Identification of Dengue virus serotype and genotype: A comprehensive study from AIIMS Patna, Bihar

PURPOSEDengue virus, a major global health concern, exhibits significant genetic diversity, leading to distinct serotypes and genotypes. Dengue is the second most common disease spread by mosquitoes that infect humans, after malaria. In recent decades, there has also been a shift in the tendencies of virus transmission from urban to peri-urban and rural settings. This research study focuses on the identification and characterization of dengue virus serotypes and genotypes.METHODSIn this study, specimens collected in one year were first subjected to serological assay (NS1 and IgM) and subsequently the NS1 positive samples were subjected to identification and characterization of the prevalent dengue virus serotypes and genotypes.RESULTSDuring the year 2021, a total of 933serum samples were tested, out of which 75 were found positive for NS1 antigen and 135 were positive for Dengue IgM antibody by ELISA. The dengue serotype specific Real-Time RT-PCR Assay detected all four serotypes in the NS1-positive samples, indicating their presence in this region. Of these, DEN-1 was detected in 4 (7.8 %), DEN-2 in 26 (50.9 %), DEN-3 in 20 (39 %), DEN-4 in 1 (1.9 %). Out of 51Real-Time RT-PCR positive samples, 33 were found positive by conventional PCR for genotyping by targeting the capsid-preMembrane (C-prM)region. The genotyping result showed that DENV-1 serotype clustering with GIII, DEN-2 serotype clustering withwide range of genotypes such as IVa, IVb, and IVc, DEN-3 clustering with GIII genotype and DEN-4 serotype showed GIII genotype.CONCLUSIONSThis study provides the recent details about the circulating serotypes along with prevalent genotypes in this region.

A Modular Biosensor Design for Quantitative Measurement of Free Nedd8

The objective of our study was to develop a genetically encoded biosensor for quantification of Nedd8, a post-translational modifier that regulates cellular signals through conjugation to other proteins. Perturbations in the balance of free (i.e., unconjugated) and conjugated Nedd8 caused by defects in Nedd8 enzymes or cellular stress are implicated in various diseases. Despite the biological and biomedical importance of Nedd8 dynamics, no method exists for direct quantification of free Nedd8, hindering the study of Nedd8 and activities of its associated enzymes. Genetically encoded biosensors are established as tools to study other dynamic systems, but limitations of current biosensor design methods make them poorly suited for free Nedd8 quantification. We have developed a modular method to design genetically encoded biosensors that employs a target binding domain and two reporter domains positioned on opposite sides of the target binding site. Target quantification is based on competition between target binding and the interaction of the reporter domains. We applied our design strategy to free Nedd8 quantification by developing a selective binder for free Nedd8 and combining it with fluorescent or split nanoluciferase reporters. Our sensors produced quantifiable and specific signals for free Nedd8 and enabled real-time monitoring of deneddylation by DEN1 with a physiological substrate. Our sensor design will be useful for high-throughput screening for deneddylation inhibitors, which have potential in treatment of cancers such as acute lymphoblastic leukemia. The modular design strategy can be extended to develop genetically encoded quantitative biosensors for other proteins of interest.

Etoposide-induced SENP8 confers a feed-back drug resistance on acute lymphoblastic leukemia cells

Chemotherapy is the most common treatment for acute lymphoblastic leukemia (ALL). However, many ALL patients eventually develop relapse and treating relapsed ALL has always been challenging. Therefore, exploring the resistance mechanism of chemotherapeutic drugs and proposing feasible intervention strategies are of great significance for ALL treatment. Here, we show that SENP8, whose coding protein is an important deNEDDylase targeting the substrate for deNEDDylation, is highly expressed in relapsed ALL specimens. Interestingly, overexpressing SENP8 specifically reduces the chemosensitivity of ALL cells to etoposide (VP-16) and significantly alleviates the proapoptotic effect of VP-16 on ALL cells. By contrast, NEDDylation inhibition reduces the chemosensitivity of ALL cells to VP-16. Furthermore, VP-16 induces SENP8 accumulation and the instability of MDM2 as well as the stabilization of p53 in ALL cells, and SENP8 knockdown can sensitize ALL cells to VP-16. Our study reveals a novel function of SENP8 in ALL and that VP-16-induced SENP8 confers a feed-back drug resistance on ALL cells, suggesting a possibility of overcoming the chemotherapeutic resistance to VP-16 via targeting SENP8.
