Identification of Specific Biomarkers for Anaplastic Thyroid Carcinoma Through Spatial Transcriptomic and Immunohistochemical Profiling

Anaplastic thyroid carcinoma (ATC) is an aggressive malignancy with a poor prognosis. Despite its rarity, identifying predictive molecular markers that distinguish ATC from follicular cell-derived non-anaplastic thyroid carcinomas is critical for improving diagnosis and treatment strategies. This study aimed to identify and validate key mRNA and protein markers associated with ATC progression and dedifferentiation. We performed spatial transcriptomic analysis on an index case of ATC coexisting with papillary thyroid carcinoma (PTC) and identified eight differentially expressed mRNA markers. These findings were validated in a large cohort using immunohistochemistry on tissue microarrays across various thyroid tumor types, including follicular adenoma, PTC, poorly differentiated thyroid carcinoma, medullary thyroid carcinoma, and ATC. Additionally, the impact of BRAF p.V600E mutation status on these markers was evaluated. COL7A1, LAMC2, SPHK1, and SRPX2 mRNA and protein levels were significantly overexpressed in ATCs. Conversely, CD24, EPHX1, GPX3, and RBM47 mRNA and protein levels were markedly downregulated in ATCs. Functional enrichment analysis, based on mRNA expression data, identified the role of these proteins in tumor invasion, epithelial-mesenchymal transition, extracellular matrix remodeling, and immune evasion. The expression levels of these markers were independent of BRAF p.V600E mutation status, highlighting their potential as diagnostic markers. In summary, this study identified eight molecular markers that can distinguish ATC from other thyroid tumors. The validation of these markers at both the mRNA and protein levels underscores their clinical relevance in ATC diagnosis and tumor characterization. These findings provide a foundation for future biomarker-driven diagnostic and therapeutic strategies for ATC.

Spatially resolved multi-omics reveals the renal cortex-metabolic reprogramming of Shenhua Tablet for intervention on IgA nephropathy

BACKGROUNDShenhua tablet (SHT) is a clinically used Chinese patent medicine, which has garnered attention for its effectiveness in treating IgA nephropathy (IgAN). Nevertheless, early researches lacked anatomical and metabolic data, hindering a comprehensive understanding of the therapeutic mechanisms of SHT in spatial contexts.PURPOSEWe aimed to explore the molecular mechanism of SHT intervention in IgAN by utilizing spatial multi-omics strategies.STUDY DESIGNWe injected Thy-1 into tail vein to induce IgAN rat model and administer SHT. Classical pharmacological parameters were used to evaluate the efficacy of SHT. The distribution of active components of SHT and their regulation for metabolites and upstream genes in the cortex were examined to determine the intervention mechanism of SHT.METHODSAfter establishing the animal models and administering SHT treatment, Kidney injury were assessed using biochemical indexes and histopathology. Classical and spatial metabolomics were employed to detect metabolites in serum and kidney. Spatial transcriptomics was used to detect mRNA levels in renal sections adjacent to the spatial metabolomics. In addition, mass-spectrometry-imaging and cell experiments were used to explore and verify the active components of SHT.RESULTSSHT reduced inflammation and mesangial cell proliferation, and reversed kidney damage. Mechanically, in renal tubules, SHT regulated glutathione metabolism by reversing the expression of Gclc and Gpx3. It was further found that Pck1 and G6pc1 were increased to inhibit glycolysis. In glomeruli, SHT downregulated Oat and Odc1 and reduced spermidine and l-proline levels to inhibit mesangial cell proliferation. Finally, formononetin, calycosin and curzerenone were identified as the main active components of SHT and showed their distribution in the cortex.CONCLUSIONSSHT ameliorated renal injury by regulating glutathione metabolism, glycolysis, and l-proline metabolism, providing a more comprehensive insight into the molecular mechanisms of SHT intervention in IgAN in a spatial context, and offering new perspectives for the treatment of IgAN.

CiLi (Rosa roxburghii Tratt.) polyphenols improve colitis via gut microbiota-lipid mediator-immunity axis

Dysbiosis of gut microbiome is one of the most important factors leading to inflammatory bowel disease (IBD). Intake of phytochemicals from fruits and vegetables is an effective way to improve IBD, but how these bioactivators regulate gut microbiota to exert healthy effects remains unclear. Here, we found that pretreatment with CiLi juice, particularly its polyphenol component, alleviated dextran sulfate sodium (DSS)-induced colitis while preserving intestinal barrier integrity. CiLi polyphenols (CL_PP) reduced inflammation and oxidative stress in colon tissue and enriched fecal short-chain fatty acids. Importantly, CL_PP significantly regulated the gut microbiome diversity, increasing beneficial bacteria (e.g., Clostridia_UCG-014, f_Muribaculaceae and Ileibacterium_valens) while decreasing harmful bacteria (Escherichia-Shigella and Romboutsia). Multiomics analysis revealed that CL_PP upregulated bioactive lipid metabolites, particularly those derived from polyunsaturated fatty acids (e.g., resolvin D2, prostaglandin A1, and glycerophosphocholine) related gene expressions (Pltp, Alox15 and Pld4). Additionally, CL-PP downregulated the oxidative stress markers (oxidized glutathione and glutathione peroxidase 3), and immune cell markers (CD8 and CD68). Fecal microbiota transplantation confirmed that the fecal microbiota from CL_PP-treated mice exhibited anti-colitis effects. These effects were diminished in antibiotic-treated mice, underscoring the importance of the gut microbiota in mediating the CL_PP's anti-inflammatory benefits. This study suggests that CL_PP is a potential modulator of gut microbiome dysbiosis for the prevention and treatment of colitis.
