Introduction

Mitochondrial GTPase that mediates the disassembly of ribosomes from messenger RNA at the termination of mitochondrial protein biosynthesis (PubMed:19716793, PubMed:33878294). Acts in collaboration with MRRF (PubMed:19716793, PubMed:33878294). Promotes mitochondrial ribosome recycling by dissolution of intersubunit contacts (PubMed:33878294). GTP hydrolysis follows the ribosome disassembly and probably occurs on the ribosome large subunit (PubMed:19716793). Not involved in the GTP-dependent ribosomal translocation step during translation elongation (PubMed:19716793).

Exploring the Allosteric Response of Fascin to Its Inhibitor

Fascin is a major actin-binding protein (ABP) for stabilizing filopodia to support efficient adhesion and migration of cancer cells. Fascin is also highly expressed in metastatic tumors. Disrupting the actin-binding site (ABS) on fascin constitutes a critical approach to hindering tumor metastasis. The G2 series of small molecules was formulated with the specific purpose of obstructing the binding pocket of fascin. The determination of inhibitor-induced structural dynamics in fascin is crucial for a comprehensive of its biological functions and the strategic development of pharmacological interventions. In this study, we utilized both equilibrium and dynamical-nonequilibrium molecular dynamics (D-NEMD) to elucidate the molecular mechanisms responsible for transmitting structural changes when removing the G2 inhibitor, in both the wild type (WT) and its variants. Our findings indicate that when G2 is removed, structural dynamics in fascin originate from the G2 binding pocket of fascin and propagate signals through the conformational transformation that spans all four β-trefoil domains. Although different mutant variants demonstrated comparable conformational networks, they showed varying response times. However, the signaling pathways in mutants remained consistent in comparison to the WT fascin. This study provides valuable insights into the structural features and communication pathways of fascin and provides avenues for the development of targeted inhibitors with promising prospects in cancer therapy.

Harnessing light‐harvesting chlorophyll a/b‐binding proteins for multiple abiotic stress tolerance in Chlamydomonas reinhardtii: Insights from genomic and physiological analysis

AbstractLight‐harvesting chlorophyll a/b‐binding proteins (LHC) of photosystem II perform key functions in various processes, e.g., photosynthesis, development, and abiotic stress responses. Nonetheless, comprehensive genome‐wide investigation of LHC family genes (CrLHCs) has not been well‐reported in single‐cell alga (Chlamydomonas reinhardtii). Here, we discovered 61 putative CrLHC genes in the C. reinhardtii genome and observed that most genes demonstrate stable exon‐intron and motif configurations. We predicted five phytohormones‐ and six abiotic stress‐interrelated cis‐regulatory elements in promoter regions of CrLHC. Likewise, 19 miRNAs targeting 42 CrLHC genes from 16 unique families were discovered. Besides, we identified 400 transcription factors from 13 families, including ERF, GATA, CPP, bZIP, C3H, MYB, SBP, Dof, bHLH, C2H2, G2‐like, etc. Protein–protein interactions and 3D structures provided insight into CrLHC proteins. Gene ontology and KEGG‐based enrichment advocated their role in light responses, photosynthesis, and energy metabolisms. Expression analysis highlighted the shared and unique roles of many CrLHC genes against different abiotic stresses (UV‐C, green light, heat, nitric oxide, cadmium, nitrogen starvation, and salinity). Under salinity stress, antioxidant enzyme activity, reactive oxygen species markers, photosynthesis‐related traits and pigments were significantly affected. Briefly, this comprehensive genomic and physiological study shed light on the impact of CrLHC genes in abiotic stress tolerance and set the path for future genetic engineering experiments.

Vaccine Molecule Design Based on Phage Display and Computational Modeling against Rhabdovirus

AbstractRhabdoviruses with rich species lead a variety of high lethality and rapid transmission diseases to plants and animals around the globe. Vaccination is one of the most effective approaches to prevent and control virus disease. However, the key antigenic epitopes of glycoprotein being used for vaccine development are unclear. In this study, fish-derived Abs are employed for a Micropterus salmoides rhabdovirus (MSRV) vaccine design by phage display and bioinformatics analysis. We constructed an anti-MSRV phage Ab library to screen Abs for glycoprotein segment 2 (G2) (G129-266). Four M13-phage-displayed Abs (Ab-5, Ab-7, Ab-8 and Ab-30) exhibited strong specificity to target Ag, and Ab-7 had the highest affinity with MSRV. Ab-7 (300 μg/ml) significantly increased grass carp ovary cell viability to 83.40% and significantly decreased the titer of MSRV. Molecular docking results showed that the key region of Ag–Ab interaction was located in 10ESQEFTTLTSH20 of G2. G2Ser11 and G2Gln12 were replaced with alanine, respectively, and molecular docking results showed that the Ag–Ab was nonbinding (ΔG > 0). Then, the peptide vaccine KLH-G210–20 was immunized to M. salmoides via i.p. injection. ELISA result showed that the serum Ab potency level increased significantly (p < 0.01). More importantly, the challenge test demonstrated that the peptide vaccine elicited robust protection against MSRV invasion, and the relative percentage survival reached 62.07%. Overall, this study proposed an approach for screening key epitope by combining phage display technology and bioinformatics tools to provide a reliable theoretical reference for the prevention and control of viral diseases.
