Comparing UV and Diesel Cutaneous Damage and Evaluating the Protective Role of a Topical Antioxidant Mixture Containing Vitamin C, E and Ferulic Acid

ABSTRACTCutaneous tissue is one of the main targets of outdoor stressors, and nowadays, the effect of pollution on skin conditions and premature skin ageing has been well correlated, although the exact effect that different pollutants have on the skin has not been well defined, especially when compared to other stressors. Among the air pollutants, UV radiation and particulate matter (PM) have been found among the most aggressive in terms of skin damage, inducing oxinflammatory responses, promoting degradation of extracellular matrix (ECM) components, and compromising the cutaneous defensive barrier. Topical application of technologies able to prevent oxidative damage is still one of the best approaches to protect our skin, and considering the well‐known antioxidant network, application of an antioxidant mixture is more recommended than a single compound. In the present study, human skin explants were exposed every day for 4 days to diesel particles (DEE) or to UV after the daily pre‐treatment with a topical application of a commercially available antioxidant mixture (AOX Mix), containing 15% ascorbic acid, 0.5% ferulic acid and 1% tocopherol. Oxidative stress markers such as 4‐hydroxynonenal, skin barrier proteins such as involucrin, filaggrin, claudin‐1 and desmocollin‐1, resilience markers such as elastin and tropoelastin, and the levels of Type I and Type III collagens were assessed. Topical application was able to prevent most of the damage induced by the outdoor stressors, confirming that daily protection is needed to prevent cutaneous premature ageing.

HPV-Associated Gene Signatures in Bladder Cancer: A Comprehensive Prognostic Model and its Implications in Immunotherapy

Background: Evidence increasingly indicates that HPV infection plays a pivotal role in the initiation and progression of bladder cancer (BC). Yet, determining the predictive value of HPV-associated genes in BC remains challenging. Methods: We identified differentially expressed HPV-associated genes of BC patients from the TCGA and GEO databases. We screened prognostic genes using COX and LASSO regression, subsequently establishing a risk prediction model. The model's precision and clinical relevance were gauged using Kaplan-Meier survival analyses and ROC curves. Functional enrichment, immune cell infiltration, and drug sensitivity analyses were performed across both high-risk and low-risk sets. PCR assays were utilized to measure the expression levels of genes. Results: We identified 13 HPV-associated genes for our risk model. Among these, FLRT2, HOXC5, LDLR, SCD, GRM7, DSC1, EMP1, and HMGA1 were identified as risk contributors, while LPA, SERPINA6, ZNF124, ETV7, and SCO2 were deemed protective. Cox regression analysis verified that our model provides an independent prediction of overall survival (OS) in bladder cancer (BC) patients. Gene Ontology (GO) analysis revealed predominant gene enrichment in wound healing, extracellular matrix composition, and collagen-rich extracellular matrices. KEGG pathway analysis highlighted primary enrichment areas, including focal adhesion, the PI3K-Akt signalling pathway, and ECM-receptor interaction. Risk scores were correlated with tumor microenvironment (TME) scores, immune cell infiltration, and sensitivities to both chemotherapy and immunotherapy. Conclusion: We have formulated a risk-assessment model pinpointing 13 central HPV-associated genes in BC. These genes present potential as prognostic indicators and therapeutic targets, emphasizing the intertwined relationship between HPV-induced BC progression and the immune landscape.

Immunohistopathological analyses of a case of pemphigus vegetans with antibodies against desmoglein 1 and desmocollins 1–3

AbstractPemphigus vegetans is a rare type of pemphigus characterized by vegetative lesions primarily localized to the intertriginous area. Despite its unique clinical presentation, the underlying pathomechanism remains unclear owing to the rarity of the disease. We report a case of pemphigus vegetans with antibodies against desmoglein 1 and desmocollins 1–3. Furthermore, immunohistochemical analyses were performed to address the pathogenesis of this disease. A 73‐year‐old man presented with multiple vegetative plaques with erythema on the trunk, groins, and extremities. Mucosal lesions were not observed. Laboratory examinations revealed mild leukocytosis with eosinophilia. A histopathological examination of the skin lesion showed epidermal hyperplasia and intraepidermal abscesses with marked infiltration of neutrophils and eosinophils, and infiltration of lymphocytes and eosinophils into the upper derms. Bacterial culture of the skin tissue was positive for Staphylococcus aureus. Direct immunofluorescence showed deposits of IgG and C3 on keratinocyte surfaces in the epidermis. Autoantibodies against desmoglein 1 and autoantibodies against desmocollin 1, desmocollin 2, and desmocollin 3 were detected by enzyme‐linked immunosorbent assays. The diagnosis of pemphigus vegetans was made. Initiation of prednisolone (1.0 mg/kg/day) gradually improved his skin symptoms. We performed immunohistochemical analyses of the lesional skin, which revealed infiltration of CD3‐positive, CD4‐positive, and CD68‐positive cells in the upper dermis, but CD20‐ or CD56‐positive cells were negative. In addition, the present case showed more prominent infiltration of IL‐17A‐ and IL‐22‐positive cells in the upper dermis than in pemphigus foliaceus, a type of pemphigus with autoantibodies against desmoglein 1. Furthermore, these cells co‐expressed CD3 and CD68. We hypothesized that IL‐22 and IL‐17A produced by T cells and macrophages and their dysregulation might be involved in the pathogenesis of pemphigus vegetans. Additionally, skin colonization and/or infection with Staphylococcus aureus could potentially contribute to the pathogenesis of the disease.
