Introduction

Myoblast-specific protein that mediates myoblast fusion, an essential step for the formation of multi-nucleated muscle fibers (PubMed:28681861). Actively participates in the membrane fusion reaction by mediating the mixing of cell membrane lipids (hemifusion) upstream of MYMX. Acts independently of MYMX (By similarity). Involved in skeletal muscle regeneration in response to injury by mediating the fusion of satellite cells, a population of muscle stem cells, with injured myofibers (By similarity). Also involved in skeletal muscle hypertrophy, probably by mediating the fusion of satellite cells with myofibers (By similarity).

Virtual screening and characterization of novel myogenic peptides from bovine collagen hydrolysates: Targeting myomaker

The increasing prevalence of muscle aging, exacerbated by an aging population, poses a significant threat to public health, necessitating the development of more effective interventions. This study primarily aimed to elucidate the mechanism by which bovine bone collagen facilitates muscle differentiation and regeneration. Initially, peptide sequences within bovine bone collagen hydrolysate were identified using peptidomics. Molecular docking and dynamics simulations subsequently demonstrated that the peptide AGPPGPPGPAGK could form a stable complex with Myomaker, suggesting its potential to regulate myoblast differentiation by targeting Myomaker. The physicochemical properties of AGPPGPPGPAGK were predicted using various deep learning tools, providing insight into its functional capabilities. Further molecular and cellular experiments confirmed that the peptide could enhance myoblast differentiation by regulating energy metabolism. Transcriptome analysis further supported these findings, revealing that the peptide modulated energy metabolism during myoblast differentiation. Finally, a combined bioinformatic and transcriptomic analysis indicated a potential regulatory role of Hrh1 in energy metabolism during cell differentiation, a finding that warrants further investigation.

Dysfunctional terminal differentiation of satellite cells in the wooden breast muscle of Ross 308 broiler chickens

Muscle satellite cells (MuSCs), defined as skeletal muscle-resident stem cells, serve as the principal cellular mediators of the repair and regeneration processes in impaired skeletal muscle tissue. However, myofibers with degeneration and necrosis in wooden breast (WB) myopathy are still not completely repaired, and this study aims to elucidate the functional changes of MuSCs between normal and WB-affected pectoralis major (PM) muscle of broilers. A total of 100 Ross 308 broilers (42 days old) fed the same diet were randomly selected. Initial identification of normal and WB phenotypes was conducted through behavioral assessments and manual palpation of the PM muscle. Ultimately, PM samples were harvested from 8 chickens per experimental group, classified into normal (NOR), moderate (MOD), and severe (SEV) groups based on necropsy grading. The superficial cranial region of each PM sample was isolated for quantitative analysis of MuSCs functional markers. One-way ANOVA was used to compare differences among groups, with p < 0.05 indicating significant differences. Results showed that breast muscle weight and morphological indices (length, top/middle/bottom heights) of MOD and SEV groups were higher than those of NOR group (P < 0.05). The SEV group exhibited larger myofiber diameter and cross-sectional area but lower fiber density compared to NOR group (P < 0.05). There was higher mitotic activity of MuSCs, populations of MuSCs and myoblasts in severe WB muscle, characterized by elevated BrdU⁺, Pax7⁺, and MyoD⁺ cells (P < 0.05). The mRNA or protein expressions of Myf5, MyoD, MyoG, Myomaker, Myomerger, MSTN, HGF, IGF-1 and IGF-1R were upregulated in SEV group, along with downregulation of MRF4 protein (P < 0.05). Taken together, the proliferation of MuSCs is promoted in WB muscle, while terminal differentiation and fusion were failure, inhibiting the formation of multinucleated myotubes and mature myofibers. These findings elucidate novel mechanistic insights into the pathogenesis of WB myopathy and potential strategies for stem cell-mediated WB prophylaxis in poultry industry.

Genome-wide association analysis reveals genetic loci and candidate genes for white diarrhea in Jingyuan chickens

In order to investigate the basic genetic structure of dysentery in Jingyuan chickens and to explore the genetic markers associated with dysentery resistance in chickens, the present study was based on the genome-wide association analysis (GWAS) technique to investigate the candidate SNPs and genes associated with dysentery resistance in Jingyuan chickens, and a total of 12 SNPs were associated with dysentery resistance in Jingyuan chickens. In addition, some important candidate genes inciuding frizzled class receptor 4 (FZD4), DDB1 and CUL4 associated factor 13(DCAF13), regulating synaptic membrane exocytosis 2 (RIMS2), transmembrane protein 8C (TMEM8C), and RIC1 homolog (RIC1) were identified by selection signal analysis, gene annotation, and enrichment analysis. These results can be used as potential molecular selection markers for chicken dysentery resistance in Jingyuan chickens in order to improve the breeding of disease resistance in Jingyuan chickens.
