Introduction

Serine/threonine-protein kinase involved in the control of the eukaryotic cell cycle, whose activity is controlled by an associated cyclin. Acts as a cell-cycle regulator of Wnt signaling pathway during G2/M phase by mediating the phosphorylation of LRP6 at 'Ser-1490', leading to the activation of the Wnt signaling pathway. Acts as a regulator of cell cycle progression and cell proliferation via its interaction with CCDN3. Phosphorylates RB1 in vitro, however the relevance of such result remains to be confirmed in vivo. May also play a role in meiosis, neuron differentiation and may indirectly act as a negative regulator of insulin-responsive glucose transport.

Cross-talk between immune cells and tumor cells in non-Hodgkin lymphomas arising in common variable immunodeficiency

INTRODUCTIONCVID is the commonest and most symptomatic primary immune deficiency of adulthood. NHLs are the most prevalent malignancies in CVID. The cross-talk between tumor cells and immune cells may be an important risk factor in lymphomagenesis.AREAS COVEREDThe present review highlights immune cell, genetic and histopathological alterations in the CVID-associated NHLs.EXPERT OPINIONCVID patients exhibit some notable immune defects that may predispose to lymphomas. T/NK cell defects including reduced T cells, naïve CD4+T cells, T regs, and Th17 cells, increased CD8+T cells with reduced T cell proliferative and cytokine responses and reduced iNKT and NK cell count and cytotoxicity. B cell defects include increased transitional and CD21^low B cells, clonal IgH gene rearrangements, and increased BCMA levels. Increase in IL-9, sCD30 levels, and upregulation of BAFF-BAFFR signaling are associated with lymphomas in CVID. Increased expression of PFTK1, duplication of ORC4L, germline defects in TACI, NFKB1, and PIK3CD, and somatic mutations in NOTCH2 and MYD88 are reported in CVID-associated lymphomas. Upregulation of PD-L1-PD-1 pathway may also promote lymphomagenesis in CVID. These abnormalities need to be explored as prognostic or predictive markers of CVID-associated NHLs by large multicentric studies.

CDK14 is regulated by IGF2BP2 and involved in osteogenic differentiation via Wnt/β-catenin signaling pathway in vitro

AIMSCyclin-dependent kinase (CDK) family proteins involve in various cellular processes via regulating the cell cycle; however, their expression during osteogenic differentiation and postmenopausal osteoporosis remains poorly understood.MAIN METHODSUsing bioinformatics, we screened for CDK14 bound to Insulin-like growth factor 2 mRNA-binding protein 2 (IGF2BP2) and explored its expression in vitro with time-gradient model and in a mouse model of postmenopausal osteoporosis, building on prior research. Subsequently, we investigated its effect on osteoblast proliferation, cell cycle dynamics, and osteogenic differentiation by administering CDK14 siRNA and the covalent inhibitor FMF-04-159-2. Furthermore, we examined the interaction between IGF2BP2 and CDK14. Finally, we validated the regulatory role of CDK14 on the Wnt/β-catenin pathway.KEY FINDINGSOur findings demonstrate a time-dependent CDK14 expression patterns during osteogenic differentiation of MC3T3-E1 cell line, with an initial increase followed by gradual decline over time. Notably, CDK14 expression exhibited significant reduction in bone tissue of postmenopausal osteoporosis mouse model. CDK14 inhibition altered osteoblast cell cycle dynamics, significantly reduced cellular proliferation capacity, and impaired osteogenic differentiation ability. IGF2BP2 interacted with CDK14 mRNA, and stabilizing mRNA's structure and inhibiting its degradation. Additionally, CDK14 facilitated Low-density lipoprotein receptor-related protein 6 (LRP6) and Glycogen synthase kinase 3β (GSK3β) phosphorylation, thus regulating β-catenin levels.SIGNIFICANCEThese findings provide further insight into the molecular mechanisms governing osteoblast proliferation, differentiation and osteoporosis.

Relationships between Circulating Monocyte and Brain Transcriptional Profiles in Nonhuman Primates Consuming Mediterranean versus Western Like Diets

AbstractBackgroundMediterranean diets may reduce Alzheimer’s disease (AD) risk and preserve cognitive function relative to Western diets by protecting against inflammation. In a long term controlled randomized trial of Mediterranean vs. Western diet consumption in cynomolgus macaques (Macaca fascicularis), difficult to conduct in humans, we found significant anti‐inflammatory effects of Mediterranean diet on circulating monocyte and brain temporal cortex transcriptional profiles. Here we examine relationships between monocyte and temporal cortex gene expression profiles.Methods38 middle‐aged (mean = 12 years of age) female macaques were fed Western or Mediterranean diets for 2.5 years in a randomized nonhuman primate trial. RNAseq was used to assess transcriptional profiles in CD14+ monocytes after 15 months of diet consumption while temporal cortex was obtained after 2.5 years.ResultsCortex expression of cyclin dependent kinase 14 (CDK14), a proinflammatory regulator, was lower in the Mediterranean group. Six other cortex transcripts [i.e., “lunatic fringe” (LFNG), mannose receptor C type 2 (MRC2), solute carrier family 3 member 2 (SLCA32), butyrophilin subfamily 2 member A1 (BTN2A1), katanin regulatory subunit B1 (KATNB1), and transmembrane protein 268 (TMEM268)] higher in the Mediterranean group were associated with anti‐inflammatory/neuroprotective pathways. Monocyte patterns of gene expression predicted expression of these targets in the brain, with consistent anti‐inflammatory effects of the Mediterranean Diet relative to the Western Diet (Figure 1). Monocyte transcripts associated with inflammation, including proinflammatory components of the constitutive transcriptional response to adversity (CTRA, Cole et al. 2015), NFKB1, NFKB2, and IL6, were positively associated with temporal cortex transcript level of the pro‐inflammatory gene CDK14 (all p’s≤0.0?). In general, these proinflammatory monocyte transcript levels were negatively correlated with temporal cortex anti‐inflammatory transcripts LFNG, SLC3A2, and MRC2 (8 of 12 correlations p<0.05; 11 of 12 correlations p<0.10), consistent with an overall anti‐inflammatory effect of the Mediterranean diet.ConclusionCirculating monocyte gene expression profiles correlated with temporal cortex transcript levels consistent with anti‐inflammatory effects of the Mediterranean Diet relative to the Western Diet, suggesting that peripheral inflammation may promote neuroinflammation. Interventions including diet to target inflammation may have a role in prevention or treatment of systemic as well as neuro‐inflammation and neuropathology.
