Relationship between the expression levels of myogenic regulatory factor genes and carcass characteristics in Kivircik and Hungarian Merino lambs

This study aimed to investigate the expression profiles of the myogenic regulatory genes MYOD1, MYOG, MYF5, MYF6, and MSTN in longissimus dorsi muscle, as well as the correlation of the expression levels of these genes with carcass characteristics and growth performance in the Kivircik and Hungarian Merino sheep breeds. The expression levels of the MYF5, MYF6, and MYOG genes were found to be significantly correlated with the rib proportion, the expression level of the MYOG gene was identified as being the main determinant of variations in the rib proportion in the Kivircik lambs. The regression analysis results revealed that the expression levels of the MYF5 and MSTN genes played an essential role in determining the cold carcass dressing percentage in Hungarian Merino lambs. Further, as a result of the regression analysis, the model including the expression level of the MYF6 gene demonstrated that this gene could be responsible for 36.4% of the differences observed in cold carcass weight. In conclusion, the findings of this study suggest that the expression levels of the MYF5, MYF6, and MYOG genes were associated with various carcass traits, particularly in the Kivircik breed, and these genes hold potential as markers for enhancing breed productivity.

Dysfunctional terminal differentiation of satellite cells in the wooden breast muscle of Ross 308 broiler chickens

Muscle satellite cells (MuSCs), defined as skeletal muscle-resident stem cells, serve as the principal cellular mediators of the repair and regeneration processes in impaired skeletal muscle tissue. However, myofibers with degeneration and necrosis in wooden breast (WB) myopathy are still not completely repaired, and this study aims to elucidate the functional changes of MuSCs between normal and WB-affected pectoralis major (PM) muscle of broilers. A total of 100 Ross 308 broilers (42 days old) fed the same diet were randomly selected. Initial identification of normal and WB phenotypes was conducted through behavioral assessments and manual palpation of the PM muscle. Ultimately, PM samples were harvested from 8 chickens per experimental group, classified into normal (NOR), moderate (MOD), and severe (SEV) groups based on necropsy grading. The superficial cranial region of each PM sample was isolated for quantitative analysis of MuSCs functional markers. One-way ANOVA was used to compare differences among groups, with p < 0.05 indicating significant differences. Results showed that breast muscle weight and morphological indices (length, top/middle/bottom heights) of MOD and SEV groups were higher than those of NOR group (P < 0.05). The SEV group exhibited larger myofiber diameter and cross-sectional area but lower fiber density compared to NOR group (P < 0.05). There was higher mitotic activity of MuSCs, populations of MuSCs and myoblasts in severe WB muscle, characterized by elevated BrdU⁺, Pax7⁺, and MyoD⁺ cells (P < 0.05). The mRNA or protein expressions of Myf5, MyoD, MyoG, Myomaker, Myomerger, MSTN, HGF, IGF-1 and IGF-1R were upregulated in SEV group, along with downregulation of MRF4 protein (P < 0.05). Taken together, the proliferation of MuSCs is promoted in WB muscle, while terminal differentiation and fusion were failure, inhibiting the formation of multinucleated myotubes and mature myofibers. These findings elucidate novel mechanistic insights into the pathogenesis of WB myopathy and potential strategies for stem cell-mediated WB prophylaxis in poultry industry.

Mechanochemistry-Induced Universal Hydrogel Surface Modification for Orientation and Enhanced Differentiation of Skeletal Muscle Myoblasts

Micropatterned surface substrates containing topographic cues offer the possibility of programming tissue organization as a cell template by guiding cell alignment, adhesion, and function. In this study, we developed and used a force stamp method to grow aligned micropatterns with tunable chemical properties and elasticity on the surface of hydrogels based on a force-triggered polymerization mechanism of double-network hydrogels to elucidate the underlying mechanisms by which cells sense and respond to their mechanical and chemical microenvironments. In this work, we describe the impact of aligned micropatterns on the combined effects of microstructural chemistry and mechanics on the selective adhesion, directed migration, and differentiation of myoblasts. Our investigations revealed that topographically engineered substrates with hydrophobic and elevated surface roughness significantly enhanced myoblast adhesion kinetics. Concurrently, spatially ordered architectures facilitated cytoskeletal reorganization in myocytes, establishing biomechanically favorable niches for syncytial myotube development through extracellular matrix (ECM) physical guidance. Reverse transcription PCR analysis and immunofluorescence revealed that the expression of differentiation-specific genes, myosin heavy chain, and myogenic regulatory factors Myf5 and MyoD was upregulated in muscle cells on the aligned patterned scaffolds. These results suggest that the aligned micropatterns can promote muscle cell differentiation, making them potential scaffolds for enhancing skeletal differentiation.
