Introduction

Catalyzes the transfer of a sialic acid from a CMP-linked sialic acid donor onto a terminal alpha-2,3-, alpha-2,6-, or alpha-2,8-linked sialic acid of an N-linked glycan protein acceptor through alpha-2,8-linkages (PubMed:10766765, PubMed:11279095, PubMed:28810663, PubMed:9774483). Therefore, participates in polysialic acid synthesis on various sialylated N-acetyllactosaminyl oligosaccharides, including NCAM1 N-glycans, FETUB N-glycans and AHSG (PubMed:10766765, PubMed:11279095, PubMed:28810663, PubMed:9774483). It is noteworthy that alpha-2,3-linked sialic acid is apparently a better acceptor than alpha-2,6-linked sialic acid (PubMed:9774483).

Influence of operation sequences on phosphorus enrichment by polyphosphate accumulating organisms biofilm: Performance, phosphorus transfer and phosphate metabolism in biofilm

This study investigated the impact of different operational sequences on phosphorus removal and enrichment in biofilm phosphorus enrichment system. The research com-pared two distinct operational modes, analyzing phosphorus uptake and release characteristics in cells and extracellular polymeric substances (EPS) over a single cycle, while also examining microbial community composition and associated functional genes. After long-term acclimation, the Ae/An system achieved higher phosphorus concentration (120 mg/L) than the An/Ae system (65 mg/L). However, the An/Ae system showed stronger phosphorus uptake and release capabilities due to higher phosphorus load during the aerobic phase. In both systems, Mg-P and Ca-P dominated in cells and EPS. Compared to the Ae/An system, the An/Ae system stored phosphorus mainly in EPS, with higher orthophosphate content. However, EPS-associated phosphorus is more easily released, explaining the An/Ae system's higher aerobic phosphorus load but lower overall storage capacity. Microbial analysis revealed higher abundance of phosphorus accumulating organisms (PAOs) in the An/Ae system (25.99 % vs. 19.69 %), while glycogen accumulating organisms (GAOs) showed the opposite trend. Candidatus Competibacter was abundant in both systems and correlated with phosphorus metabolism genes. The An/Ae system expressed the pst system more, whereas the Ae/An favored the pit system, suggesting that transfer system variations affect enrichment solution concentration. Lower expression of polyphosphate kinase (ppk1) in the An/Ae system may explain its unsatisfied phosphorus enrichment performance. Mantel analysis confirmed connections among environmental factors, kinetic parameters, phosphorus metabolism genes, and phosphorus morphology in EPS, demonstrating their combined influence on enrichment solution concentration.

Positive Preoperative Stress Test is Associated with Increased Risk of Postoperative Myocardial Infarction in Patients Undergoing Aortoiliac Reconstruction for Chronic Limb-Threatening Ischemia

BACKGROUNDAortofemoral or aortoiliac bypasses (AFBs/AIBs) are considered the gold standard treatment for patients with aortoiliac occlusive disease. Myocardial infarction (MI) is a postoperative risk in AFB/AIB surgeries and preoperative stress test (PST) is frequently utilized to inform operative planning and preoperative risk discussions. The aim of this study is to assess the utility of PST in determining outcomes following AFB/AIB in patients with chronic limb-threatening ischemia (CLTI).METHODSThis is a retrospective study from Vascular Quality Initiative database. Patients undergoing AFB/AIB for chronic limb-threatening ischemia during 2009-2023 were stratified by PST result. Primary outcomes included postoperative MI and 30-day mortality. Secondary outcomes included in-hospital death, in-hospital death/MI, postoperative respiratory complications, postoperative congestive heart failure, intensive care unit stay > 3 days, and 1-year mortality. Logistic and Cox regressions were used for multivariate analyses.RESULTSThe study included 2 cohorts of patients: negative (N = 1,809, 81.2%) and positive PST (N = 420, 18.8%). The patients with positive PST had increased rate of postoperative MI compared to the patients with negative PST (6.7% vs. 2.9%, P < 0.001). The rate of 30-day mortality was greater for patients with positive PST but the difference was not significant (3.8% vs. 2.9%, P = 0.315). After adjusting for potential confounders, positive PST was associated with increased risk of postoperative MI (adjusted odds ratio = 1.89 [95% confidence interval: 1.04-3.43], P = 0.038). However, positive PST was not associated with 30-day mortality. Pulmonary complications, congestive heart failure, and intensive care unit stay >3 days were also not associated with PST result. The overall survival rate at 1-year was 93.3% and 92.0% for patients with negative and positive PSTs (P = 0.343), respectively. After adjusting for potential confounders, postoperative MI was associated with increased hazards of death at 1 year (adjusted hazard ratio = 3.62 [95% confidence interval: 2.21-5.92], P < 0.001).CONCLUSIONThis study emphasizes the important of appropriate preoperative planning. A positive stress test before open aortic reconstruction for aortoiliac occlusive disease is associated with increased risk of postoperative MI. Although this was neither associated with increased risk of 30-day nor 1-year mortality, the negative impact of MI on patient functionality, quality of life, and 1-year death cannot be ignored.

Altered volume of thalamic nuclei and genetic expression in first-episode psychotic patients, and their association with childhood adversity

Childhood maltreatment is a significant risk factor for schizophrenia, and there are correlations between these adversities and thalamic gray matter density. The thalamus, a subcortical structure with various nuclei with specific connections, relays sensory information and participates in higher cognitive processes. Thalamic alterations are evident in psychotic disorders, and early-life adversities may affect its development, potentially contributing to psychosis. However, no evidence exists of volumetric alterations in thalamic nuclei in first-episode psychosis (FEP) patients related to early traumatic events. This study recruited 70 FEP patients and 68 age-matched healthy controls, who underwent 3 T structural MRI and clinical scales, including the Childhood Trauma Questionnaire (CTQ). The thalamus was analyzed for shape and segmented into nuclei to assess volume. Additionally, peripheral blood was analyzed for the expression of VCAN, CSGALNACT1, ST8SIA4, NRGN, SP4, and TOX genes, which are related to neuronal plasticity in the thalamus and psychosis. Results showed volumetric reductions in the whole thalamus and specific nuclei (lateral posterior, lateral geniculate, medial geniculate, ventrolateral, centromedian, anteroventral, mediodorsal, and pulvinar). The thalamus did not show shape alterations. A significant association was observed between physical neglect during childhood and the volume of the left thalamus and its anteroventral nucleus. Reduced expression of ST8SIA4 and SP4 genes was detected in FEP patients compared to healthy controls, with correlations between thalamic nuclei volumes and gene expression differing between groups. In conclusion, this study links thalamic nuclei volume with childhood adversities in FEP and highlights changes in ST8SIA4 and SP4 expression, correlating with thalamic nuclei volumes.
