The molecular basis of Staphylococcus aureus resistance to linezolid and the role of silver nanoparticles in attenuating linezolid resistance

BACKGROUNDSeveral linezolid resistance mechanisms have been reported in human pathogen methicillin-resistant Staphylococcus aureus (MRSA). This study investigated the molecular basis of methicillin resistance in S. aureus with increased linezolid minimum inhibitory concentrations (MICs).METHODS AND RESULTSThe antimicrobial efficacy of silver nanoparticles (AgNPs) derived from S. aureus and rosemary leaves (Rosmarinus officinalis) was evaluated against linezolid-resistant S. aureus strains. 81% of the isolates were MRSA, and 20% were linezolid resistant. Twelve isolates were linezolid-resistant S. aureus (LRSA) via the agar dilution. Molecular confirmation of linezolid-resistant S. aureus was performed by detecting the nuc gene and sequencing the 16 S rRNA gene. Molecular analysis of the resistant isolates revealed that mutations in domain V of the 23 S rRNA, rplC and rplV genes were detected in all the isolates, whereas the cfr gene was not assigned. DNA sequencing revealed that the major mutation in our isolates was a G to A mutation at different positions despite the presence of the T2500A mutation.CONCLUSIONSAgNPs synthesized from rosemary leaves and S. aureus had a significant antimicrobial effect on LRSA isolates. In addition, the LRSA isolates became sensitive to linezolid after mixing linezolid with the synthesized AgNPs.

Active capture-directed bimetallic nanosubstrate for enhanced SERS detection of Staphylococcus aureus by combining strand exchange amplification and wavelength-selective machine learning

Staphylococcus aureus (S. aureus) is the leading risk factor for food safety and human health. Herein, a novel wavelength-selective machine learning -driven adaptive strand exchange amplification (SEA)/SERS biosensor was developed for rapid detection of S. aureus. The study operates via three innovative routes: i) the exceptional specificity triggered through SEA of the nuc target gene (nuc T') from S. aureus, ii) anodic aluminum oxide filter membrane-supported gold and silver bimetallic nanoflowers modified with 4-ATP (Au/Ag FL@AAO-4-ATP) were prepared as SERS nanosubstrate for actively capturing the nuc T' through a nucleophilic addition reaction, and for SERS signal enhancement, and finally iii) the integration of a wavelength-selective machine learning tool for further refinement and accuracy of the S. aureus detection process. The proposed wavelength-selective machine learning-driven adaptive Au/Ag FL@AAO-4-ATP nanosubstrate administers prediction performance for the quantitative detection of S. aureus using interval combined population analysis-partial least squares (ICPA-PLS) with root mean square error of prediction and residual predictive deviation values of 0.9626 and 3.56, respectively. The effectiveness of the proposed ICPA-PLS method in real milk samples was validated by a standard fluorescent quantitative PCR method in terms of t-test with no significant differences at P = 0.05. This study offers a new avenue for rapid and straightforward detection of S. aureus, focusing on key genes. The proposed SERS/SEA/machine learning integrated platform can be adapted to other bacterial species via engineering appropriate amplification primers, thus, inspiring potential applications in food safety and biomedical research.

MeRIP-seq data analysis and validation reveal the regulatory role of m6A modified circRNAs in the apoptosis of secondary hair follicle cells in Inner Mongolia cashmere goats

As a widely epigenetic modification, m⁶A (N⁶-methyladenosine, m⁶A) can regulate the degradation, translation, and other biological functions of circRNAs through dynamic reversible processes. It plays an important role in regulating the life activities of biological organisms, particularly in cell differentiation, apoptosis, embryonic development, stress response, and innate immunity. In this study, bioinformatics analysis, qRT-PCR identification, FISH subcellular localization, and ceRNA network construction were performed on m⁶A modified circRNAs regulating the apoptosis of secondary hair follicle cells of Inner Mongolia Albas white cashmere goats based on the skin m⁶A sequencing data of secondary hair follicles in anagen and catagen. The results showed that 8 m⁶A modified circRNAs regulating the cell apoptosis of secondary hair follicles, namely circRNA_2130, circRNA_0013, circRNA_1203, circRNA_1462, circRNA_1242, circRNA_2308, circRNA_2654 and circRNA_1442 were identified, and they are respectively derived from ANGEL2, APP, GKAP1, HNRNPC, PTBP3, NUCB1, SNRK and ZNF609 genes. Among them, circRNA_0013, circRNA_1442 and circRNA_1462 were located in the cytoplasm of the secondary hair follicle papilla, while circRNA_1203, circRNA_1242, circRNA_2130, circRNA_2308 and circRNA_2654 were located in the nucleus. There are complex and diverse regulatory relationships among 8 circRNAs, with each circRNA targeting one or more miRNAs, revealing that each m⁶A circRNA can exert regulatory effects through multiple potential miRNA-mRNA axes, to regulate the apoptosis of secondary hair follicle cells of cashmere goats during the growth cycles. This result provides a direction for further elucidating the regulatory mechanism of m⁶A modified circRNAs in cashmere growth and exploring biomarkers.
