Light intensity-regulated glycogen synthesis and pollutant removal in microalgal-bacterial granular sludge for wastewater treatment

As light intensity plays a pivotal role in the microalgal-bacterial granular sludge (MBGS) process, understanding its impact on system performance and energy dynamics is essential. This study investigated the effects of varying light intensities (20, 100, 200, and 300 μ mol/m²/s) on the performance of MBGS in urban wastewater treatment, with a particular focus on glycogen accumulation and pollutant removal. The results demonstrated that light intensity significantly influenced microbial community structure, glycogen accumulation, and pollutant removal efficiency. At the lowest light intensity (20 μ mol/m²/s), filamentous Cyanobacteria dominated, leading to poor settling performance but enhancing phosphate removal due to increased polyphosphate metabolism. As light intensity increased, the microbial community shifted from Cyanobacteria to eukaryotic algae (e.g., Chlorella) at higher intensities. The optimal light condition (200 μ mol/m²/s) favored glycogen synthesis and improved synergistic interactions between microalgae and bacteria, ensuring stable pollutant removal. Notably, increased light intensity upregulated the expression of genes encoding glycogen branching enzyme (EC: 2.4.1.18), promoting glycogen accumulation in the MBGS system. These findings provide valuable insights for optimizing MBGS systems to achieve efficient urban wastewater treatment and resource recovery.

Identification of copper metabolism-related markers in Parkinson’s disease

OBJECTIVESThis study aimed to identify key genes related to copper metabolism in Parkinson's disease (PD), providing insight into their roles in disease progression.METHODSUsing bioinformatic analyses, the study identified hub genes related to copper metabolism in PD patients. Differentially expressed genes (DEGs) were identified using the limma package, and copper-metabolism-related genes (CMRGs) were sourced from the Genecard database. Immune cell-related genes were derived through immune infiltration and Weighted Gene Co-expression Network Analysis (WGCNA). Hub genes were pinpointed by integrating DEGs, CMRGs, and immune cell-related genes. Functional analyses included Receiver Operating Characteristic (ROC) analysis, Ingenuity Pathway Analysis (IPA), and networks for miRNA-mRNA-transcription factor (TF), Competitive Endogenous RNA (ceRNA), and hub gene-drug interactions. Validation was performed in cerebrospinal fluid (CSF) samples from PD patients, while in vitro experiments utilized GBE1- overexpressing SH-SY5Y cells to examine cell proliferation, migration, and viability.RESULTSNine hub genes (HPRT1, GLS, SNCA, MDH1, GBE1, DDC, STXBP1, ACHE, and AGTR1) were identified from 753 CMRGs, 416 DEGs, and 951 immune cell-related genes. ROC analysis showed high predictive accuracy for PD, and principal component analysis (PCA) effectively distinguished PD patients from controls. IPA identified 20 significant pathways, and various networks highlighted miRNA, TF, and drug interactions with the hub genes. Hub gene expression was validated in PD CSF samples. GBE1-overexpressing cells displayed enhanced proliferation, migration, and viability.CONCLUSIONSThe study identified nine copper metabolism-related genes as potential therapeutic targets in PD, highlighting their relevance in PD pathology and possible treatment pathways.

The impact of Cas9-mediated mutagenesis of genes encoding potato starch-branching enzymes on starch structural properties and in vitro digestibility

The digestibility of starch is affected by amylose content, and increasing amylopectin chain length which can be manipulated by alterations to genes encoding starch-branching enzymes (SBEs). We investigated the impact of Cas9-mediated mutagenesis of SBEs in potato on starch structural properties and digestibility. Four potato starches with edited SBE genes were tested. One lacked SBE1 and SBE2, two lacked SBE2 and had reduced SBE1, and one had reduced SBE2 only. Starch structure and thermal properties were characterised by DSC and XRD. The impact of different thermal treatments on digestibility was studied using an in vitro digestion protocol. All native potato starches were resistant to digestion, and all gelatinised starches were highly digestible. SBE modified starches had higher gelatinisation temperatures than wild type potatoes and retrograded more rapidly. Gelatinisation and 18 h of retrogradation, increased gelatinisation enthalpy, but this did not translate to differences in digestion. Following 7 days of retrogradation, starch from three modified SBE starch lines was less digestible than starch from wild-type potatoes, likely due to the recrystallisation of the long amylopectin chains. Our results indicate that reductions in SBE in potato may be beneficial to health by increasing the amount of fibre reaching the colon after retrogradation.
