of Sqr‐Binding Organosulfur Compounds And Its Catalytic Modulation Underscored That Expanded Substrates Might Benefit Sulfide Homeostasis In Thiol‐Degrading Bacteria.

Genetic resources and genes/QTLs for gram pod borer (Helicoverpa armigera Hübner) resistance in chickpea from the Western Himalayas

AbstractHelicoverpa armigera (also known as gram pod borer) is a serious threat to chickpea production in the world. A set of 173 chickpea genotypes were evaluated for H. armigera resistance, including mean larval population (MLP), percentage pod damage (PPD), and pest resistance (PR) for 2 consecutive years (year 2020 and 2021). The same core set was also genotyped with 50K Axiom CicerSNP Array. The trait data and 50,000 single nucleotide polymorphism genotypic data were used together to work out marker–trait associations (MTAs) using different genome‐wide association studies models. For MLP, a total of 53 MTAs were identified, including 25 MTAs in year 2020 and 28 MTAs in year 2021. A set of three MTAs was found common in both environments. For PPD, two MTAs in year 2020 and five MTAs in year 2021 were identified. A set of two MTAs were common in both environments. Similarly, for PR, only two MTAs common in both environments were identified. Interestingly, a common MTA (Affx_123255526) on chromosome 2 (Ca2) was found to be associated with all the three component traits (MLP, PPD, and PR) of pod borer resistance in chickpea. Further, we report key genes that encode SCAMPs (that facilitates the secretion of defense‐related molecules), quinone oxidoreductase (enables the production of reactive oxygen species that promotes diapause of gram pod borer), and NB‐LRR proteins that have been implicated in plant defense against H. armigera. The resistant chickpea genotypes, MTAs, and key genes reported in the present study may prove useful in the future for developing pod borer–resistant chickpea varieties.
