Role of the premotor and the precentral negative motor area in praxis: A direct electrical stimulation study with behavioral analysis

OBJECTIVEAlthough the negative motor area (NMA) is defined as the area where electrical cortical stimulation inhibits voluntary movements, detail functions of NMA on praxis have not been elucidated. We investigated its role in praxis by motion analysis during stimulation at a smaller intensity.METHODSPatients were six intractable partial epilepsy patients undergoing implantation of intracranial electrodes. Motion impairments by stimulation were studied in finger tapping, reach-to-grasp, finger gesture, and pantomime of tool use.RESULTSNMAs were identified on the precentral gyrus (4 patients), ventral premotor area (1), and at their border (1). In patients with precentral NMA, quantitative analysis revealed decreased tapping stroke and grasping aperture, while reaching velocity and pantomime did not change. As for more rostral NMA, quantitative stroke, aperture, and reaching velocity were decreased. One patient showed the arrest of finger gestures and pantomime, and the other had prolongation of reaction time. These two NMAs showed distinct connectivity pattern in connectivity analysis.CONCLUSIONSPrecentral NMA seemed to play a role in elementary finger movement control, whereas more rostral NMA in complex movement. The findings indicate functional differences within NMAs.SIGNIFICANCEThese findings elucidated the contribution of the human premotor area to the highly skilled hand movements.

SP1 regulates porcine primary adipocyte differentiation by modulating BAMBI transcriptional activity

Obesity is a critical metabolic disorder in modern society, necessitating urgent research and intervention. Compared to mice, pigs are increasingly been recognized as valuable model organisms for such studied due to their closer genetic similarity to humans. The BAMBI gene plays a pivotal role in adipocyte differentiation and adipose tissue development, rendering it a potent target for exploring the mechanisms of obesity and related metabolic disorders. This study conducted an in-depth exploration of the intricate regulatory mechanisms governing the BAMBI gene, with a particular emphasis on the transcription factor SP1. Our experiments strongly provided that SP1 modulates BAMBI transcription by directly binding to its promoter. Using primary porcine adipocytes, we further demonstrated that SP1 regulates adipocyte differentiation occurs via the BAMBI gene. These findings enhance understanding of the critical role played by the BAMBI gene in adipocyte differentiation and establish a theoretical foundation for novel therapeutic strategies targeting obesity and type 2 diabetes. Importantly, this study emphasizes the potential of SP-BAMBI pathways in interventions within the context of metabolic diseases, addressing the global challenges posed by obesity and its associated complications.

Comparative analysis of neural crest development in the chick and mouse

A core framework of the gene regulatory network (GRN) governing neural crest (NC) cell development has been generated by integrating separate inputs from diverse model organisms rather than direct comparison. This has limited insights into the diversity of genes in the NC cell GRN and extent of conservation of newly identified transcriptional signatures in cell differentiation and invasion. Here, we address this by leveraging the strengths and accessibility of the avian embryo to precise developmental staging by egg incubation and use an integrated analysis of chick (HH13) and mouse (E9.5) embryo tissue samples collected during NC cell migration into pharyngeal arches 1-2 (PA1 and PA2). We successfully identify a cluster of NC cells containing both mouse and chick cells that share expression of Lmo4, Tfap2B, Sox10, and Twist1, and distinct genes that lack known conserved roles in NC. Importantly, we discovered a cluster of cells exhibiting a conserved transcriptional signature associated with the NC cell migratory wavefront in both mouse and chick, including KAZALD1, BAMBI, DES, and GPC3. We confirm their expression is restricted to leader mouse NCs by multiplexed FISH. Together, these data offer novel insights into the transcriptional programs that underlie NC cell migration and establish the foundation for future comparative functional analyses.
