Introduction

Extracellular matrix protein that seems to be a ligand for ITGA8:ITGB1, ITGAV:ITGB1 and ITGA4:ITGB1 (By similarity) (PubMed:17909022). Involved in neurite outgrowth and cell migration in hippocampal explants (By similarity). During endochondral bone formation, inhibits proliferation and differentiation of proteoblasts mediated by canonical WNT signaling (By similarity). In tumors, stimulates angiogenesis by elongation, migration and sprouting of endothelial cells (PubMed:19884327). Expressed in most mammary tumors, may facilitate tumorigenesis by supporting the migratory behavior of breast cancer cells (PubMed:17909022).

Genomic Signatures of Recurrence After Resection of Early-Stage Node-Negative Colon Cancer

BACKGROUNDWhereas clinicopathologic risk factors for colon cancer (CC) recurrence are well-established, the role of genomic predictors remains understudied. This study aimed to identify genomic factors associated with recurrence after resection of early-stage, node-negative CC.METHODSA retrospective cohort study was performed using clinicopathologic data, somatic mutations, and mRNA expression profiles from the cBioPortal database. The study enrolled patients with T1-3, N0, or M0 CC who underwent surgical resection and primary tumor sequencing. Those with fewer than 6 months of follow-up evaluation were excluded. Gene expression profiles were classified into consensus molecular subtypes (CMSs). Somatic mutations and CMS groups were analyzed for associations with recurrence.RESULTSOf the 305 patients analyzed, 46 (15%) experienced recurrence. The median age at diagnosis was 70 years (interquartile range [IQR], 62-76 years), the gender distribution was balanced. The median follow-up period was 38 months, and the median time to recurrence was 13 months. The following 12 mutated genes were significantly associated with recurrence: KRAS (odds ratio [OR], 2.19), PIK3CA (OR, 2.13), DNAH11 (OR, 3.28), NALCN (OR, 4.69), COL6A3 (OR, 3.12), GRIN2A (OR, 4.92), COL6A1 (OR, 4.25), TNN (OR, 3.25), NEXMIF (OR, 6.97), PKHD1L1 (OR, 3.24), CDH4 (OR, 3.29), and BCL9 (OR, 4.03). Additionally, tumors classified as CMS4/mesenchymal subtype had a greater risk of recurrence (OR, 4.67) than the CMS2/canonical subtype. Patients with CMS4 or at least four mutations associated with recurrence (n = 77) had a 5-year disease-free survival rate of 57.7%.CONCLUSIONThis study identified novel genomic signatures that may improve risk stratification in early-stage node-negative CC, potentially guiding the selection of high-risk patients for adjuvant therapy.

Analysis of hydrogen sulfide-related signatures of breast cancer identified 2 distinct subtypes: Implications for individualized therapeutics

Hydrogen sulfide (H2S) is a vital gasotransmitter involved in breast cancer (BC) pathogenesis. This study aims to employ hydrogen sulfide-related genes (HSRGs) for molecular classification of BC and, accordingly, to establish a robust prognostic risk signature. Transcriptomic, clinical, and mutational data of BC patients were collected from the cancer genome atlas and gene expression omnibus databases. Prognostic relevance was evaluated using Cox regression analysis, while consensus clustering analysis was employed for molecular subtyping. Gene expression profiles, prognosis, immune infiltration patterns, drug sensitivity, and response to immunotherapy were compared between subtypes. Multiple-gene prognostic features were developed and assessed along with a nomogram. The gene expression was validated in clinical samples using quantitative polymerase chain reaction. Among 282 HSRGs, 46 exhibited significant correlations with BC prognosis. Consensus clustering identified 2 distinct molecular subtypes (C1 and C2). C1 displayed significantly improved prognosis compared to C2, accompanied by increased infiltration of B cells, T cells, monocytes, and mast cells but decreased macrophage infiltration. Moreover, C1 demonstrated higher drug sensitivity and immunotherapeutic response relative to C2. Enrichment analysis revealed suppressed immune-related processes and pathways in C2 while cell cycle regulation and chromosomal processes were significantly activated. Additionally, a risk feature comprising 6 differentially expressed genes between subtypes was constructed; this feature performed well in prognostic prediction. Integration of this feature with other clinical parameters (radiotherapy/chemotherapy status, clinical stage, N stage) into a nomogram further enhanced prognostic accuracy. Clinical samples further validated the high expression of ATP13A5, LRTM2, MAFA, and SPDYC and the low expression of CYP4F12 and TNN in BC. Our findings highlight the clinical relevance of HSRGs in BC, providing a basis for precise molecular classification and prognosis evaluation. The developed risk feature and nomogram offer practical tools for guiding personalized treatment strategies in clinical practice.

Clinical value of multi-slice spiral CT in evaluating preoperative TNN staging and postoperative recurrence and metastasis of colon carcinoma

OBJECTIVETo evaluate the clinical value of multi-slice spiral CT in preoperative TNN staging and postoperative recurrence and metastasis of colon carcinoma, and to provide evidence for the reliability of CT in the diagnosis of colon carcinoma METHODS: 89 patients with colon carcinoma diagnosed pathologically in our hospital from July 2020 to April 2023 were selected retrospectively. The preoperative TNN staging and postoperative recurrence and metastasis were monitored by 64 row 128 layer spiral CT. The diagnostic coincidence rate, TNM staging coincidence rate and postoperative recurrent TNM staging accuracy were evaluated according to the pathological diagnosis RESULTS: The diagnostic coincidence rate of multi-slice spiral CT was 97.8 % (87/89), and the detection rate of lymph nodes was 86.1 % (31/36). The coincidence rate of T staging was 93.3 % (83/89), N staging was 91.0 % (81/89), M staging was 100 % (Kappa=0.897,0.879, 1.000). The diagnosis of recurrent TNM stage was consistent (Kappa=0.893, 0.801, 1.000) CONCLUSION: Multi-slice spiral CT is of high diagnostic coincidence rate, high accuracy of TNM staging and rapid noninvasive examination. It can obtain reliable results in preoperative staging and postoperative recurrence and metastasis diagnosis, which is worth popularizing in clinic.
