Elevated pCO2 may increase the edible safety risk of clams exposed to toxic Alexandrium spp.

Toxic harmful algal blooms (HABs) have received increasing attention owing to their threat to the health of aquatic life and seafood consumers. This study evaluated the impacts of elevated atmospheric partial pressure of CO₂ (pCO₂) on the production of paralytic shellfish toxins (PSTs) in different Alexandrium spp. strains, together with its further effects on the bioaccumulation/elimination dynamics of PSTs in bivalves contaminated with PSTs from toxic dinoflagellates. Our results showed that elevated pCO₂ stimulated the growth of the two Alexandrium spp. (A. catenella and A. pacificum) isolated from the northern and southern coastal areas of China, respectively, and affected PST production including content and toxicity of the two strains differently. Further PSTs bioaccumulation/elimination in PSTs-contaminated Manila clam, Ruditapes philippinarum under high pCO₂ also occurred. It is worth noting the biotransformation of neosaxitoxin (NEO) with high toxicity through trophic transfer with effect of elevated pCO₂. When in microalgae cultured under the control (410 ppm) and elevated pCO₂ conditions (495 and 850 ppm), the proportion of NEO in the PST content produced by A. catenella was reduced from 11.1 to 6.4 and 2.6 %, while the proportion of NEO in A. pacificum was increased from 3.1 to 3.6 and 4.7 %, respectively. NEO accounted for >50 % of total PST contents in clams, which were biotransformed via transfer from dinoflagellates and higher pCO₂ enhanced this biotransformation leading to increased NEO accumulation. The negatively affected elimination of PSTs, especially NEO, in clams fed with A. catenella or A. pacificum, indicates that the detoxification of PSTs-contaminated clams may be more difficult under elevated pCO₂. This study provides reference for developing models to assess the safety of bivalves under the co-stress of environmental change and toxic HABs, suggesting that ocean acidification may lead to the higher safety risk of Manila clams exposed to toxic HAB dinoflagellates.

Novel insights on ecological responses of short- and long-chain perfluorocarboxylic acids in constructed wetlands coupled with modified basalt fiber bio-nest

The first investigation based on constructed wetlands coupled with modified basalt fiber bio-nest (MBF-CWs) was performed under exposure of short- and long-chain perfluorocarboxylic acids (PFCAs). In general, both perfluorooctanoic acid (PFOA) and perfluorobutanoic acid (PFBA) caused significant decline of chemical oxygen demand removal by 10.83 % and 4.73 %. However, only PFOA led to marked inhibition on total phosphorus removal by 12.51 % in whole duration. Suppression of removal performance resulted from side impacts on microbes by PFOA. For instance, activities of key enzymes like dehydrogenase (DHA), urease (URE), and phosphatase (PST) decreased by 52.77 %, 40.70 %, and 56.94 % in maximum under PFOA stress, while URE could alleviate over time. By contrast, distinct inhibition was only found on PST in later phases with PFBA exposure. PFCAs had adverse influence on alpha diversity of MBF-CWs, particularly long-chain PFOA. Both PFCAs caused enrichment of Proteobacteria, owing to increase of Gammaproteobacteria and Plasticicumulans by 22.04-35.79 % and 22.91-219.77 %. Nevertheless, some dominant phyla (like Bacteroidota and Acidobacteriota) and genera (like SC-I-84, Thauera, Subgroup_10, and Ellin6067) were only suppressed by PFOA, causing more hazards to microbial decontamination than PFBA did. As for plants, chlorophyll contents tend to decrease with PFOA treatment. Whereas, higher antioxidase activities and more lipid peroxidation products were uncovered in PFOA group, demonstrating more reactive oxygen species brought by long-chain PFCAs. This work offered new findings about ecological effects of MBF-CWs under PFCAs exposure, evaluating stability and sustainability of MBF-CW systems to treat sewage containing complex PFCAs.

Isolation and Identification of Highly Sb-Resistant Rhodotorula glutinis Strain J5 and its Mechanism of Resistance to Sb(III)

Sb-resistant strains can detoxify antimony through metabolic mechanisms such as oxidation and affect the migration, transformation, and ultimate fate of antimony in the environment. In this study, a strain of Sb-resistant fungi, Rhodotorula glutinis sp. Strain J5, was isolated from Xikuangshan mine and its growth characteristics, gene expression differences, and functional annotation under Sb(III) stress were further investigated to reveal the mechanism of resistance to Sb(III). We identified strain J5 as belonging to the Rhodotorula glutinis species optimally growing at pH 5.0 and at 28 °C of temperature. According to gene annotation and differential expression, the resistance mechanism of Strain J5 includes: reducing the endocytosis of antimony by aquaporin AQP8 and transmembrane transporter pst, enhancing the efflux of Sb(III) by the gene expression of acr2, acr3 and ABC, improving the oxidation of Sb(III) by iron-sulfur protein and Superoxide dismutase (SOD), glutathione (GSH) and cysteine (Cys) chelation, methylation of methyltransferase and N-methyltransferase, accelerating cell damage repair and EPS synthesis and other biochemical reaction mechanisms. FT-IR analysis shows that the -OH, -COOH, -NH, -PO, C-O, and other active groups of Strain J5 can be complexed with Sb(III), resulting in chemical adsorption. Strain J5 displays significant resistance to Sb(III) with the MIC of 1300 mg/L, playing a crucial role in the global biochemical transformation of antimony and its potential application in soil microbial remediation.
